    Arabinoxyloglucan Oligosaccharides May Contribute to the Antiadhesive Properties of Porcine Urine after Cranberry Consumption

    © 2019 As part of our continuing investigation for interesting biological activities of native medicinal plants, thirty-nine plants, obtained from diverse areas in Saudi Arabia and Yemen, were screened for insecticidal activity against yellow fever mosquito Aedes aegypti (L.). Out of the 57 organic extracts, Saussurea lappa, Ocimum tenuiflorum, Taraxacum officinale, Nigella sativa, and Hyssopus officinalis exhibited over 80% mortality against adult female Ae. aegypti at 5 μg/mosquito. In the larvicidal bioassay, the petroleum ether extract of Aloe perryi flowers showed 100% mortality at 31.25 ppm against 1st instar Ae. aegypti larvae. The ethanol extract of Saussurea lappa roots was the second most active displaying 100% mortality at 125 and 62.5 ppm. Polar active extracts were processed using LC-MS/MS to identify bioactive compounds. The apolar A. perryi flower extract was analyzed by headspace SPME-GC/MS analysis. Careful examination of the mass spectra and detailed interpretation of the fragmentation pattern allowed the identification of various biologically active secondary metabolites. Some compounds such as caffeic and quinic acid and their glycosides were detected in most of the analyzed fractions. Additionally, luteolin, luteolin glucoside, luteolin glucuronide and diglucuronide were also identified as bioactive compounds in several HPLC fractions. Scientific Reports. 2020;10(1): 4239.Caenorhabditis elegans presents functioning, biologically relevant phenotypes and is frequently used as a bioindicator of toxicity. However, most C. elegans in vivo effect-assessment methods are laborious and time consuming. Therefore, we developed a novel method to measure the oxygen consumption rate of C. elegans as a sublethal endpoint of toxicity. This protocol was tested by exposing 50 larval stage one C. elegans individuals for 48h (at 20 degrees C) to different concentrations of two toxicants i.e. benzylcetyldimethylammonium chloride (BAC-C16) and cadmium (Cd). Following exposures, the oxygen consumption rate of the C. elegans individuals were measured using the high-throughput functionality of the Seahorse XF(e)96 Extracellular Flux Analyzer. Dose-response curves for BAC-C16 (R-2=0.93; P=0.001) and Cd (R-2=0.98; P=0.001) were created. Furthermore, a strong, positive correlation was evidenced between C. elegans oxygen consumption rate and a commonly used, ecologically relevant endpoint of toxicity (growth inhibition) for BAC-C16 (R-2=0.93; P=0.0001) and Cd (R-2=0.91; P=0.0001). The data presented in this study show that C. elegans oxygen consumption rate can be used as a promising functional measurement of toxicity

    A prospective evaluation of the feasibility and utility of additional tools to obtain information from recently diagnosed HIV infected patients

    QUESTION UNDER STUDY: Can additional information be obtained from recently HIV diagnosed individuals? METHODS: A 1-year prospective Swiss study, including all newly diagnosed HIV-infected patients. Information on circumstances of HIV infection was collected through physician- and patient questionnaires and patient interviews. Information on timing of infection was linked with an HIV-antibody avidity assay. RESULTS: Of 710 newly HIV diagnosed patients, 543 (76%) physician questionnaires (PhyQ) and 145 (20%) patient questionnaires (PaQ) were returned. PhyQ required fewer reminders (57% vs 28% spontaneous return). Patients whose doctors had returned the PhyQ were comparable to total population group. In contrast, a strong bias towards well educated recently infected Swiss men having sex with men (MSM) was seen in patients returning PaQ or agreeing to an interview. 83% of patients claimed that they knew the infection source and 85% infection place. Unprotected sexual contact was the most frequently cited infection source (92%; n = 404). Men mainly claimed occasional (43%) and women steady (61%) partners as the most likely source of HIVinfection. Serum for timing of infection was available in 98% of patients. Recent infections (RI) were highest in MSM (51%) and intravenous drug users (IDU, 54%). Compared to women, heterosexual men were more than twice as likely to be diagnosed with a RI. CONCLUSION: Relevant additional information on circumstances of HIV infection in newly diagnosed patients can easily be collected from treating physicians. Collecting information from patients is not a feasible option, with the exception of qualitative interviews in a selected group of patients

    Antimicrobial and Antiinsectan Phenolic Metabolites of Dalea searlsiae

    Continued interest in the chemistry of Dalea spp. led to investigation of Dalea searlsiae, a plant native to areas of the western United States. Methanol extractions of D. searlsiae roots and subsequent chromatographic fractionation afforded the new prenylated and geranylated flavanones malheurans A–D (1–4) and known flavanones (5 and 6). Known rotenoids (7 and 8) and isoflavones (9 and 10) were isolated from aerial portions. Structure determination of pure compounds was accomplished primarily by extensive 1D- and 2D-NMR spectroscopy. The absolute configurations of compounds 1–5, 7, and 8 were assigned using electronic circular dichroism spectroscopy. Antimicrobial bioassays revealed significant activity concentrated in the plant roots. Compounds 1–6 exhibited MICs of 2–8 μg/mL against Streptococcus mutans, Bacillus cereus, and oxacillin-sensitive and -resistant Staphylococcus aureus. Aerial metabolites 7–10 were inactive against these organisms, but the presence of 7 and 8 prompted investigation of the antiinsectan activity of D. searlsiae metabolites toward the major crop pest Spodoptera frugiperda (fall armyworm). While compounds 1–10 all caused significant reductions in larval growth rates, associated mortality (33–66%) was highest with flavanone 4 and rotenoids 7 and 8. These findings suggest a differential allocation of antimicrobial and antiinsectan plant resources to root and aerial portions of the plant, respectively

    Antimalarials and Phytotoxins from Botryosphaeria dothidea Identified from a Seed of Diseased Torreya taxifolia

    The metabolic pathways in the apicoplast organelle of Plasmodium parasites are similar to those in plastids in plant cells and are suitable targets for malaria drug discovery. Some phytotoxins released by plant pathogenic fungi have been known to target metabolic pathways of the plastid; thus, they may also serve as potential antimalarial drug leads. An EtOAc extract of the broth of the endophyte Botryosphaeria dothidea isolated from a seed collected from a Torreya taxifolia plant with disease symptoms, showed in vitro antimalarial and phytotoxic activities. Bioactivity-guided fractionation of the extract afforded a mixture of two known isomeric phytotoxins, FRT-A and flavipucine (or their enantiomers, sapinopyridione and (-)-flavipucine), and two new unstable γ-lactam alkaloids dothilactaenes A and B. The isomeric mixture of phytotoxins displayed strong phytotoxicity against both a dicot and a monocot and moderate cytotoxicity against a panel of cell lines. Dothilactaene A showed no activity. Dothilactaene B was isolated from the active fraction, which showed moderate in vitro antiplasmodial activity with high selectivity index. In spite of this activity, its instability and various other biological activities shown by related compounds would preclude it from being a viable antimalarial lead

    Factors affecting diet, habitat selection and breeding success of the African Crowned Eagle Stephanoaetus coronatus in a fragmented landscape

    This study aimed to identify variables that affect habitat selection and nesting success of the African Crowned Eagle Stephanoaetus coronatus, the largest forest raptor, in north-eastern South Africa. A preference for nesting in the Northern Mistbelt Forest vegetation type was established and 82% of all nests were located in indigenous trees. Nest abandonment was less common when distances to the nearest neighbour were greater. The diet of this species was investigated by examination of prey remains beneath nests and verified by comparison with museum specimens. In total, 156 remains were found, representing a minimum of 75 prey individuals. The diet of African Crowned Eagles constituted almost entirely mammals (99%), which were predominantly antelopes (61%) and monkeys (25%). It was also found that the proportion of primates in the diet correlates with latitude: populations in equatorial latitudes have a higher proportion of primates in their diets, whereas further south antelopes are a much more common diet component

    The fluorescence shown by extracts of the heartwood of Pterocarpus marsupium is attributed to salts of the new compound 1, whose structure was elaborated using detailed spectroscopic/ spectrometric studies. The plant material also contains the nonfluorescent compounds 2 and 3. The absolute configuration of 1 was determined by experimental and theoretically calculated electronic CD spectra, while that of 3 was deduced from ECD comparison with reported results in the α-hydroxydihydrochalcone series

    In this contribution, an overview of the distinct waysin which the interplay between knowledge, values, and beliefs tookshape in the South African context since 1948 is offered. This is framedagainst the background of the paleontological significance of SouthAfrica and an appreciation of indigenous knowledge systems, but alsoof the ideological distortion of knowledge and education during theapartheid era through the legacy of neo-Calvinism. The overviewincludes references to discourse on human rationality (as an implicitcritique against ideology), on the use of social sciences in theologicalreflection, on the teaching of evolution in public schools, on scienceand religion, and on religion and ecology. The essay concludes witha survey of some of the major voices regarding the interface betweenreligion and science in South Africa
