Hepatic accumulation of intestinal cholesterol is decreased and fecal cholesterol excretion is increased in mice fed a high-fat diet supplemented with milk phospholipids
<p>Abstract</p> <p>Background</p> <p>Milk phospholipids (PLs) reduce liver lipid levels when given as a dietary supplement to mice fed a high-fat diet. We have speculated that this might be due to reduced intestinal cholesterol uptake.</p> <p>Methods</p> <p>Mice were given a high-fat diet for 3 or 5 weeks that had no added PL or that were supplemented with 1.2% by wt PL from cow's milk. Two milk PL preparations were investigated: a) a PL-rich dairy milk extract (PLRDME), and b) a commercially-available milk PL concentrate (PC-700). Intestinal cholesterol uptake was assessed by measuring fecal and hepatic radioactivity after intragastric administration of [<sup>14</sup>C]cholesterol and [<sup>3</sup>H]sitostanol. Fecal and hepatic lipids were measured enzymatically and by ESI-MS/MS.</p> <p>Results</p> <p>Both PL preparations led to significant decreases in total liver cholesterol and triglyceride (-20% to -60%, <it>P </it>< 0.05). Hepatic accumulation of intragastrically-administered [<sup>14</sup>C]cholesterol was significantly less (-30% to -60%, <it>P </it>< 0.05) and fecal excretion of [<sup>14</sup>C]cholesterol and unlabeled cholesterol was significantly higher in PL-supplemented mice (+15% to +30%, <it>P </it>< 0.05). Liver cholesterol and triglyceride levels were positively correlated with hepatic accumulation of intragastrically-administered [<sup>14</sup>C]cholesterol (<it>P </it>< 0.001) and negatively correlated with fecal excretion of [<sup>14</sup>C]cholesterol (<it>P </it>< 0.05). Increased PL and ceramide levels in the diet of mice supplemented with milk PL were associated with significantly higher levels of fecal PL and ceramide excretion, but reduced levels of hepatic PL and ceramide, specifically, phosphatidylcholine (-21%, <it>P </it>< 0.05) and monohexosylceramide (-33%, <it>P </it>< 0.01).</p> <p>Conclusion</p> <p>These results indicate that milk PL extracts reduce hepatic accumulation of intestinal cholesterol and increase fecal cholesterol excretion when given to mice fed a high-fat diet.</p
Classical Monopoles: Newton, NUT-space, gravomagnetic lensing and atomic spectra
Stimulated by a scholium in Newton's Principia we find some beautiful results
in classical mechanics which can be interpreted in terms of the orbits in the
field of a mass endowed with a gravomagnetic monopole. All the orbits lie on
cones! When the cones are slit open and flattened the orbits are exactly the
ellipses and hyperbolae that one would have obtained without the gravomagnetic
monopole.
The beauty and simplicity of these results has led us to explore the similar
problems in Atomic Physics when the nuclei have an added Dirac magnetic
monopole. These problems have been explored by others and we sketch the
derivations and give details of the predicted spectrum of monopolar hydrogen.
Finally we return to gravomagnetic monopoles in general relativity. We
explain why NUT space has a non-spherical metric although NUT space itself is
the spherical space-time of a mass with a gravomagnetic monopole. We
demonstrate that all geodesics in NUT space lie on cones and use this result to
study the gravitational lensing by bodies with gravomagnetic monopoles.
We remark that just as electromagnetism would have to be extended beyond
Maxwell's equations to allow for magnetic monopoles and their currents so
general relativity would have to be extended to allow torsion for general
distributions of gravomagnetic monopoles and their currents. Of course if
monopoles were never discovered then it would be a triumph for both Maxwellian

Combined Use of Serum Adiponectin and Tumor Necrosis Factor-Alpha Receptor 2 Levels Was Comparable to 2-Hour Post-Load Glucose in Diabetes Prediction
Background: Adipose tissue inflammation and dysregulated adipokine secretion are implicated in obesity-related insulin resistance and type 2 diabetes. We evaluated the use of serum adiponectin, an anti-inflammatory adipokine, and several proinflammatory adipokines, as biomarkers of diabetes risk and whether they add to traditional risk factors in diabetes prediction. Methods: We studied 1300 non-diabetic subjects from the prospective Hong Kong Cardiovascular Risk Factor Prevalence Study (CRISPS). Serum adiponectin, tumor necrosis factor-alpha receptor 2 (TNF-α R2), interleukin-6 (IL-6), adipocyte-fatty acid binding protein (A-FABP) and high-sensitivity C-reactive protein (hsCRP) were measured in baseline samples. Results: Seventy-six participants developed diabetes over 5.3 years (median). All five biomarkers significantly improved the log-likelihood of diabetes in a clinical diabetes prediction (CDP) model including age, sex, family history of diabetes, smoking, physical activity, hypertension, waist circumference, fasting glucose and dyslipidaemia. In ROC curve analysis, "adiponectin + TNF-α R2" improved the area under ROC curve (AUC) of the CDP model from 0.802 to 0.830 (P = 0.03), rendering its performance comparable to the "CDP + 2-hour post-OGTT glucose" model (AUC = 0.852, P = 0.30). A biomarker risk score, derived from the number of biomarkers predictive of diabetes (low adiponectin, high TNF-α R2), had similar performance when added to the CDP model (AUC = 0.829 [95% CI: 0.808-0.849]). Conclusions: The combined use of serum adiponectin and TNF-α R2 as biomarkers provided added value over traditional risk factors for diabetes prediction in Chinese and could be considered as an alternative to the OGTT. © 2012 Woo et al.published_or_final_versio
An investigation of the problem of optimizing a search tactic for a searchlight type sonar.
A searchlight type sonar is one of the systems that
small navies use to counteract the danger which submarines
present to their lines of supply and transport.
In this paper, a standard search pattern for this
type of sonar is compared with search patterns which are
based on a consideration of the tactical value of detecting
a submarine as a function of the relative location of the
submarine.
The results of the comparison suggest that is possible
to increase the effectiveness of a searchlight type sonar
by using a search pattern in which the sweep time allocated

The KCNJ11 E23K Polymorphism and Progression of Glycaemia in Southern Chinese: A Long-Term Prospective Study
Context: The KCNJ11 E23K variant is associated with type 2 diabetes mellitus (T2DM) in cross-sectional studies, but conflicting findings have been reported from prospective studies. Objective: This study aimed to evaluate whether the E23K variant could predict glycaemic progression in a Southern Chinese population. Methods/Principal Findings: We performed a long-term prospective study on 1912 subjects from the Hong Kong Cardiovascular Risk Factors Prevalence Study (CRISPS). The KCNJ11 E23K variant was associated with the progression to prediabetes after a median interval of 12 years on multinomial logistic regression analysis, even after adjustment for traditional risk factors (OR 1.29, P age, sex, BMI and fasting plasma glucose [FPG] adjusted = 0.02). Based on Cox proportional hazard regression analysis, the E23K variant also predicted incident prediabetes (HR 1.18, P age, sex, BMI and FPG adjusted = 0.021). However, E23K was not associated with the progression to T2DM in either multinomial or Cox regression analysis, and the association of E23K with glycaemic progression to either prediabetes or T2DM was significant only in unadjusted Cox regression analysis (P = 0.039). In a meta-analysis of eight prospective studies including our own, involving 15680 subjects, the E23K variant was associated with incident T2DM (fixed effect: OR 1.10, P = 4×10 -3; random effect: OR 1.11, P = 0.035). Conclusions: Our study has provided supporting evidence for the role of the E23K variant in glycaemic progression in Chinese, with its effect being more evident in the early stage of T2DM, as the subjects progressed from normal glucose tolerance to prediabetes. © 2011 Cheung et al.published_or_final_versio
Understanding Work Practices of Autonomous Agile Teams: A Social-psychological Review
The purpose of this paper is to suggest additional aspects of social
psychology that could help when making sense of autonomous agile teams. To make
use of well-tested theories in social psychology and instead see how they
replicated and differ in the autonomous agile team context would avoid
reinventing the wheel. This was done, as an initial step, through looking at
some very common agile practices and relate them to existing findings in
social-psychological research. The two theories found that I argue could be
more applied to the software engineering context are social identity theory and
group socialization theory. The results show that literature provides
social-psychological reasons for the popularity of some agile practices, but
that scientific studies are needed to gather empirical evidence on these
under-researched topics. Understanding deeper psychological theories could
provide a better understanding of the psychological processes when building
autonomous agile team, which could then lead to better predictability and
intervention in relation to human factors
Evaluation of the combined use of adiponectin and C-reactive protein levels as biomarkers for predicting the deterioration in glycaemia after a median of 5.4 years
Aims/hypothesis: Hypoadiponectinaemia and raised C-reactive protein (CRP) level are obesity-related biomarkers associated with glucose dysregulation. We evaluated the combined use of these two biomarkers in predicting the deterioration of glycaemia in a prospective study after a median of 5.4 years. Methods: In total 1,288 non-diabetic participants from the Hong Kong Cardiovascular Risk Factor Prevalence Study-2, with high-sensitivity CRP (hsCRP) and total adiponectin levels measured were included. OGTT was performed in all participants. Two hundred and six participants had deterioration of glycaemia at follow-up, whereas 1,082 participants did not. Results: Baseline age, hsCRP and adiponectin levels were significant independent predictors of the deterioration of glycaemia in a Cox regression analysis after adjusting for baseline age, sex, BMI, hypertension, triacylglycerols, 2 h post-OGTT glucose and homeostasis model assessment of insulin resistance index (all p < 0.01). The introduction of hsCRP or adiponectin level to a regression model including the other biomarker improved the prediction of glycaemic progression significantly in all participants, especially in women (all p < 0.01). The combined inclusion of the two biomarkers resulted in a modest improvement in model discrimination, compared with the inclusion of either one alone. Among participants with impaired fasting glucose/impaired glucose tolerance (IFG/IGT) at baseline, hsCRP and adiponectin levels were not predictive of progression or improvement of glycaemic status. Conclusions/interpretation: Adiponectin and hsCRP levels are independent factors in predicting the deterioration of glycaemia, supporting the role of adiposity-related inflammation in the development of type 2 diabetes. A novel immunomodulatory function of neutrophils on rhinovirus-Activated monocytes in vitro
© 2016 Published by the BMJ Publishing Group Limited. Background Rhinovirus (RV) infections are the major precipitant of asthma exacerbations. While neutrophilic lung inflammation occurs during such infections, its role remains unclear. Neutrophilic inflammation is associated with increased asthma severity and steroid refractory disease. Neutrophils are vital for controlling infections but also have immunomodulatory functions. Previously, we found that neutrophils respond to viral mimetics but not replication competent RV. We aimed to investigate if neutrophils are activated and/or modulate immune responses of monocytes during RV16 infection. Methods Primary human monocytes and autologous neutrophils were cocultured with or without RV16, in direct contact or separated by transwells. RV16-stimulated monocytes were also exposed to lysed neutrophils, neutrophil membrane components or soluble neutrophil intracellular components. Interleukin 6 (IL-6) and C-X-C motif (CXC)L8 mRNA and proteins were measured by quantitative PCR and ELISA at 24â €..hours. Results RV16 induced IL-6 and CXCL8 in monocytes, but not neutrophils. RV16-induced IL-6 and CXCL8 from monocytes was reduced in the presence of live neutrophils. Transwell separation abolished the inhibitory effects. Lysed neutrophils inhibited RV16-induced IL-6 and CXCL8 from monocytes. Neutrophil intracellular components alone effectively inhibited RV16-induced monocyte-derived IL-6 and CXCL8. Neutrophil intracellular components reduced RV16-induced IL-6 and CXCL8 mRNA in monocytes. Conclusions Cell contact between monocytes and neutrophils is required, and preformed neutrophil mediator(s) are likely to be involved in the suppression of cytokine mRNA and protein production. This study demonstrates a novel regulatory function of neutrophils on RV-Activated monocytes in vitro, challenging the paradigm that neutrophils are predominantly proinflammatory
Search for effective Lorentz and CPT violation using ZEUS data
Lorentz and CPT symmetry in the quark sector of the Standard Model are studied in the context of an effective field theory using ZEUS e±p data. Symmetry-violating effects can lead to time-dependent oscillations of otherwise time-independent observables, including scattering cross sections. An analysis using five years of inclusive neutral-current deep inelastic scattering events corresponding to an integrated HERA luminosity of 372 pb-1 at s=318 GeV has been performed. No evidence for oscillations in sidereal time has been observed within statistical and systematic uncertainties. Constraints, most for the first time, are placed on 42 coefficients parametrizing dominant CPT-even dimension-four and CPT-odd dimension-five spin-independent modifications to the propagation and interaction of light quarks
