    Roles of a forest corridor between Marojejy, Anjanaharibe- Sud and Tsaratanana protected areas, northern Madagascar, in maintaining endemic and threatened Malagasy taxa

    Site-based conservation is widely recognized as a fundamental requirement for the maintenance of  biodiversity. We carried out a rapid biological assessment from 17 March to 14 May 2011 in the southwestern part of the provisionally protected rainforest corridor between Marojejy, Anjanaharibe-Sud and Tsaratanana protected areas (COMATSA), northern Madagascar, to assess its biodiversity richness and species conservation status. We recorded 248 endemic vertebrate species, including 80 amphibians, 52 reptiles, 79 birds, 27 small mammals and 10 lemurs. Of these 248 species, 36 are threatened including one Critically Endangered (one lemur), nine Endangered (two amphibians, four reptiles and three lemurs) and 26 Vulnerable (10 amphibians, six reptiles, four birds, one small mammal and five lemurs). For herpetofauna species, the pair COMATSAc –cMarojejy was the only site that had a similarity value greater than 0.50. For birds and  lemurs, values of similarity in three pairs of sites COMATSAc –c Marojejy, COMATSA c –c Anjanaharibe-Sud and Marojejyc –c Anjanaharibe-Sud exceeded those of herpetofauna in COMATSAc –c Marojejy. The inclusion of the COMATSA into the new protected area network in Madagascar is supported by our findings. Il est dorénavant largement reconnu que pour maintenir la biodiversité, il est fondamental de réaliser la  conservation de la nature au niveau des sites. Un inventaire biologique rapide a été mené du 17 mars au 14 mai 2011 dans la partie sud-ouest du couloir forestier reliant les parcs et réserves du Marojejy,  d’Anjanaharibe- Sud et du Tsaratanana (COMATSA). L’inventaire de ce couloir qui bénéficie d’un statut de protection temporaire dans la partie septentrionale de Madagascar, était destiné à évaluer la richesse de sa biodiversité et le statut de conservation des espèces de vertébrés qu’il héberge. Les résultats ont   montré que parmi les 248 espèces endémiques de vertébrés recensées, 80 sont des amphibiens, 52 des  reptiles, 79 des oiseaux, 27 des petits mammifères mammifères et 10 des lémuriens. Sur ces 248  espèces, 36 sont menacées, dont une espèce qui est en Danger Critique d’Extinction (lémurien), neuf en Danger (deux amphibiens, quatre reptiles et trois lémuriens) et 26 Vulnérables (1 0 amphibiens, six  reptiles, quatre oiseaux, un micromammifère et cinq lémuriens). Ce couloir forestier présente un  coefficient de similarité élevé, d’une valeur supérieure à 0.50 avec le Marojejy pour les amphibiens et les reptiles. Pour les oiseaux et les lémuriens, les coefficients sont encore plus importants, plus  particulièrement dans les comparaisons COMATSAc –c Marojejy, COMATSAc –c Anjanaharibe- Sud et Marojejyc –c Anjanaharibe-Sud.     Epidemiological characteristics of cryptococcal meningoencephalitis associated with Cryptococcus neoformans var. grubii from HIV-infected patients in Madagascar : a cross-sectional study

    Cryptococcal meningoencephalitis (CM) remains the most prevalent invasive fungal infection worldwide. The main objective of this study was to describe the prevalence of CM and cryptococcal infection in HIV-infected patients in Madagascar. The secondary objectives were to assess the adjusted prevalence of CM according to clinical presentation and patient characteristics, to determine crude 90-day survival according to cryptococcal antigen (CrAg) status and CM, and to identify the genotypes of Cryptococcus clinical isolates. This cross-sectional study was carried out at two urban hospitals in Antananarivo (central highlands) and Toamasina (east coast) between November 2014 and December 2016. Consecutive HIV-infected adults presenting with CD4 cell counts 64200/\u3bcl were enrolled. Lateral flow immunoassays of CrAg were performed on serum for all patients, and on cerebrospinal fluid for patients with CM symptoms. MALDI-ToF MS, ITS sequencing, and determinations of the molecular and mating types of the isolates were performed. Fluconazole is the only drug for CM treatment available in Madagascar. Patients were treated orally, with high doses (1200 mg/day) for 10-12 weeks and then with 200 mg/day. Minimum inhibitory concentrations were determined for amphotericin B, flucytosine, voriconazole and fluconazole in E-tests. Overall prevalence was 13.2% (95% CI 7.9-20.3) for cryptococcal infection and 10.9% (95% CI 6.1-17.5) for CM, among the 129 HIV-infected patients studied. The 90-day mortality rate was 58.8% (10/17) in CrAg-positive patients and 17.9% (20/112) in CrAg-negative patients (p<0.001). The 13 Cryptococcus strains obtained at baseline were all Cryptococcus neoformans var. grubii, genotypes VNI-\u3b1A (3 isolates), VNII-\u3b1A (4 isolates) or hybrid VNI/VNII-\u3b1AA\u3b1 (6 isolates), suggesting high diversity. Two strains acquired fluconazole resistance after four and five months of exposure, respectively. The prevalence of cryptococcosis is high in Madagascar and this serious condition is life-threatening in HIV-infected patients. These findings will be used to raise the awareness of national authorities to strengthen the national HIV/AIDS control program

    Taenia solium porcine cysticercosis in Madagascar: Comparison of immuno-diagnostic techniques and estimation of the prevalence in pork carcasses traded in Antananarivo city

    Taenia solium cysticercosis was reported in official veterinary and medical statistics to be highly prevalent in pigs and humans in Madagascar, but few estimates are available for pigs. This study aimed to estimate the seroprevalence of porcine cysticercosis among pigs slaughtered in Antananarivo abattoirs. Firstly, the diagnostic performance of two antigen-ELISA techniques (B158B60 Ag-ELISA and HP10 Ag-ELISA) and an immunoblotting method were compared with meat inspection procedures on a sample of pigs suspected to be infected with (group 1; n = 250) or free of (group 2; n = 250) T. solium based on direct veterinary inspection in Madagascar. Sensitivity and specificity of the antigen ELISAs were then estimated using a Bayesian approach for detection of porcine cysticercosis in the absence of a gold standard. Then, a third set of pig sera (group 3, n = 250) was randomly collected in Antananarivo slaughterhouses and tested to estimate the overall prevalence of T. solium contamination in pork meat traded in Antananarivo. The antigen ELISAs showed a high sensitivity (>84%), but the B158B60 Ag-ELISA appeared to be more specific than the HP10 Ag-ELISA (model 1: 95% vs 74%; model 2: 87% vs 71%). The overall prevalence of porcine cysticercosis in Antananarivo slaughterhouses was estimated at 2.3% (95% credibility interval [95%CrI]: 0.09–9.1%) to 2.6% (95%CrI: 0.1–10.3%) depending on the model and priors used. Since the sample used in this study is not representative of the national pig population, village-based surveys and longitudinal monitoring at slaughter are needed to better estimate the overall prevalence, geographical patterns and main risk factors for T. solium contamination, in order to improve control policies.     Competing biosecurity and risk rationalities in the Chittagong poultry commodity chain, Bangladesh

    This paper anthropologically explores how key actors in the Chittagong live bird trading network perceive biosecurity and risk in relation to avian influenza between production sites, market maker scenes and outlets. They pay attention to the past and the present, rather than the future, downplaying the need for strict risk management, as outbreaks have not been reported frequently for a number of years. This is analysed as ‘temporalities of risk perception regarding biosecurity’, through Black Swan theory, the idea that unexpected events with major effects are often inappropriately rationalized (Taleb in The Black Swan. The impact of the highly improbable, Random House, New York, 2007). This incorporates a sociocultural perspective on risk, emphasizing the contexts in which risk is understood, lived, embodied and experienced. Their risk calculation is explained in terms of social consent, practical intelligibility and convergence of constraints and motivation. The pragmatic and practical orientation towards risk stands in contrast to how risk is calculated in the avian influenza preparedness paradigm. It is argued that disease risk on the ground has become a normalized part of everyday business, as implied in Black Swan theory. Risk which is calculated retrospectively is unlikely to encourage investment in biosecurity and, thereby, points to the danger of unpredictable outlier events

    A large-scale study of a poultry trading network in Bangladesh: implications for control and surveillance of avian influenza viruses

    Since its first report in 2007, avian influenza (AI) has been endemic in Bangladesh. While live poultry marketing is widespread throughout the country and known to influence AI dissemination and persistence, trading patterns have not been described. The aim of this study is to assess poultry trading practices and features of the poultry trading networks which could promote AI spread, and their potential implications for disease control and surveillance. Data on poultry trading practices was collected from 849 poultry traders during a cross-sectional survey in 138 live bird markets (LBMs) across 17 different districts of Bangladesh. The quantity and origins of traded poultry were assessed for each poultry type in surveyed LBMs. The network of contacts between farms and LBMs resulting from commercial movements of live poultry was constructed to assess its connectivity and to identify the key premises influencing it

SUPPLEMENTARY CODE 1: Example of an R code to correct for pseudoreplication.Bushpigs (Potamochoerus larvatus) play a major role in the socio-ecosystem of Madagascar, particularly in rural areas. They are largely hunted by rural populations as a major source of income and protein. They can also represent a potential source of pathogens for domestic animals and people. For example, it is hypothesized that bushpigs might compromise African swine fever (ASF) eradication programs by sporadically transmitting the virus to domestic pigs. However, available knowledge on the distribution of bushpigs in Madagascar is limited. In this study, we estimated the distribution of bushpigs on Madagascar using a species distribution model (SDM). We retrieved 206 sightings of bushpigs in Madagascar during 1990–2016 and predicted the distribution by using 37 climatic, geographic, and agricultural/human variables related to the presence of bushpigs and running a presence-background maximum entropy SDM. Our model identified three main areas with a high suitability for bushpigs: in the north, central-western, and east of the island (AUC = 0.84). The main contributors to the model were the vegetation index (51.3%), percentage of land covered by trees (17.6%), and annual averaged monthly precipitation (12.6%). In addition, we identified areas in central Madagascar with a high density of domestic pigs and a high suitability score for bushpigs. These results may help to identify bushpig areas at the interface with domestic pigs to assess the risk of pathogen transmission and to design ecological assessments, wildlife management studies, or targeted surveillance and research studies related to many bushpig-borne pathogens, such as ASF, which is an endemic problem in the country, as well as zoonotic diseases such as cysticercosis and hepatitis E.
