    Purification and biochemical characterization of a serine alkaline protease TC4 from a new isolated Bacillus alcalophilus TCCC11004 in detergent formulations

    An extracellular alkaline protease producing strain was isolated from alkaline soil and identified as Bacillus alcalophilus TCCC11004 on the basis of 16S rDNA gene sequencing and biochemical properties. The most appropriate medium for the protease production was composed of (g/l): maltodextrin 110, yeast extract 17.5, cotton seed meal 29.3, K2HPO4 18, trisodium citrate 3.3 and CaCl2 2.6. The alkaline protease TC4 was purified from the culture supernatant by ammonium sulfate precipitation, Sephadex G-75 gel filtration and SP-Sepharose HP ion exchange chromatography, with a 6.8 fold increase in specific activity and 15.2% recovery. The molecular weight was estimated to be 26 kDa on SDS-PAGE. The protease was highly active from pH 9.0-12.0 with an optimal at pH 11.0. It was active at 30 - 60°C and exhibited maximal activity at 50°C. The thermostability of the protease was increased by the addition of CaCl2. It retained 70 and 81% of its initial activity after heating for 2 h at 50°C, in the absence or presence of 2 mM CaCl2, respectively. The enzyme was inactivated by diisopropyl fluorophosphate and phenylmethylsulfonyl fluoride, suggesting that it is a serine protease. The protease was stable in 0.5% SDS and retained 70.3% of its initial activity after 1 h of incubation. It was active in the presence of 3% Triton X-100 with 100% activity and stable towards oxidizing agent with 69.2% activity in the presence of 1% H2O2. The enzyme showed excellent compatibility with commercial detergents such as TaiZi, BiLang, DiaoPai and TianQing, retaining more than 90% of its initial activity in the tested detergents after 1 h of preincubation at 40°C.Keywords: Serine alkaline protease, Bacillus alcalophilus, stability, detergent compatibility

    Comparing metrics at large: harmonic vs quo-harmonic coordinates

    To compare two space-times on large domains, and in particular the global structure of their manifolds, requires using identical frames of reference and associated coordinate conditions. In this paper we use and compare two classes of time-like congruences and corresponding adapted coordinates: the harmonic and quo-harmonic classes. Besides the intrinsic definition and some of their intrinsic properties and differences we consider with some detail their differences at the level of the linearized approximation of the field equations. The hard part of this paper is an explicit and general determination of the harmonic and quo-harmonic coordinates adapted to the stationary character of three well-know metrics, Schwarzschild's, Curzon's and Kerr's, to order five of their asymptotic expansions.     Radiative decays of heavy and light mesons in a quark triangle approach

    The radiative meson decays V→PγV\to P\gamma and P→γγP\to \gamma\gamma are analyzed using the quark triangle diagram. Experimental data yield well determined estimates of the universal quark-antiquark-meson couplings gVqqˉ′g_{Vq\bar{q}'} and gPqqˉ′g_{Pq\bar{q}'} for the light meson sector.     Prediction of preterm birth with and without preeclampsia using mid-pregnancy immune and growth-related molecular factors and maternal characteristics.

    OBJECTIVE:To evaluate if mid-pregnancy immune and growth-related molecular factors predict preterm birth (PTB) with and without (±) preeclampsia. STUDY DESIGN:Included were 400 women with singleton deliveries in California in 2009-2010 (200 PTB and 200 term) divided into training and testing samples at a 2:1 ratio. Sixty-three markers were tested in 15-20 serum samples using multiplex technology. Linear discriminate analysis was used to create a discriminate function. Model performance was assessed using area under the receiver operating characteristic curve (AUC). RESULTS:Twenty-five serum biomarkers along with maternal age <34 years and poverty status identified >80% of women with PTB ± preeclampsia with best performance in women with preterm preeclampsia (AUC = 0.889, 95% confidence interval (0.822-0.959) training; 0.883 (0.804-0.963) testing). CONCLUSION:Together with maternal age and poverty status, mid-pregnancy immune and growth factors reliably identified most women who went on to have a PTB ± preeclampsia

    Clades and clans: a comparison study of two evolutionary models

    The Yule-Harding-Kingman (YHK) model and the proportional to distinguishable arrangements (PDA) model are two binary tree generating models that are widely used in evolutionary biology. Understanding the distributions of clade sizes under these two models provides valuable insights into macro-evolutionary processes, and is important in hypothesis testing and Bayesian analyses in phylogenetics. Here we show that these distributions are log-convex, which implies that very large clades or very small clades are more likely to occur under these two models. Moreover, we prove that there exists a critical value Îş(n)\kappa(n) for each nâ©ľ4n\geqslant 4 such that for a given clade with size kk, the probability that this clade is contained in a random tree with nn leaves generated under the YHK model is higher than that under the PDA model if 1<k<Îş(n)1<k<\kappa(n), and lower if Îş(n)<k<n\kappa(n)<k<n. Finally, we extend our results to binary unrooted trees, and obtain similar results for the distributions of clan sizes.Comment: 21page
