Dominant negative phenotype of Bacillus thuringiensis Cry1Ab, Cry11Aa and Cry4Ba mutants suggest hetero-oligomer formation among different Cry toxins.
Background - Bacillus thuringiensis Cry toxins are used worldwide in the control of different insect pests important in agriculture or in human health. The Cry proteins are pore-forming toxins that affect the midgut cell of target insects. It was shown that non-toxic Cry1Ab helix a-4 mutants had a dominant negative (DN) phenotype inhibiting the toxicity of wildtype Cry1Ab when used in equimolar or sub-stoichiometric ratios (1:1, 0.5:1, mutant:wt) indicating that oligomer formation is a key step in toxicity of Cry toxins. Methodology/Principal Findings - The DN Cry1Ab-D136N/T143D mutant that is able to block toxicity of Cry1Ab toxin, was used to analyze its capacity to block the activity against Manduca sexta larvae of other Cry1 toxins, such as Cry1Aa, Cry1Ac, Cry1Ca, Cry1Da, Cry1Ea and Cry1Fa. Cry1Ab-DN mutant inhibited toxicity of Cry1Aa, Cry1Ac and Cry1Fa. In addition, we isolated mutants in helix a-4 of Cry4Ba and Cry11Aa, and demonstrate that Cry4Ba-E159K and Cry11Aa-V142D are inactive and completely block the toxicity against Aedes aegypti of both wildtype toxins, when used at sub-stoichiometric ratios, confirming a DN phenotype. As controls we analyzed Cry1Ab-R99A or Cry11Aa-E97A mutants that are located in helix a-3 and are affected in toxin oligomerization. These mutants do not show a DN phenotype but were able to block toxicity when used in 10:1 or 100:1 ratios (mutant:wt) probably by competition of binding with toxin receptors. Conclusions/Significance - We show that DN phenotype can be observed among different Cry toxins suggesting that may interact in vivo forming hetero-oligomers. The DN phenotype cannot be observed in mutants affected in oligomerization, suggesting that this step is important to inhibit toxicity of other toxin
A nongravitational wormhole
Using the effective metric formalism for photons in a nonlinear
electromagnetic theory, we show that a certain field configuration in
Born-Infeld electromagnetism in flat spacetime can be interpreted as an
ultrastatic spherically symmetric wormhole. We also discuss some properties of
the effective metric that are valid for any field configuration.Comment: LaTex, 9 pages with 5 figures, minor changes, accepted for

Dynamics for a 2-vertex Quantum Gravity Model
We use the recently introduced U(N) framework for loop quantum gravity to
study the dynamics of spin network states on the simplest class of graphs: two
vertices linked with an arbitrary number N of edges. Such graphs represent two
regions, in and out, separated by a boundary surface. We study the algebraic
structure of the Hilbert space of spin networks from the U(N) perspective. In
particular, we describe the algebra of operators acting on that space and
discuss their relation to the standard holonomy operator of loop quantum
gravity. Furthermore, we show that it is possible to make the restriction to
the isotropic/homogeneous sector of the model by imposing the invariance under
a global U(N) symmetry. We then propose a U(N) invariant Hamiltonian operator
and study the induced dynamics. Finally, we explore the analogies between this
model and loop quantum cosmology and sketch some possible generalizations of
it.Comment: 28 pages, v2: typos correcte
Running Coupling with Minimal Length
In models with large additional dimensions, the GUT scale can be lowered to
values accessible by future colliders. Due to modification of the loop
corrections from particles propagating into the extra dimensions, the
logarithmic running of the couplings of the Standard Model is turned into a
power law. These loop-correction are divergent and the standard way to achieve
finiteness is the introduction of a cut-off. The question remains, whether the
results are reliable as they depend on an unphysical parameter.
In this paper, we show that this running of the coupling can be calculated
within a model including the existence of a minimal length scale. The minimal
length acts as a natural regulator and allows us to confirm cut-off
computations.Comment: 26 pages, 5 figures, typos corrected, replaced with published versio
Vortex nucleation as a case study of symmetry breaking in quantum systems
Mean-field methods are a very powerful tool for investigating weakly
interacting many-body systems in many branches of physics. In particular, they
describe with excellent accuracy trapped Bose-Einstein condensates. A generic,
but difficult question concerns the relation between the symmetry properties of
the true many-body state and its mean-field approximation. Here, we address
this question by considering, theoretically, vortex nucleation in a rotating
Bose-Einstein condensate. A slow sweep of the rotation frequency changes the
state of the system from being at rest to the one containing one vortex. Within
the mean-field framework, the jump in symmetry occurs through a turbulent phase
around a certain critical frequency. The exact many-body ground state at the
critical frequency exhibits strong correlations and entanglement. We believe
that this constitutes a paradigm example of symmetry breaking in - or change of
the order parameter of - quantum many-body systems in the course of adiabatic
evolution.Comment: Minor change
Holomorphic Simplicity Constraints for 4d Spinfoam Models
Within the framework of spinfoam models, we revisit the simplicity
constraints reducing topological BF theory to 4d Riemannian gravity. We use the
reformulation of SU(2) intertwiners and spin networks in term of spinors, which
has come out from both the recently developed U(N) framework for SU(2)
intertwiners and the twisted geometry approach to spin networks and spinfoam
boundary states. Using these tools, we are able to perform a
holomorphic/anti-holomorphic splitting of the simplicity constraints and define
a new set of holomorphic simplicity constraints, which are equivalent to the
standard ones at the classical level and which can be imposed strongly on
intertwiners at the quantum level. We then show how to solve these new
holomorphic simplicity constraints using coherent intertwiner states. We
further define the corresponding coherent spin network functionals and
introduce a new spinfoam model for 4d Riemannian gravity based on these
holomorphic simplicity constraints and whose amplitudes are defined from the
evaluation of the new coherent spin networks.Comment: 27 page
Models of Neutrino Masses and Mixings
We review theoretical ideas, problems and implications of neutrino masses and
mixing angles. We give a general discussion of schemes with three light
neutrinos. Several specific examples are analyzed in some detail, particularly
those that can be embedded into grand unified theories.Comment: 44 pages, 2 figures, version accepted for publication on the Focus

Bose-Einstein correlations in hadron-pairs from lepto-production on nuclei ranging from hydrogen to xenon
Bose-Einstein correlations of like-sign charged hadrons produced in
deep-inelastic electron and positron scattering are studied in the HERMES
experiment using nuclear targets of H, H, He, He, N, Ne, Kr,
and Xe. A Gaussian approach is used to parametrize a two-particle correlation
function determined from events with at least two charged hadrons of the same
sign charge. This correlation function is compared to two different empirical
distributions that do not include the Bose-Einstein correlations. One
distribution is derived from unlike-sign hadron pairs, and the second is
derived from mixing like-sign pairs from different events. The extraction
procedure used simulations incorporating the experimental setup in order to
correct the results for spectrometer acceptance effects, and was tested using
the distribution of unlike-sign hadron pairs. Clear signals of Bose-Einstein
correlations for all target nuclei without a significant variation with the
nuclear target mass are found. Also, no evidence for a dependence on the
invariant mass W of the photon-nucleon system is found when the results are
compared to those of previous experiments
Loop quantum gravity: the first twenty five years
This is a review paper invited by the journal "Classical ad Quantum Gravity"
for a "Cluster Issue" on approaches to quantum gravity. I give a synthetic
presentation of loop gravity. I spell-out the aims of the theory and compare
the results obtained with the initial hopes that motivated the early interest
in this research direction. I give my own perspective on the status of the
program and attempt of a critical evaluation of its successes and limits.Comment: 24 pages, 3 figure
