    Fibronectin in immune responses in organ transplant recipients.

    The immune response to an organ allograft involves perpetuation of T cell infiltration and activation. Advances in understanding the mechanisms of T cell activation have placed particular emphasis on the interactions between the T-cell receptor and antigen presenting cells, with little reference to the fact that in vivo activation occurs in the physical context of extracellular matrix proteins (ECM). Indeed, the possibility that ECM proteins may have a determining role in lymphocyte adhesion and tissue localization and function is now becoming more appreciated in view of growing evidence indicating that integrins and other T cell antigens bind ECM components, with some of these components exerting synergistic effects on T-cell activation. This review focuses on the importance of interactions between lymphocytes and fibronectin, a prominent ECM component, for cell migration and function in organ allograft recipients. It explores novel therapeutic approaches based on the assumption that fibronectin represents an active element in the process of T cell activation in the immune cascade triggered by organ transplantation

    Site-specific genomic (SSG) and random domain-localized (RDL) mutagenesis in yeast

    BACKGROUND: A valuable weapon in the arsenal available to yeast geneticists is the ability to introduce specific mutations into yeast genome. In particular, methods have been developed to introduce deletions into the yeast genome using PCR fragments. These methods are highly efficient because they do not require cloning in plasmids. RESULTS: We have modified the existing method for introducing deletions in the yeast (S. cerevisiae) genome using PCR fragments in order to target point mutations to this genome. We describe two PCR-based methods for directing point mutations into the yeast genome such that the final product contains no other disruptions. In the first method, site-specific genomic (SSG) mutagenesis, a specific point mutation is targeted into the genome. In the second method, random domain-localized (RDL) mutagenesis, a mutation is introduced at random within a specific domain of a gene. Both methods require two sequential transformations, the first transformation integrates the URA3 marker into the targeted locus, and the second transformation replaces URA3 with a PCR fragment containing one or a few mutations. This PCR fragment is synthesized using a primer containing a mutation (SSG mutagenesis) or is synthesized by error-prone PCR (RDL mutagenesis). In SSG mutagenesis, mutations that are proximal to the URA3 site are incorporated at higher frequencies than distal mutations, however mutations can be introduced efficiently at distances of at least 500 bp from the URA3 insertion. In RDL mutagenesis, to ensure that incorporation of mutations occurs at approximately equal frequencies throughout the targeted region, this region is deleted at the same time URA3 is integrated. CONCLUSION: SSG and RDL mutagenesis allow point mutations to be easily and efficiently incorporated into the yeast genome without disrupting the native locus

    The effect of foot type on lower extremity muscle activity and center of pressure

    The purpose of this study was to examine how foot type and a foot orthotic device (FOD) affect electromyography (EMG) and kinetic data (COP). Thirty-nine individuals (20 with normal and 19 with pronated feet) were tested in overground walking in a pretestposttest design. Results of statistical analyses show no significant differences with regards to onset and duration of the tibialis anterior, peroneus longus, vastus medialis (VMO), and gluteus medius muscles. These results suggest there is no difference in onset and duration, between normal and pronated feet. There was a trend specifically with VMO duration, suggesting a possible shorter duration time with FOD intervention. The COP examination indicated that the normal group had significantly greater COP excursion during the first 50% of stance phase. Also, a similar trend in greater normal foot COP excursion existed between the groups for the entire stance phase

    AVIRIS spectra correlated with the chlorophyll concentration of a forest canopy

    Imaging spectrometers have many potential applications in the environmental sciences. One of the more promising applications is that of estimating the biochemical concentrations of key foliar biochemicals in forest canopies. These estimates are based on spectroscopic theory developed in agriculture and could be used to provide the spatial inputs necessary for the modeling of forest ecosystem dynamics and productivity. Several foliar biochemicals are currently under investigation ranging from those with primary absorption features in visible to middle infrared wavelengths (e.g., water, chlorophyll) to those with secondary to tertiary absorption features in this part of the spectrum (e.g., nitrogen, lignin). The foliar chemical of interest in this paper is chlorophyll; this is a photoreceptor and catalyst for the conversion of sunlight into chemical energy and as such plays a vital role in the photochemical synthesis of carbohydrates in plants. The aim of the research reported here was to determine if the chlorophyll concentration of a forest canopy could be correlated with the reflectance spectra recorded by the Airborne Visible/Infrared Imaging Spectrometer (AVIRIS)

    Different sets of QTLs influence fitness variation in yeast

    We have carried out a combination of in-lab-evolution (ILE) and congenic crosses to identify the gene sets that contribute to the ability of yeast cells to survive under alkali stress.Each selected line acquired a different set of mutations, all resulting in the same phenotype. We identified a total of 15 genes in ILE and 17 candidates in the congenic approach, and studied their individual contribution to the phenotype.The total additive effect of the QTLs was much larger than the difference between the ancestor and the evolved strains, suggesting epistatic interactions between the QTLs.None of the genes identified encode structural components of the pH machinery. Instead, most encode regulatory functions, such as ubiquitin ligases, chromatin remodelers, GPI anchoring and copper/iron sensing transcription factors

    EVALUATING EXTENSIVE SHEEP FARMING SYSTEMS

    Data from each of 5 commercial, extensive sheep farms in Cumbria, UK were used as parameters in a linear program (LP) representing labour and grazing management in such farming systems. The LP maximised ewe enterprise gross margin subject to constraints dictated by the labour availability and land types on each farm. Under the assumptions used, labour availability and price restricted ewe numbers well below those observed in practice on 2 farms i.e. land resources were adequate for the farming system practiced. On two other farms stocking levels and hence returns were limited by the availability of forage and hence feed input prices relative to output. On one farm, greater grassland productivity was the key determinant of system performance.     Impacts of labour on interactions between economics and animal welfare in extensive sheep farms

    This study quantified interactions between animal welfare and farm profitability in British extensive sheep farming systems. Qualitative welfare assessment methodology was used to assess welfare from the animal's perspective in 20 commercial extensive sheep farms and to estimate labour demand for welfare, based on the assessed welfare scores using data collected from farm inventories. The estimated labour demand was then used as a coefficient in a linear program based model to establish the gross margin maximising farm management strategy for given farm situations, subject to constraints that reflected current resource limitations including labour supply. Regression analysis showed a significant relationship between the qualitative welfare assessment scores and labour supply on the inventoried farms but there was no significant relationship between current gross margin and assessed welfare scores. However, to meet the labour demand of the best welfare score, a reduction in flock size and in the average maximum farm gross margin was often required.
