    Towards Reflective AI:Needs, Challenges and Directions for Future Research

    Harnessing benefits and preventing harms of AI cannot be solved alone through technological fixes and regulation. It depends on a complex interplay between technology, societal governance, individual behaviour, organizational and societal dynamics. Enabling people to understand AI and the consequences of its use and design is a crucial element for ensuring responsible use of AI.In this report we suggest a new framework for the development and use of AI technologies in a way that harnesses the benefits and prevents the harmful effects of AI. We name it Reflective AI. The notion of Reflective AI that we propose calls for adopting a holistic approach in the research and development of AI to investigate both what people need to learn about AI systems to develop better mental models i.e. an experiential knowledge of AI, to be able to use it safely and responsibly, as well as how this can be done and supported

    Die ZelladhĂ€sion von Leukozyten/ Metastasenzellen an Endothelzellen bei EntzĂŒndungsreaktionen/ Arteriosklerose und Tumormetastasierung ist ein mehrstufiger Prozess. Im ersten Schritt kommt es zu einer Interaktion zwischen E-Selektin-Rezeptoren auf den Endothelzellen und komplexen Kohlenhydraten, den Polylaktosaminen und ihren Derivaten, den Lewis-Antigenen. Im Rahmen dieser Arbeit wurden durch transiente Transfektion fĂŒnf rekombinante Glykoproteine erzeugt (hLysII/IV-FucTIII-VII). Durch hLysII/IV-FucTVI konnte die ZelladhĂ€sion von U937 an HUVEC Zellen signifikant gehemmt werden. Fukosyltransferase VI scheint in der Lage zu sein, die Bildung von endstĂ€ndigen sLex-Antigenen an den Polylaktosaminketten von hLys zu ermöglichen. Auch durch eine Transfektion der Kolonkarzinomzelllinie SW480 und Colo 206 konnten Lysozymmutanten hergestellt werden, mit denen eine signifikante AdhĂ€sionblockade möglich ist. Man kann sich hLysII/IV-FucTVI- SW480 und- Colo206 als potentielle therapeutische Substanzen vorstellen, die zum Beispiel zur EntzĂŒndungs- oder Metastasierungshemmung eingesetzt werden.The initial step in diapedesis is the interaction between e-selectin and its ligand, the lewis antigen. In this study we examine the possibility to block this interaction to reduce metastasis and inflammatory processes. The plasmid hLysII/IV is expressed in CHO cells with active alpha1,3fucosyltransferases (III-VII). The resulting five recombinant glycoproteins are purified from cultur supernatants and static cell binding assays are performed. hLysII/IV-FucTVI was shown to bind to cells expressing e-selectin on its cell surface. It inhibited the binding of the monocytic cell line U937 to human endothelial cells activated with TNFalpha to upregulate the e-selectin expression. Alpha1,3fucosyltransferase VI seems to make possible the biosynthesis of the sLex and the hLysII/IV-FucTVI glycoprotein seems to be an interesting target to reduce inflammation and metastasis

    Validation of an open source high voltage grid model for AC load flow calculations in a delimited region

    Large shares of renewable energy production in the electricity grid make grid expansion and new technologies necessary. The unavailability of grid models to address upcoming research questions led to the development of open source grid models. Our work contributes to establish the open_eGo model for grid simulations by validating its assumptions and results for a rural region with high share of wind energy. In particular, assumptions on electrical parameters and the graph structure of the model are compared to the grid owner's model along with a validation of AC load flow results at the model boundaries. It was found that the graph structure deviates in the degree of nodes and connection characteristic. These deviations are less exterior nodes and a lower maximum degree of nodes as well as a higher number of parallel lines in the open_eGo model. The AC load flow results differ slightly in active power and significantly in reactive power, but are more reliable than an aggregation of loads and generation to the extra high voltage (EHV) nodes. Concluding, the open_eGo model has a limited usability for simulating, understanding and optimising DSO grid operation but can enhance EHV-only analysis in large area contexts

    Visualization of ligand-induced dopamine D2S and D2L receptor internalization by TIRF microscopy

    G protein-coupled receptors (GPCRs), including the dopamine receptors, represent a group of important pharmacological targets. Upon agonist binding, GPCRs frequently undergo internalization, a process that is known to attenuate functional responses upon prolonged exposure to agonists. In this study, internalization was visualized by means of total internal reflection fluorescence (TIRF) microscopy at a level of discrete single events near the plasma membrane with high spatial resolution. A novel method has been developed to determine the relative extent of internalized fluorescent receptor-ligand complexes by comparative fluorescence quantification in living CHO cells. The procedure entails treatment with the reducing agent sodium borohydride, which converts cyanine-based fluorescent ligands on the membrane surface to a long-lived reduced form. Because the highly polar reducing agent is not able to pass the cell membrane, the fluorescent receptor-ligand complexes located in internalized compartments remain fluorescent under TIRF illumination. We applied the method to investigate differences of the short (D2S) and the long (D2L) isoforms of dopamine D2 receptors in their ability to undergo agonist-induced internalization

    Near-UV circular dichroism reveals structural transitions of vimentin subunits during intermediate filament assembly

    In vitro assembly of vimentin intermediate filaments (IFs) proceeds from soluble, reconstituted tetrameric complexes to mature filaments in three distinct stages: (1) within the first seconds after initiation of assembly, tetramers laterally associate into unit-length filaments (ULFs), on average 17 nm wide; (2) for the next few minutes, ULFs grow by longitudinal annealing into short, immature filaments; (3) almost concomitant with elongation, these immature filaments begin to radially compact, yielding approximately 11-nm-wide IFs at around 15 min. The near-UV CD signal of soluble tetramers exhibits two main peaks at 285 and 278 nm, which do not change during ULF formation. In contrast, the CD signal of mature IFs exhibits two major changes: (1) the 278-nm band, denoting the transition of the tyrosines from the ground state to the first vibrational mode of the excited state, is lost; (2) a red-shifted band appears at 291 nm, indicating the emergence of a new electronic species. These changes take place independently and at different time scales. The 278-nm signal disappears within the first minute of assembly, compatible with increased rigidity of the tyrosines during elongation of the ULFs. The rise of the 291-nm band has a lifetime of approximately 13 min and denotes the generation of phenolates by deprotonation of the tyrosines' hydroxyl group after they relocalize into a negatively charged environment. The appearance of such tyrosine-binding "pockets" in the assembling filaments highlights an essential part of the molecular rearrangements characterizing the later stages of the assembly process, including the radial compaction
