    Hdo And SO2 Thermal Mapping On Venus: Evidence For Strong SO2 Variability

    We have been using the TEXES high-resolution imaging spectrometer at the NASA Infrared Telescope Facility to map sulfur dioxide and deuterated water over the disk of Venus. Observations took place on January 10-12, 2012. The diameter of Venus was 13 arcsec, with an illumination factor of 80%. Data were recorded in the 1344-1370 cm(-1) range (around 7.35 mu m) with a spectral resolving power of 80 000 and a spatial resolution of about 1.5 arcsec. In this spectral range, the emission of Venus comes from above the cloud top (z = 60-80 km). Four HDO lines and tens of SO2 lines have been identified in our spectra. Mixing ratios have been estimated from HDO/CO2 and SO2/CO2 line depth ratios, using weak neighboring transitions of comparable depths. The HDO maps, recorded on Jan. 10 and Jan. 12, are globally uniform with no significant variation between the two dates. A slight enhancement of the HDO mixing ratio toward the limb might be interpreted as a possible increase of the D/H ratio with height above the cloud level. The mean H2O mixing ratio is found to be 1.5 +/-0.75 ppm, assuming a D/H ratio of 0.0312 (i.e. 200 times the terrestrial value) over the cloud deck. The SO2 maps, recorded each night from Jan. 10 to Jan. 12, show strong variations over the disk of Venus, by a factor as high as 5 to 10. In addition, the position of the maximum SO2 mixing ratio strongly varies on a timescale of 24 h. The maximum SO2 mixing ratio ranges between 75 +/-25 ppb and 125 +/-50 ppb between Jan. 10 and Jan. 12.     Creatine metabolism: detection of creatine and guanidinoacetate in saliva of healthy subjects.

    Creatine (Cr) plays an important role in storage and transmission of phosphate-bound energy. Cerebral creatine deficiency syndromes comprise three inherited defects in Cr biosynthesis and transport. The aim of this study was to investigate whether Cr and Guanidinoacetate (GAA) can be detected in saliva of healthy subjects and to establish the relationship between salivary and plasma levels of these molecules. An adapted gas chromatography (GC) method is described for the quantification of Cr and GAA biomarkers in saliva. Reference values were established for GAA and Cr in saliva. These values were age dependent (p= 0.001). No difference between genders was observed. We detected a difference between GAA and Cr concentrations in saliva and in plasma. The GC method for simultaneous determination of GAA and Cr in human saliva is fast, reliable, sensitive, non-invasive and precise to use as a biochemical approach in early detection of cerebral creatine deficiency syndromes.publishedVersio

    Seasonal Variations in Titan's Stratosphere Observed with Cassini/CIRS: Temperature, Trace Molecular Gas and Aerosol Mixing Ratio Profiles

    Titan's northern spring equinox occurred in August 2009. General Circulation Models (e.g. Lebonnois et al., 2012) predict strong modifications of the global circulation in this period, with formation of two circulation cells instead of the pole-to-pole cell that occurred during northern winter. This winter single cell, which had its descending branch at the north pole, was at the origin of the enrichment of molecular abundances and high stratopause temperatures observed by Cassini/CIRS at high northern latitudes (e.g. Achterberg et al., 2011, Coustenis et al., 2010, Teanby et al., 2008, Vinatier et al., 2010). The predicted dynamical seasonal variations after the equinox have strong impact on the spatial distributions of trace gas, temperature and aerosol abundances. We will present here an analysis of CIRS limb-geometry datasets acquired in 2010 and 2011 that we used to monitor the seasonal evolution of the vertical profiles of temperature, molecular (C2H2, C2H6, HCN, ..) and aerosol abundances

    Urea cycle disorders in Argentine patients: clinical presentation, biochemical and genetic findings

The present study is the first to thoroughly assess the clinical and molecular profiles of UCD patients examined at a single reference center in Argentina. RESULTS: Forty-nine UCD cases were collected. About half (26/49, 53%) manifested neonatally with classical presentation and had a high mortality (25/26, 96%). Ornithine transcarbamylase deficiency (OTCD) was the most common UCD (26 patients). Argininosuccinate synthetase deficiency (ASSD) was detected in 19 cases, while argininosuccinate lyase deficiency (ASLD) was diagnosed in 4 cases. Molecular genetic analysis revealed 8 private OTC mutations and two large deletion/duplication events in the OTC gene. Most mutations in the ASS1 and ASL genes were recurrent missense changes, and four alterations were novel. The clinical outcome of our UCD cohort was poor, with an overall mortality of 57% (28/49 cases), and a 28% (6/21) disability rate among the survivors. CONCLUSIONS: Most patients in our case series showed severe neonatal onset, with high morbidity/mortality. We detected in total 19 mutations, most of them recurrent and of high frequency worldwide. Noteworthy, we highlight the presence of a geographic cluster with high prevalence of a point mutation in the ASS1 gene. This study suggests that these disorders may be more frequent than commonly assumed, and stresses the need for increased awareness amongst health professionals and greater availability of diagnostic tools for accurate identification, early diagnosis, and timely treatment.This study was supported by grants from the Secretaría de Ciencia y Tecnología, Universidad Nacional de Córdoba and Consejo Nacional de Investigaciones Científicas y Técnicas (CONICET), Argentina.     Detection of interstellar CH_3

    Observations with the Short Wavelength Spectrometer (SWS) onboard the {\it Infrared Space Observatory} (ISO) have led to the first detection of the methyl radical CH3{\rm CH_3} in the interstellar medium. The ν2\nu_2 QQ-branch at 16.5 μ\mum and the RR(0) line at 16.0 μ\mum have been unambiguously detected toward the Galactic center SgrA^*. The analysis of the measured bands gives a column density of (8.0±\pm2.4)×1014\times10^{14} cm2^{-2} and an excitation temperature of (17±2)(17\pm 2) K. Gaseous CO{\rm CO} at a similarly low excitation temperature and C2H2{\rm C_2H_2} are detected for the same line of sight. Using constraints on the H2{\rm H_2} column density obtained from C18O{\rm C^{18}O} and visual extinction, the inferred CH3{\rm CH_3} abundance is (1.3+2.20.7)×108(1.3{{+2.2}\atop{-0.7}}) \times 10^{-8}. The chemically related CH4{\rm CH_4} molecule is not detected, but the pure rotational lines of CH{\rm CH} are seen with the Long Wavelength Spectrometer (LWS). The absolute abundances and the CH3/CH4{\rm CH_3/CH_4} and CH3/CH{\rm CH_3/CH} ratios are inconsistent with published pure gas-phase models of dense clouds. The data require a mix of diffuse and translucent clouds with different densities and extinctions, and/or the development of translucent models in which gas-grain chemistry, freeze-out and reactions of H{\rm H} with polycyclic aromatic hydrocarbons and solid aliphatic material are included.Comment: 2 figures.     Perturbation of serine enantiomers homeostasis in the striatum of MPTP-lesioned monkeys and mice reflects the extent of dopaminergic midbrain degeneration

    Loss of dopaminergic midbrain neurons perturbs L-serine and D-serine homeostasis in the post-mortem caudate putamen (CPu) of Parkinson's disease (PD) patients. However, it is unclear whether the severity of dopaminergic nigrostriatal degeneration plays a role in deregulating serine enantiomers' metabolism. Here, through high -performance liquid chromatography (HPLC), we measured the levels of these amino acids in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated monkeys and MPTP-plus-probenecid (MPTPp)-treated mice to determine whether and how dopaminergic midbrain degeneration affects the levels of serine enantiomers in various basal ganglia subregions. In addition, in the same brain regions, we measured the levels of key neuro-active amino acids modulating glutamatergic neurotransmission, including L-glutamate, glycine, L-aspartate, D- aspartate, and their precursors L-glutamine, L-asparagine. In monkeys, MPTP treatment produced severe denervation of nigrostriatal dopaminergic fibers (⁓75%) and increased the levels of serine enantiomers in the rostral putamen (rPut), but not in the subthalamic nucleus, and the lateral and medial portion of the globus pallidus. Moreover, this neurotoxin significantly reduced the protein expression of the astrocytic serine trans-porter ASCT1 and the glycolytic enzyme GAPDH in the rPut of monkeys. Conversely, concentrations of D-serine and L-serine, as well as ASCT1 and GAPDH expression were unaffected in the striatum of MPTPp-treated mice, which showed only mild dopaminergic degeneration (⁓30%). These findings unveil a link between the severity of dopaminergic nigrostriatal degeneration and striatal serine enantiomers concentration, ASCT1 and GAPDH expression. We hypothesize that the up-regulation of D-serine and L-serine levels occurs as a secondary response within a homeostatic loop to support the metabolic and neurotransmission demands imposed by the degener-ation of dopaminergic neurons
