Currently, there is a trend towards the use of minimally invasive techniques for the treatment of posterior heel pain. In our hospital, we prefer to use the tendon splitting approach through a four-shaped cutaneous incision. It provides a better visualization of retrocalcaneal space. This type of surgical approach allows an easier and complete surgery with a very low complication rate
Post-transcriptional regulation of the yeast high affinity iron transport system.
Saccharomyces cerevisiae transcriptionally regulates the expression of the plasma membrane high affinity iron transport system in response to iron need. This transport system is comprised of the products of the FET3 and FTR1 genes. We show that Fet3p and Ftr1p are post-translationally regulated by iron. Incubation of cells in high iron leads to the internalization and degradation of both Fet3p and Ftr1p. Yeast strains defective in endocytosis (Deltaend4) show a reduced iron-induced loss of Fet3p-Ftr1p. In cells with a deletion in the vacuolar protease PEP4, high iron medium leads to the accumulation of Fet3p and Ftr1p in the vacuole. Iron-induced degradation of Fet3p-Ftr1p is significantly reduced in strains containing a deletion of a gene, VTA1, which is involved in multivesicular body (MVB) sorting in yeast. Sorting through the MVB can involve ubiquitination. We demonstrate that Ftr1p is ubiquitinated, whereas Fet3p is not ubiquitinated. Iron-induced internalization and degradation of Fet3p-Ftr1p occurs in a mutant strain of the E3 ubiquitin ligase RSP5 (rsp5-1), suggesting that Rsp5p is not required. Internalization of Fet3p-Ftr1p is specific for iron and requires both an active Fet3p and Ftr1p, indicating that it is the transport of iron through the iron permease Ftr1p that is responsible for the internalization and degradation of the Fet3p-Ftr1p complex
Serafino Zappacosta: An Enlightened Mentor and Educator
With this article, the authors aim to honor the memory of Serafino Zappacosta, who had been their mentor during the early years of their career in science. The authors discuss how the combination of Serafino Zappacosta's extraordinary commitment to teaching and passion for science created a fostering educational environment that led to the creation of the “Ruggero Ceppellini Advanced School of Immunology.” The review also illustrates how the research on the MHC and the inspirational scientific context in the Zappacosta's laboratory influenced the authors' early scientific interests, and subsequent professional work as immunologists
Conversion of the Mycotoxin Patulin to the Less Toxic Desoxypatulinic Acid by the Biocontrol Yeast Rhodosporidium kratochvilovae Strain LS11
Se describe en este artículo el descubrimiento de la degradación de la micotoxina patulina por una levaduraThe infection of stored apples by the fungus Penicillium expansum causes the contamination of fruits and fruit-derived
products with the mycotoxin patulin, which is a major issue in food safety. Fungal attack can be prevented by beneficial
microorganisms, so-called biocontrol agents. Previous time-course thin layer chromatography analyses showed that the aerobic
incubation of patulin with the biocontrol yeast Rhodosporidium kratochvilovae strain LS11 leads to the disappearance of the
mycotoxin spot and the parallel emergence of two new spots, one of which disappears over time. In this work, we analyzed the
biodegradation of patulin effected by LS11 through HPLC. The more stable of the two compounds was purified and characterized by
nuclear magnetic resonance as desoxypatulinic acid, whose formation was also quantitated in patulin degradation experiments. After
R. kratochvilovae LS11 had been incubated in the presence of 13C-labeled patulin, label was traced to desoxypatulinic acid, thus
proving that this compound derives from the metabolization of patulin by the yeast. Desoxypatulinic acid was much less toxic than
patulin to human lymphocytes and, in contrast to patulin, did not react in vitro with the thiol-bearing tripeptide glutathione. The
lower toxicity of desoxypatulinic acid is proposed to be a consequence of the hydrolysis of the lactone ring and the loss of functional
groups that react with thiol groups. The formation of desoxypatulinic acid from patulin represents a novel biodegradation pathway
that is also a detoxification process
REGULATION OF PROSTAGLANDIN GENERATION IN CARRAGEENAN-INDUCED PLEURISY BY INDUCIBLE NITRIC OXIDE SYNTHASE IN KNOCKOUT MICE
In the present study, by comparing the responses in wild-type mice (iNOSWT) and mice lacking (iNOSKO) the inducible (or type 2) nitric oxide synthase (iNOS), we investigated the correlation between endogenous nitric oxide (NO) and prostaglandin (PG) generation in carrageenan-induced pleurisy. The inflammatory response in iNOSKO mice was significantly reduced in respect to iNOSWT animals, as demonstrated by the exudate volume (-63%) and numbers of infiltrating cells (-62%). The levels of NOx in the pleural exudate from carrageenan-treated mice were significantly (p < 0.01) decreased in iNOSKO mice (16 ± 7.6 nmoles/mice) compared to iNOSWT animals (133 ± 9 nmoles/mice). Similarly, the amounts of PGE2 in the pleural exudates of carrageenan-treated animals were significantly (p < 0.01) lower in iNOSKO compared to iNOSWT mice (120 ± 20 pg/mice vs. 308 ± 51 pg/mice). Also the amounts of 6-keto-PGF1α produced by lungs from carrageenan-treated iNOSKO mice (1.01 ± 0.10 ng/tissue mg) were significantly (p < 0.01) reduced compared to iNOSWT carrageenan-treated mice (2.1 ± 0.09 ng/tissue mg). In conclusion our results confirm, by the use of iNOSKO mice that in carrageenan-induced pleurisy NO positively modulates PG biosynthesis
Two years of flight of the Pamela experiment: results and perspectives
PAMELA is a satellite borne experiment designed to study with great accuracy
cosmic rays of galactic, solar, and trapped nature in a wide energy range
(protons: 80 MeV-700 GeV, electrons 50 MeV-400 GeV). Main objective is the
study of the antimatter component: antiprotons (80 MeV-190 GeV), positrons (50
MeV-270 GeV) and search for antinuclei with a precision of the order of

Ab-initio study of model guanine assemblies: The role of pi-pi coupling and band transport
Several assemblies of guanine molecules are investigated by means of
first-principle calculations. Such structures include stacked and
hydrogen-bonded dimers, as well as vertical columns and planar ribbons,
respectively, obtained by periodically replicating the dimers. Our results are
in good agreement with experimental data for isolated molecules, isolated
dimers, and periodic ribbons. For stacked dimers and columns, the stability is
affected by the relative charge distribution of the pi orbitals in adjacent
guanine molecules. pi-pi coupling in some stacked columns induces dispersive
energy bands, while no dispersion is identified in the planar ribbons along the
connections of hydrogen bonds. The implications for different materials
comprised of guanine aggregates are discussed. The bandstructure of dispersive
configurations may justify a contribution of band transport (Bloch type) in the
conduction mechanism of deoxyguanosine fibres, while in DNA-like configurations
band transport should be negligible.Comment: 21 pages, 6 figures, 3 tables, to be published in Phys. Rev.
Bladder irrigation with tap water to reduce antibiotic treatment for catheter-associated urinary tract infections: an evaluation of clinical practice
IntroductionCatheter-associated urinary tract infection (CAUTI) is a common complication among patients with urinary catheters and is often treated with antibiotics. With increasing rates of antibiotic resistance, it is necessary to explore alternative treatment options for CAUTIs. The aims of this study were 1) to assess the efficacy and treatment satisfaction of bladder irrigation (BI) with tap water to prevent and treat CAUTIs, 2) and to evaluate the current use of BI for CAUTIs among Dutch clinicians.MethodsThe first part of this study consisted of a cross-sectional study among patients with intermittent or indwelling catheters who performed BI with tap water between March 2020 and May 2021. Efficacy, treatment satisfaction, and Patient Global Impression of Improvement (PGI-I) were assessed using questionnaires. Outcomes were compared between neurogenic lower urinary tract dysfunction (NLUTD) and non-NLUTD patients. Factors associated with overall treatment satisfaction were determined using logistic regression analysis. Second, a nationwide survey of Dutch clinicians was conducted to evaluate the current use of BI for CAUTIs.ResultsA total of 99 patients who were performing BI for at least three months were included. The median age was 61.9 years, 41.4% had NLUTD, and 72.2% performed BI >1 year. The majority of both NLUTD (65.9%) and non-NLUTD patients (68.4%) were (very) satisfied with BI. Women had higher odds of reporting higher satisfaction and each additional CAUTI decreased the odds. Most NLUTD (85.4%) and non-NLUTD (65.5%) patients reported an improvement on the PGI-I with a difference in favour of NLUTD patients (p=0.002). In addition, 40.4% of the patients had no CAUTI, and 59.6% reported 1.39 (SD 2.06) CAUTIs. Only half of these self-reported CAUTIs were treated with antibiotics. In addition, 33 (58.9%) clinicians used BI for CAUTIs, of which ten used tap water as irrigation agent.DiscussionThis study provides first evidence supporting the efficacy of BI with tap water in the treatment of CAUTIs and reducing the use of antibiotics. Patients are overall satisfied and experience improvement in their condition with BI. In addition, the majority of the surveyed Dutch clinicians use BI for CAUTIs. However, irrigation with tap water is still not widely used
Launch of the Space experiment PAMELA
PAMELA is a satellite borne experiment designed to study with great accuracy
cosmic rays of galactic, solar, and trapped nature in a wide energy range
protons: 80 MeV-700 GeV, electrons 50 MeV-400 GeV). Main objective is the study
of the antimatter component: antiprotons (80 MeV-190 GeV), positrons (50

The experiment, housed on board the Russian Resurs-DK1 satellite, was launched
on June, 15, 2006 in a 350*600 km orbit with an inclination of 70 degrees. The
detector is composed of a series of scintillator counters arranged at the
extremities of a permanent magnet spectrometer to provide charge,
Time-of-Flight and rigidity information. Lepton/hadron identification is
performed by a Silicon-Tungsten calorimeter and a Neutron detector placed at
the bottom of the device. An Anticounter system is used offline to reject false
triggers coming from the satellite. In self-trigger mode the Calorimeter, the
neutron detector and a shower tail catcher are capable of an independent
measure of the lepton component up to 2 TeV. In this work we describe the
experiment, its scientific objectives and the performance in the first months
after launch.Comment: Accepted for publication on Advances in Space Researc
