Lipid membrane instability and poration driven by capacitive charging
A new model for the interaction of an electric pulse with a lipid membrane is
proposed. Using this model we show that when a DC electric pulse is applied to
an insulating lipid membrane separating fluids with different conductivities,
the capacitive charging current through the membrane drives electrohydrodynamic
flow that destabilizes the membrane. The instability is transient and decays as
the membrane charges. The bulk conductivity mismatch plays an essential role in
this instability because it results in a different rate of charge accumulation
on the membrane's physical surfaces. Shearing stresses created by the electric
field acting on its own induced free charge are non-zero as long as the charge
imbalance exists. Accordingly, the most unstable mode is related to the ratio
of membrane charging time and the electrohydrodynamic time.Comment: 4 pages, 4 figure
Reflections on the Ethical Content of the IT Honours Program Project Module
Part 4: Syllabus DesignInternational audienceHonours programs in South African universities must include a research project module. There are external pressures from professional bodies and government that influences the nature of the project module. This paper presents a reflection on the process of managing the project module at a South African university taking into account internal and external demands. The focus for this paper is on the ethical content of the projects. Examples of projects are presented after which the ethical awareness of the students is discussed. Not all projects have ethical, legal or social issues, however ethical aspects need to be reflected upon and awareness of these issues is essential during the planning of the project. The awareness that students have on ethical, social and legal issues is investigated in this paper
Age modification of the relationship between C-reactive protein and fatigue: findings from Understanding Society (UKHLS)
Background: Systemic inflammation may play a role in the development of idiopathic fatigue, that is, fatigue not explained by infections or diagnosed chronic illness, but this relationship has never been investigated in community studies including the entire adult age span. We examine the association of the inflammatory marker C-reactive protein (CRP) and fatigue assessed annually in a 3-year outcome period for UK adults aged 16–98. Methods: Multilevel models were used to track fatigue 7, 19, and 31 months after CRP measurement, in 10 606 UK individuals. Models accounted for baseline fatigue, demographics, health conditions diagnosed at baseline and during follow-up, adiposity, and psychological distress. Sensitivity analyses considered factors including smoking, sub-clinical disease (blood pressure, anaemia, glycated haemoglobin), medications, ethnicity, and alcohol consumption. Results: Fatigue and CRP increased with age, and women had higher values than men. CRP was associated with future self-reported fatigue, but only for the oldest participants. Thus, in those aged 61–98 years, high CRP ( > 3 mg/L) independently predicted greater fatigue 7, 19, and 31 months after CRP measurement [odds ratio for new-onset fatigue after 7 months: 1.88, 95% confidence interval (CI) 1.21–2.92; 19 months: 2.25, CI 1.46–3.49; 31 months: 1.65, CI 1.07–2.54]. No significant longitudinal associations were seen for younger participants. Conclusions: Our findings support previously described CRP–fatigue associations in older individuals. However, there are clear age modifications in these associations, which may reflect a contribution of unmeasured sub-clinical disease of limited relevance to younger individuals. Further work is necessary to clarify intervening processes linking CRP and fatigue in older individuals
Pointcatcher software:analysis of glacial time-lapse photography and integration with multi-temporal digital elevation models
Terrestrial time-lapse photography offers insight into glacial processes through high spatial and temporal resolution imagery. However, oblique camera views complicate measurement in geographic coordinates, and lead to reliance on specific imaging geometries or simplifying assumptions for calculating parameters such as ice velocity. We develop a novel approach that integrates time-lapse imagery with multi-temporal digital elevation models to derive full 3D coordinates for natural features tracked throughout a monoscopic image sequence. This enables daily independent measurement of horizontal (ice flow) and vertical (ice melt) velocities. By combining two terrestrial laser scanner surveys with a 73-day sequence from Sólheimajökull, Iceland, variations in horizontal ice velocity of ~10% were identified over timescales of ~25 days. An overall surface elevation decrease of ~3.0 m showed rate changes asynchronous with the horizontal velocity variations, demonstrating a temporal disconnect between the processes of ice surface lowering and mechanisms of glacier movement. Our software, ‘Pointcatcher’, is freely available for user-friendly interactive processing of general time-lapse sequences and includes Monte Carlo error analysis and uncertainty projection onto DEM surfaces. It is particularly suited for analysis of challenging oblique glacial imagery, and we discuss good features to track, both for correction of camera motion and for deriving ice velocities
Synonymous codons direct cotranslational folding toward different protein conformations.
In all genomes, most amino acids are encoded by more than one codon. Synonymous codons can modulate protein production and folding, but the mechanism connecting codon usage to protein homeostasis is not known. Here we show that synonymous codon variants in the gene encoding gamma-B crystallin, a mammalian eye-lens protein, modulate the rates of translation and cotranslational folding of protein domains monitored in real time by Forster resonance energy transfer and fluorescence-intensity changes. Gamma-B crystallins produced from mRNAs with changed codon bias have the same amino acid sequence but attain different conformations, as indicated by altered invivo stability and invitro protease resistance. 2D NMR spectroscopic data suggest that structural differences are associated with different cysteine oxidation states of the purified proteins, providing a link between translation, folding, and the structures of isolated proteins. Thus, synonymous codons provide a secondary code for protein folding in the cell
A Global Health Partnership's Use of Time-Limited Support to Catalyze Health Practice Change: The Case of GAVI's Injection Safety Support
This paper presents the findings of a study to assess the effectiveness and sustainability of a GAVI (Global Alliance of Vaccines and Immunization) sponsored, time-limited Injection Safety (INS) support. The support came in two forms: 1) in-kind, in the form of AD syringes and safety boxes, and 2) in cash, for those countries that already had a secure, multi-year source of AD syringes and safety boxes, but proposed to use INS support to strengthen their injection safety activities. In total, GAVI gave INS support for a three-year period to 58 countries: 46 with commodities and 12 with cash support. To identify variables that might be associated with financial sustainability, frequencies and cross-tabulations were run against various programmatic and socio-economic variables in the 58 countries. All but two of the 46 commodity-recipient countries were able to replace and sustain the use of AD syringes and safety boxes after the end of their GAVI INS support despite the fact that standard disposable syringes are less costly than ADs (10–15 percent differential). In addition, all 12 cash-recipient countries continued to use AD syringes and safety boxes in their immunization programs in the years following GAVI INS assistance. At the same time, countries were often not prepared for the increased waste management requirements associated with the use of the syringes, suggesting the importance of anticipating challenges with the introduction of new technologies. The sustained use of AD syringes in countries receiving injection safety support from GAVI, in a majority of cases through government financing, following the completion of three years of time-limited support, represents an early indication of how GHPs can contribute to improved health outcomes in immunization safety in the world's poorest countries in a sustainable way
Interfacial effects on droplet dynamics in poiseuille flow
Many properties of emulsions arise from interfacial rheology, but a theoretical understanding of the effect of interfacial viscosities on droplet dynamics is lacking. Here we report such a theory, relating to isolated spherical drops in a Poiseuille flow. Stokes flow is assumed in the bulk phases, and a jump in hydrodynamic stress at the interface is balanced by Marangoni and surface viscous forces according to the Boussinesq–Scriven constitutive law. Our model employs a linear equation of state for the surfactant. Our analysis predicts slip, cross-stream migration and droplet-circulation velocities. These results and the corresponding interfacial parameters are separable: e.g., cross-stream migration occurs only if gradients in surfactant concentration are present; slip velocity depends on viscosity contrast and dilatational properties, but not on shear Boussinesq number. This separability allows a new and advantageous means to measure surface viscous and elastic forces directly from the drop interface
