The basic representations about electrochemical processes and systems are outlined. The reactions and principles of chemical sources of electric energy and electrolysis systems functioning are analyzed. A general idea is given about the chemical properties of metals, corrosion resistance in environments of various aggressiveness, and the protection principles are given.     In 178 patients with different clinical forms of pulmonary tuberculosis carried out pharmacogenetic testing to determine gene polymorphism glutathione S-transferase system (GSTM1, GSTT1). Estimated the severity and duration of clinical manifestations of the syndrome of intoxication, depending on the gene polymorphism of glutathione S-transferase system (GSTM1, GSTT1).     Stat1 nuclear translocation by nucleolin upon monocyte differentiation

    BACKGROUND: Members of the signal transducer and activator of transcription (Stat) family of transcription factors traverse the nuclear membrane through a specialized structure, called the nuclear pore complex (NPC), which represents a selective filter for the import of proteins. Karyophilic molecules can bind directly to a subset of proteins of the NPC, collectively called nucleoporins. Alternatively, the transport is mediated via a carrier molecule belonging to the importin/karyopherin superfamily, which transmits the import into the nucleus through the NPC. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we provide evidence for an alternative Stat1 nuclear import mechanism, which is mediated by the shuttle protein nucleolin. We observed Stat1-nucleolin association, nuclear translocation and specific binding to the regulatory DNA element GAS. Using expression of nucleolin transgenes, we found that the nuclear localization signal (NLS) of nucleolin is responsible for Stat1 nuclear translocation. We show that this mechanism is utilized upon differentiation of myeloid cells and is specific for the differentiation step from monocytes to macrophages. CONCLUSIONS/SIGNIFICANCE: Our data add the nucleolin-Stat1 complex as a novel functional partner for the cell differentiation program, which is uniquely poised to regulate the transcription machinery via Stat1 and nuclear metabolism via nucleolin

    A molecular assembly system for presentation of antigens on the surface of HBc virus-like particles

    AbstractHepatitis B virus-like particles, icosahedral structures formed by multiple core protein dimers, are promising immune-enhancing vaccine carriers for foreign antigens. Insertions into the surface-exposed immunodominant loop are especially immunogenic. However, the need to conserve the particulate structure to ensure high immunogenicity imposes restraints on the nature of the heterologous sequence that can be inserted. We propose a new approach to constructing HBc particles linked to the target epitopes that relies on non-covalent interactions between the epitope and pre-assembled unmodified HBc particles. Interaction was enabled by fusion of the epitope to the GSLLGRMKGA peptide, binding to the spike tips. This peptide may be used as a “binding tag” allowing in vitro construction of HBc particles carrying the target peptide. Such virus-like particles carrying multiple copies of the extracellular domain of the M2 protein of different influenza strains appeared to be highly immunogenic and protected immunised mice against a lethal influenza challenge

    ИСПОЛЬЗОВАНИЕ ФЛУОРИМЕТРИИ В БЛИЖНЕЙ ИК-ОБЛАСТИ С ФОТОГРАФИЧЕСКОЙ РЕГИСТРАЦИЕЙ СИГНАЛА В МЕТОДЕ «ОТПЕЧАТКОВ ПАЛЬЦЕВ» С ДОБАВКОЙ ФЛУОРОФОРА К ОБЪЕКТУ: ДИСКРИМИНАЦИЯ ЯБЛОЧНЫХ СОКОВ

    The application of dyes, that fluoresce in the near infrared (NIR, 700-800 nm) region, for the recognition of samples using a fingerprinting method with the addition of fluorophores to the samples (“fluorescent eye”) is proposed. The technique has been successfully applied to the classification of samples of various nature. In the current work, this strategy has been tested on the example of discrimination of 17 samples of apple juice from different manufacturers, purchased at different times. An indolenine series heptamethine carbocyanine dye in the presence of surfactants was used as the added fluorophore, red LEDs were used as an excitation source, and the signal was recorded using a digital camera with an additional IR filter installed; a spectrofluorimeter with a 96-well plate accessory was used to record the spectra. Photographic images were processed using Unscrambler X and Excel software. The results were presented using the following coordinates: intensity of NIR fluorescence - intensity of visible light reflection (using the photographic images). It was found that such presentation allowed the samples to be divided into groups associated with the manufacturer. We have also obtained intrinsic fluorescence spectra, including those with the addition of NIR dye, and these results were processed by the principal component analysis. It was possible to distinguish 5–6 groups of samples by their intrinsic emission, not counting the blank, while the spectra with the addition of the dye allowed to isolate the largest number of groups of samples (9). At the same time, the classification using spectra did not allow juices to be grouped by the producer. Also, obtaining photographs using a visualizer was easier and faster than recording the fluorescence spectra. В данной работе метод опробован на примере дискриминации 17 образцов яблочного сока разных производителей, выпущенных в разное время. В качестве добавляемого флуорофора использовали гептаметиновый карбоцианиновый краситель индоленинового ряда в присутствии ПАВ, в качестве источника излучения – красные светодиоды, а сигнал регистрировали с помощью цифрового фотоаппарата с дополнительным ИК-светофильтром; для записи спектров применяли спектрофлуориметр с приставкой для 96-луночного флуориметрического планшета. Фотографические изображения обрабатывали с помощью стандартного программного обеспечения Unscrambler X и Excel. Результаты представили в координатах: интенсивность БИК-флуоресценции – интенсивность отражения видимого света (с использованием соответствующих фотографий). Обнаружили, что такое представление позволяет разделить образцы на группы, связанные с производителем. Получали также спектры собственной флуоресценции, в том числе с добавкой БИК-красителя, обрабатывая эти результаты методом главных компонент. По собственной эмиссии можно выделить 5–6 групп образцов, не считая контрольного, тогда как по спектрам с добавкой красителя удается добиться выделения наибольшего числа групп образцов (девять). При этом классификация с использованием спектров не позволяет группировать соки по производителям. Кроме того, получение фотографий с помощью визуализатора проще и экспресснее, чем регистрация спектров флуоресценции.     ABOUT NON-TRADITIONAL FEED ADDITIVES INFLUENCE ON SAFETY OF THE YAKUT BREED HORSES LIVE WEIGHT

    Horse breeding is one of the rapidly developing branches of animal husbandry in Yakutia. Horses of the Yakut breed are only type of farm animals that are kept year-round in harsh conditions of the sharply continental climate of Yakutia. In winter, animals experience a significant lack of nutrients and minerals. This is due to fact that basic diet consists of natural feed winter pasture and hay. Therefore, the goal was to study the effect of complex feed additives from local natural raw materials on the change in live weight and biochemical composition of the blood of animals. This study used standard animal research methods. The use of non-traditional feed additives in feeding of horses contributed to an increase in the supply of nutrients and minerals to the rations. Improving feeding conditions affected the live weight of horses (live weight loss was 5.75 % и 5.45 %). The change caused by inclusion of non-traditional feed additives in the diets of horses had a positive effect on the morphological and biochemical composition of the blood animals. In horses from the experimental groups, an increase was found in the blood in total protein by 0.50 % and 0.30 %, albumin by 0.36 and 0.20 %, globulin by 0.13 and 0.10 %, hemoglobin by 2.91 and 1.33 %, calcium by 3.56 and 1.29%, phosphorus by 5.71 and 1.90 %. Thus, use of experimental non-traditional feed additives in feeding horses in winter contributes to the best preservation of live weight. This is due to the improvement of mineral and vitamin nutrition in a difficult period of deficiency of macro- and microelements, vitamins in the diets of horses. This improves metabolism, which is reflected in the morphological and biochemical composition of blood of horses

    The Challenge of Machine Learning in Space Weather Nowcasting and Forecasting

    The numerous recent breakthroughs in machine learning (ML) make imperative to carefully ponder how the scientific community can benefit from a technology that, although not necessarily new, is today living its golden age. This Grand Challenge review paper is focused on the present and future role of machine learning in space weather. The purpose is twofold. On one hand, we will discuss previous works that use ML for space weather forecasting, focusing in particular on the few areas that have seen most activity: the forecasting of geomagnetic indices, of relativistic electrons at geosynchronous orbits, of solar flares occurrence, of coronal mass ejection propagation time, and of solar wind speed. On the other hand, this paper serves as a gentle introduction to the field of machine learning tailored to the space weather community and as a pointer to a number of open challenges that we believe the community should undertake in the next decade. The recurring themes throughout the review are the need to shift our forecasting paradigm to a probabilistic approach focused on the reliable assessment of uncertainties, and the combination of physics-based and machine learning approaches, known as gray-box.Comment: under revie
