Standardization of an experimental model of intradural injection after spinal cord injury in rats
OBJECTIVES: The intrathecal route has not yet been thoroughly standardized and evaluated in an experimental model of spinal cord injury (SCI) in Wistar rats. The objective of this study was to standardize and evaluate the effect of intradural injection in this animal model. METHOD: The animals were divided into 6 groups: 1) laminectomy and intradural catheter; 2) laminectomy, intradural catheter and infusion; 3) only SCI; 4) SCI and intradural catheter; 5) SCI, intradural catheter and infusion; and 6) control (laminectomy only). Motor evaluations were performed using the Basso, Beattie and Bresnahan (BBB) scale and the horizontal ladder test; motor evoked potentials were measured for functional evaluation, and histological evaluation was performed as well. All experimental data underwent statistical analysis. RESULTS: Regarding motor evoked potentials, the groups with experimental SCI had worse results than those without, but neither dural puncture nor the injection of intrathecal solution aggravated the effects of isolated SCI. Regarding histology, adverse tissue effects were observed in animals with SCI. On average, the BBB scores had the same statistical behaviour as the horizontal ladder results, and at every evaluated timepoint, the groups without SCI presented scored significantly better than those with SCI (po0.05). The difference in performance on motor tests between rats with and without experimental SCI persisted from the first to the last test. CONCLUSIONS: The present work standardizes the model of intradural injection in experimental SCI in rats. Intrathecal puncture and injection did not independently cause significant functional or histological changes
Análise tomográfica para colocação de parafusos em C2 nos pacientes com artrite reumatoide Tomographic analysis for C2 screw placement in rheumatoid arthritis patients
OBJETIVO: A análise morfológica da estrutura óssea de C2, em pacientes com artrite reumatoide, com o objetivo de aumentar a segurança de procedimentos de estabilização desta vértebra. MÉTODOS: Analisamos retrospectivamente 20 tomografias de coluna cervical realizadas em pacientes com artrite reumatoide, foram medidos os seguintes parâmetros: o ângulo espinolaminar, espessura e comprimento da lâmina de C2. RESULTADOS: Os valores médios encontrados são: espessura das lâminas direita 5,92mm e esquerda 5,87mm; comprimento das lâminas direita 27,75mm e esquerda 27,94mm e ângulo espinolaminar 44,7(0). CONCLUSÃO: Os valores obtidos são compatÃveis com os de estudos realizados em indivÃduos normais publicados por outros autores, não havendo aparente necessidade de alteração na técnica para colocação dos parafusos. NÃvel de Evidência IV, Série de casos.<br>OBJECTIVE: A morphological analysis of the bone structure of C2 in patients with rheumatoid arthritis in order to enhance the security of the stabilization procedures for this vertebra. METHODS: We retrospectively analyzed 20 CT scans of the cervical spine performed in patients with rheumatoid arthritis; the following parameters were measured: espinolaminar angle, thickness and length of C2 lamina. RESULTS: The mean values are: 5.92mm and 5.87mm for thickness of right and left laminae retrospectively, 27.75mm for right lamina lenght and 27.94mm for left lamina lenght, and 44.7º for espinolaminar angle. CONCLUSION: The values obtained are consistent with studies in normal subjects published by other groups, with no apparent need for change in the screw placement technique. Determinants of birthweight; gender based analysis
The objective of this cross sectional study is to look at determinants of birth weight and their association with the gender of the baby in 2,795 full term children living in the occupied Palestinian territory, derived from a stratified random sample of 2,994 households in the West Bank and 2,234 households in the Gaza Strip. The response rate was 85%. Multivariable analysis using analysis of variance for mixed models showed that sex and birth order, maternal age and education and to a lesser extent region were determinants of birth weight for all children. The effect of maternal education on birth weight differed for female and male infants, tending to be relatively unchanged for male infants and with mean birth weights increasing with maternal education in female infants. The effect of birth order differed by maternal age, with mean birth weight increasing with maternal age for first and second births; but being unaffected by maternal age for infants of birth order greater than two. We conclude that birth weight is influenced by common biological determinants across cultures, but is also influenced by social, ethnic, and environmental factors that are culture specific, of which some might be gender related
Distribution of a Ty3/gypsy-like retroelement on the A and B-chromosomes of Cestrum strigilatum Ruiz & Pav. and Cestrum intermedium Sendtn. (Solanaceae)
Retroelements are a diversified fraction of eukaryotic genomes, with the Ty1/copia and Ty3/gypsy groups being very common in a large number of plant genomes. We isolated an internal segment of the Ty3/gypsy retroelement of Cestrum strigilatum (Solanaceae) using PCR amplification with degenerate primers for a conserved region of reverse transcriptase. The isolated segment (pCs12) was sequenced and showed similarity with Ty3/gypsy retroelements of monocotyledons and dicotyledons. This segment was used as probe in chromosomes of C. strigilatum and Cestrum intermedium. Diffuse hybridization signals were observed along the chromosomes and more accentuated terminal signals in some chromosome pairs, always associated with nucleolus organizer regions (NORs). The physical relationship between the hybridization sites of pCs12 and pTa71 ribosomal probes was assessed after sequential fluorescence in situ hybridization (FISH). Hybridization signals were also detected in the B chromosomes of these species, indicating an entail among the chromosomes of A complement and B-chromosomes
Padronização da lesão de medula espinal em ratos Wistar Standardization of spinal cord injury in Wistar rats
OBJETIVO: Padronizar um modelo experimental de lesão de medula espinal em ratos Wistar, utilizaram-se um equipamento computadorizado para impacto por queda de peso e os parâmetros determinados pelo Multicenter Animal Spinal Cord Injury Study - MASCIS. MÉTODOS: Avaliaram-se 30 ratos, com idade variando entre 20 e 25 semanas de vida. O peso variou de 200 a 300 g, para as fêmeas, e de 232 a 430 g para os machos. Realizaram-se impactos com pesos de 10 g de 12,5; 25 e 50 mm de altura, controlando-se a velocidade de impacto e o coeficiente de compressão. O impacto ocorreu sobre a superfÃcie da medula espinal na altura da décima vértebra torácica, após laminectomia. Monitoraram-se os sinais vitais e realizaram-se gasometrias previamente e posteriormente à lesão da medula. O volume de lesão foi avaliado pela análise quantitativa dos Ãons de sódio e potássio. RESULTADOS: Verificaram-se correlações estatisticamente significantes entre o volume de lesão e os parâmetros mecânicos. O volume de lesão provocado por queda de 50 mm de altura foi superior aos de 12,5 e 25 mm, que não diferiram entre si. CONCLUSÃO: O modelo demonstrou-se eficaz e capaz de gerar lesões medulares padronizadas em ratos Wistar.<br>OBJECTIVE: To standardize an experimental model of spinal cord injury in Wistar rats, computerized weight fall impact equipment were used and the parameters were used determined by the multicenter animal spinal cord injury study - MASCIS. METHODS: Thirty rats were used, with age varying between 20 and 25 weeks, and weight ranging from 200 to 300g for females, and from 232 to 430g for males. The impacts were done with weights of 10g starting from 12.5, 25 and 50 mm of height, and the impact speed and compression coefficient were obtained. The impact occurred on the surface of the spinal cord at the level of the tenth thoracic vertebra after laminectomy. Vital signs were monitored and gas analysis was made before and after the spinal cord injury. The lesion volume was evaluated by the quantitative analysis of sodium and potassium ions. RESULTS: Statistically significant correlations were verified among the lesion volume and the mechanical parameters. The lesion volume caused by the fall from 50mm height was superior to that of the 12.5 and 25mm, which didn't differ from each other. CONCLUSION: The model demonstrated itself to be effective and capable of generating standard spinal cord injuries on Wistar rats
