    The shape of a woman's life: Lady Anne Barnard's <i>Memoir</i>

    In her old age, Lady Anne Barnard revised the diaries and letters which remained from her life in Scotland, London, and at the Cape. She shaped part of these records into a six-volume Memoir and since a man's memoir would concentrate attention on his public life and achievements, she cast it into a shape which recalls that of the female’ novel of the eighteenth century. She therefore presents her rejection of the lifestyle of her family, that of impoverished Scottish aristocrats, and her move to London, as a quest for a suitable marriage. This view of her life was not, however, entirely congenial to her: she had a strong sense of her achievements as an autonomous individual, rather than as the lesser partner in a marriage, and although the Memoir ends with her marriage, there is much evidence in it of her pride in having originated and achieved for herself a new lifestyle

    Feasibility of using microbeads with holographic barcodes to track DNA specimens in the clinical molecular laboratory

    We demonstrate the feasibility of using glass microbeads with a holographic barcode identifier to track DNA specimens in the molecular pathology laboratory. These beads can be added to peripheral blood specimens and are carried through automated DNA extraction protocols that use magnetic glass particles. We found that an adequate number of microbeads are consistently carried over during genomic DNA extraction to allow specimen identification, that the beads do not interfere with the performance of several different molecular assays, and that the beads and genomic DNA remain stable when stored together under regular storage conditions in the molecular pathology laboratory. The beads function as an internal, easily readable specimen barcode. This approach may be useful for identifying DNA specimens and reducing errors associated with molecular laboratory testing

    Las restricciones en el acceso a abortos seguros constituyen inequidades en el acceso a la salud integral de las mujeres basadas en las discriminaciones de género. Desde el marco de la teoría feminista, la epidemiología y la psicología social comunitaria crítica, este estudio tiene el objetivo de analizar los nudos críticos identificados por los/as psicólogos/as que participan en equipos de salud en servicios del subsistema público en el marco de la Interrupción Legal del Embarazo. Con un enfoque de Investigación cualitativa y de género, se desarrolló un estudio exploratorio-descriptivo con 7 psicólogas y 5 profesionales de equipos interdisciplinarios de Consejerías de Salud Sexual y Reproductiva con las que se realizaron entrevistas en profundidad y grupos focales. En los resultados se identificaron los obstáculos económicos, institucionales y disciplinares; la decisión como derecho de las mujeres; y, las innovaciones y soportes de las prácticas comprometidas.Restrictions on access to safe abortions constitute inequities in women’s access to comprehensive health based on gender discrimination. With a qualitative and gender research approach, an exploratory-descriptive study was developed with 7 psychologists and 5 interdisciplinary team professionals from Sexual and Reproductive Health Ministries with in-depth interviews and focus groups.     Antiplasmodial Properties and Cytotoxicity of Endophytic Fungi from Symphonia globulifera (Clusiaceae)

Natural products represent credible sources of new antiplasmodial agents for antimalarial drug development. Endophytes that widely colonize healthy tissues of plants have been shown to synthesize a great variety of secondary metabolites that might possess antiplasmodial benefits. The present study was carried out to evaluate the antiplasmodial potential of extracts from endophytic fungi isolated from Symphonia globulifera against a chloroquine-resistant strain of Plasmodium falciparum (PfINDO). Sixty-one fungal isolates with infection frequency of 67.77% were obtained from the bark of S. globulifera. Twelve selected isolates were classified into six different genera including Fusarium, Paecilomyces, Penicillium, Aspergillus, Mucor, and Bipolaris. Extracts from the 12 isolates were tested against PfINDO, and nine showed good activity (IC50 < 10 mu g.mL(-1)) with three fungi including Paecilomyces lilacinus (IC50 = 0.44 mu g.mL(-1)), Penicillium janthinellum (IC50 = 0.2 mu g.mL(-1)), and Paecilomyces sp. (IC50 = 0.55 mu g.mL(-1)) showing the highest promise. These three isolates were found to be less cytotoxic against the HEK293T cell line with selectivity indices ranging from 24.52 to 70.56. Results from this study indicate that endophytic fungi from Symphonia globulifera are promising sources of hit compounds that might be further investigated as novel drugs against malaria. Molecules. 2020;25(12): 2862.The MeOH/CH2Cl2 (1:1) extracts of the roots and leaves of Beilschmiedia louisii and B. obscura showed potent antitrypanosomal activity during preliminary screening on Trypanosoma brucei brucei. Phytochemical investigation of these extracts led to the isolation of a mixture of two new endiandric acid derivatives beilschmiedol B (1) and beilschmiedol C (2), and one new phenylalkene obscurene A (3) together with twelve known compounds (4–15). In addition, four new derivatives (11a–11d) were synthesized from compound 11. Their structures were elucidated based on their NMR and MS data. Compounds 5, 6, and 7 were isolated for the first time from the Beilschmiedia genus. Additionally, the NMR data of compound 4 are given here for the first time. The isolates were evaluated for their antitrypanosomal and antimalarial activities against Tb brucei and the Plasmodium falciparum chloroquine-resistant strain Pf3D7 in vitro, respectively. From the tested compounds, the mixture of new compounds 1 and 2 exhibited the most potent antitrypanosomal activity in vitro with IC50 value of 4.91 μM

Preprints. 2016.The chemical investigation of the anti-yeast methanol extract from the stem bark of Terminalia mantaly led to the isolation of seven compounds: 3-O-methyl-4-O-&amp;alpha;-rhamnopyranoside ellagic acid (1), 3-O-mehylellagic acid (2), arjungenin or 2,3,19,23-tetrahydroxyolean-12-en-28-o&amp;iuml;c acid (3), arjunglucoside or 2,3,19,23-tetrahydroxyolean-12-en-28-o&amp;iuml;c acid glucopyranoside (4), 2&amp;alpha;,3&amp;alpha;,24-trihydroxyolean-11,13(18)-dien-28-o&amp;iuml;c acid (5), stigmasterol (6), stigmasterol 3-O-&amp;beta;-D-glucopyranoside (7). Their structures were established by means of spectroscopic analysis and comparison with published data. Compounds 1-5 were tested in vitro for activity against three pathogenic yeast isolates, Candida albicans, Candida parapsilosis and Candida krusei. The activity of compounds 1, 2 and 4 were comparable to that of the reference compound fluconazole (MIC values below 32 &amp;micro;g/ml) against the three tested yeast isolates. They were also tested for inhibitory properties against four enzymes of metabolic significance: Glucose-6-Phosphate Deshydrogenase (G6PD), human erythrocyte Carbonic anhydrase I and II (hCA I and hCA II), Glutathione S-transferase (GST). Compound 4 showed highly potent inhibitory property against the four tested enzymes with overall IC50 values below 4 &amp;micro;M and inhibitory constant (Ki) &amp;lt;3 &amp;micro;M.</jats:p

    Mesenchymal Stromal Cell on Liver Decellularised Extracellular Matrix for Tissue Engineering

    Background: In end-stage chronic liver disease, transplantation represents the only curative option. However, the shortage of donors results in the death of many patients. To overcome this gap, it is mandatory to develop new therapeutic options. In the present study, we decellularised pig livers and reseeded them with allogeneic porcine mesenchymal stromal cells (pMSCs) to understand whether extracellular matrix (ECM) can influence and/or promote differentiation into hepatocyte-like cells (HLCs). Methods: After decellularisation with SDS, the integrity of ECM-scaffolds was examined by histological staining, immunofluorescence and scanning electron microscope. DNA quantification was used to assess decellularisation. pMSCs were plated on scaffolds by static seeding and maintained in in vitro culture for 21 days. At 3, 7, 14 and 21 days, seeded ECM scaffolds were evaluated for cellular adhesion and growth. Moreover, the expression of specific hepatic genes was performed by RT-PCR. Results: The applied decellularisation/recellularisation protocol was effective. The number of seeded pMSCs increased over the culture time points. Gene expression analysis of seeded pMSCs displayed a weak induction due to ECM towards HLCs. Conclusions: These results suggest that ECM may address pMSCs to differentiate in hepatocyte-like cells. However, only contact with liver-ECM is not enough to induce complete differentiation

    Plant-Derived Compounds against Microbial Infections and Cancers

    Plants synthesize and preserve a variety of metabolites known as natural products. Many of them are easily extractable and can be used as starting material or chemical scaffolds for various purposes, especially in drug discovery. Numbers of reports have listed valuable candidates with privilege scaffolds currently in active development as drugs. New compounds with anticancer and antiinfective activities have been discovered recently, some presented these backbones. The present book chapter aims to highlight these findings from plants which can be considered valuable for the development of new drugs against malignant cells and infective diseases. Interest in anti-infective agents is increasing due to the resistance of microorganisms to existing drugs and newly emerging infectious diseases. This resistance is also, nowadays, associated to some forms of cancers. In addition, the value of plants as essential part in the health care pipeline in low- and middle-income countries is under consideration even though these countries are almost all surrounded by a rich and untapped biodiversity. People are always relying on “modern drugs and treatment” which is unfortunately not affordable to all. Therefore, the present compilation of data on plant-derived compounds can inspire the formulation of ameliorated traditional medicines (ATM) against the targeted diseases and the conservation of species

    Anticancer Activities of Six Selected Natural Compounds of Some Cameroonian Medicinal Plants

    BACKGROUND: Natural products are well recognized as sources of drugs in several human ailments. In the present work, we carried out a preliminary screening of six natural compounds, xanthone V(1) (1); 2-acetylfuro-1,4-naphthoquinone (2); physcion (3); bisvismiaquinone (4); vismiaquinone (5); 1,8-dihydroxy-3-geranyloxy-6-methylanthraquinone (6) against MiaPaCa-2 pancreatic and CCRF-CEM leukemia cells and their multidrug-resistant subline, CEM/ADR5000. Compounds 1 and 2 were then tested in several other cancer cells and their possible mode of action were investigated. METHODOLOGY/FINDINGS: The tested compounds were previously isolated from the Cameroonian medicinal plants Vismia laurentii (1, 3, 4, 5 and 6) and Newbouldia laevis (2). The preliminary cytotoxicity results allowed the selection of xanthone V(1) and 2-acetylfuro-1,4-naphthoquinone, which were then tested on a panel of cancer cell lines. The study was also extended to the analysis of cell cycle distribution, apoptosis induction, caspase 3/7 activation and the anti-angiogenic properties of xanthone V(1) and 2-acetylfuro-1,4-naphthoquinone. IC(50) values around or below 4 µg/ml were obtained on 64.29% and 78.57% of the tested cancer cell lines for xanthone V(1) and 2-acetylfuro-1,4-naphthoquinone, respectively. The most sensitive cell lines (IC(50)<1 µg/ml) were breast MCF-7 (to xanthone V(1)), cervix HeLa and Caski (to xanthone V(1) and 2-acetylfuro-1,4-naphthoquinone), leukemia PF-382 and melanoma colo-38 (to 2-acetylfuro-1,4-naphthoquinone). The two compounds showed respectively, 65.8% and 59.6% inhibition of the growth of blood capillaries on the chorioallantoic membrane of quail eggs in the anti-angiogenic assay. Upon treatment with two fold IC(50) and after 72 h, the two compounds induced cell cycle arrest in S-phase, and also significant apoptosis in CCRF-CEM leukemia cells. Caspase 3/7 was activated by xanthone V(1). CONCLUSIONS/SIGNIFICANCE: The overall results of the present study provided evidence for the cytotoxicity of compounds xanthone V(1) and 2-acetylfuro-1,4-naphthoquinone, and bring supportive data for future investigations that will lead to their use in cancer therapy
