    Deep learning for diffusion in porous media

    We adopt convolutional neural networks (CNN) to predict the basic properties of the porous media. Two different media types are considered: one mimics the sandstone, and the other mimics the systems derived from the extracellular space of biological tissues. The Lattice Boltzmann Method is used to obtain the labeled data necessary for performing supervised learning. We distinguish two tasks. In the first, networks based on the analysis of the system's geometry predict porosity and effective diffusion coefficient. In the second, networks reconstruct the system's geometry and concentration map. In the first task, we propose two types of CNN models: the C-Net and the encoder part of the U-Net. Both networks are modified by adding a self-normalization module. The models predict with reasonable accuracy but only within the data type, they are trained on. For instance, the model trained on sandstone-like samples overshoots or undershoots for biological-like samples. In the second task, we propose the usage of the U-Net architecture. It accurately reconstructs the concentration fields. Moreover, the network trained on one data type works well for the other. For instance, the model trained on sandstone-like samples works perfectly on biological-like samples.Comment: 17 pages, 19 figure

    Spreading of a density front in the K\"untz-Lavall\'ee model of porous media

    We analyze spreading of a density front in the K\"untz-Lavall\'ee model of porous media. In contrast to previous studies, where unusual properties of the front were attributed to anomalous diffusion, we find that the front evolution is controlled by normal diffusion and hydrodynamic flow, the latter being responsible for apparent enhancement of the front propagation speed. Our finding suggests that results of several recent experiments on porous media, where anomalous diffusion was reported based on the density front propagation analysis, should be reconsidered to verify the role of a fluid flow

    Defective Awakening Response to Nocturnal Hypoglycemia in Patients with Type 1 Diabetes Mellitus

    BACKGROUND: Nocturnal hypoglycemia frequently occurs in patients with type 1 diabetes mellitus (T1DM). It can be fatal and is believed to promote the development of the hypoglycemia-unawareness syndrome. Whether hypoglycemia normally provokes awakening from sleep in individuals who do not have diabetes, and whether this awakening response is impaired in T1DM patients, is unknown. METHODS AND FINDINGS: We tested two groups of 16 T1DM patients and 16 healthy control participants, respectively, with comparable distributions of gender, age, and body mass index. In one night, a linear fall in plasma glucose to nadir levels of 2.2 mmol/l was induced by infusing insulin over a 1-h period starting as soon as polysomnographic recordings indicated that stage 2 sleep had been reached. In another night (control), euglycemia was maintained. Only one of the 16 T1DM patients, as compared to ten healthy control participants, awakened upon hypoglycemia (p = 0.001). In the control nights, none of the study participants in either of the two groups awakened during the corresponding time. Awakening during hypoglycemia was associated with increased hormonal counterregulation. In all the study participants (from both groups) who woke up, and in five of the study participants who did not awaken (three T1DM patients and two healthy control participants), plasma epinephrine concentration increased with hypoglycemia by at least 100% (p < 0.001). A temporal pattern was revealed such that increases in epinephrine in all participants who awakened started always before polysomnographic signs of wakefulness (mean ± standard error of the mean: 7.5 ± 1.6 min). CONCLUSIONS: A fall in plasma glucose to 2.2 mmol/l provokes an awakening response in most healthy control participants, but this response is impaired in T1DM patients. The counterregulatory increase in plasma epinephrine that we observed to precede awakening suggests that awakening forms part of a central nervous system response launched in parallel with hormonal counterregulation. Failure to awaken increases the risk for T1DM patients to suffer prolonged and potentially fatal hypoglycemia

    Glucokinase (GCK) Mutations and Their Characterization in MODY2 Children of Southern Italy

    Type 2 Maturity Onset Diabetes of the Young (MODY2) is a monogenic autosomal disease characterized by a primary defect in insulin secretion and hyperglycemia. It results from GCK gene mutations that impair enzyme activity. Between 2006 and 2010, we investigated GCK mutations in 66 diabetic children from southern Italy with suspected MODY2. Denaturing High Performance Liquid Chromatography (DHPLC) and sequence analysis revealed 19 GCK mutations in 28 children, six of which were novel: p.Glu40Asp, p.Val154Leu, p.Arg447Glyfs, p.Lys458_Cys461del, p.Glu395_Arg397del and c.580-2A>T. We evaluated the effect of these 19 mutations using bioinformatic tools such as Polymorphism Phenotyping (Polyphen), Sorting Intolerant From Tolerant (SIFT) and in silico modelling. We also conducted a functional study to evaluate the pathogenic significance of seven mutations that are among the most severe mutations found in our population, and have never been characterized: p.Glu70Asp, p.His137Asp, p.Phe150Tyr, p.Val154Leu, p.Gly162Asp, p.Arg303Trp and p.Arg392Ser. These seven mutations, by altering one or more kinetic parameters, reduced enzyme catalytic activity by >40%. All mutations except p.Glu70Asp displayed thermal-instability, indeed >50% of enzyme activity was lost at 50°C/30 min. Thus, these seven mutations play a pathogenic role in MODY2 insurgence. In conclusion, this report revealed six novel GCK mutations and sheds some light on the structure-function relationship of human GCK mutations and MODY2

    Nawożenie jest podstawowym czynnikiem plonotwórczym, a zużycie nawozów jest jednym ze wskaźników oceny intensywności gospodarowania. W opracowaniu określono wpływu zmian relacji rynkowych pomiędzy cenami produktów rolniczych i nawozów mineralnych na poziom zrównoważenia gospodarki nawozowej w polskim rolnictwie. Rynek nawozów mineralnych w analizowanym okresie charakteryzował się bardzo dużą dynamiką zmian. W latach 2003-2009 odnotowano wyraźną tendencje wzrostową zużycia NPK/ha UR ogółem i oddzielnie każdego ze składników pokarmowych. Bardzo niekorzystnie należy ocenić natomiast silną tendencje zmniejszenia zużycia nawozów wapniowych.The fertilization is one of mainly yielding factor, and consumption of fertilizers is one of the indicators to assess the intensity of management. The paper showed impact of changes in the market of mineral fertilizers to the sustainable level of fertilizer management in Poland. Market of mineral fertilizers in the analysed period was characterized by a very large dynamic change. In the years 2003-2009 saw the rising trend in consumption of NPK/ha UR overall and for each nutrient individually. Very adversely should be assessed and the strong tendency to reduce consumption of calcium fertilizers
