    Protective Effects of Calcium Against Chronic Waterborne Cadmium Exposure to Juvenile Rainbow Trout

    Juvenile rainbow trout (Oncorhynchus mykiss [Walbaum]) on 1% daily ration were exposed to 0 (control) or 2 Ī¼g of cadmium as Cd(NO3)2Ā·4H2O per liter added to four different calcium (Ca) concentrations: 260 (background), 470 (low), 770 (medium), or 1200 (high) Ī¼M of Ca added as Cd(NO3)2Ā·4H2O in synthetic soft water for 30 d. Mortality was highest (;80%) in the background 1 Cd treatment. Approximately 40% mortality was observed in the low 1 Cd exposure; mortality was 10% or less for all other treatments. No growth effects were seen for any of the exposures. Kidneys accumulated the greatest concentration of Cd during the 30 d, followed by gills and livers. Accumulation of Cd in gills, kidney, and liver decreased at higher water Ca concentrations. No differences in whole-body or plasma Ca concentrations were found. Swimming performance was impaired in the low + Cd-exposed fish. Influx of Ca2+ into whole bodies decreased as water Ca concentrations increased; influx of Ca2+ into background + Cdā€“treated fish was significantly reduced compared to that in control fish. Experiments that measured uptake of new Cd into gills showed that the affinity of gills for Cd (KCd-gill) and the number of binding sites for Cd decreased as water Ca concentrations increased. Acute accumulation of new Cd into gills and number of gill Cd-binding sites increased with chronic Cd exposure, whereas the affinity of gills for Cd decreased with chronic Cd exposure. Longer-term gill binding (72 h) showed reduced uptake of new Cd at higher water Ca levels and increased uptake with chronic Cd exposure. Complications were found in applying the biotic ligand model to fish that were chronically exposed to Cd because of discrepancies in the maximum number of gill Cd-binding sites among different studies

    Learning through virtual reality: Virtual Bethel case study

    Focusing on the challenges of teaching virtual reality creation and preservation, our paper will present a case study involving the virtual recreation of the Bethel AME Church sanctuary. We were particularly interested in studentsā€™ skills, the technology, and costs associated with teaching and learning virtual reality, and how these factors influence overall student learning experiences. Two courses are explored: 3D Production and Digital Preservation. We have learned that teaching and learning in this space is technology and skill intensive. By assessing the skills and technology needed as well as the costs and student experiences, we are better able to communicate the needs of these projects to potential funders and collaborators. Weā€™ve determined that without external funding, we are currently at capacity and will need funding for additional collaborative projects. The level of technical ability of the students influenced their level of satisfaction as well as their capacity to learn

    Remembering Together: Native Boarding School Stories on Display

    Recent scholarship on Native American boarding schools has focused on drawing out the complexities of boarding school history and emphasizing the plurality of experiences of students. This thesis examines how Native American boarding school stories have been displayed using two current museum exhibits: ā€œAway from Home: American Indian Boarding School Storiesā€ at the Heard Museum, and the Phoenix Indian School Visitors Center, a small gallery in one of the remaining school buildings. For this analysis I interviewed key players in both current exhibits and did close readings of the exhibits themselves, in conjunction with archival research about two model schoolhouse exhibits at worldā€™s fairs. Each of these exhibits, old and new, was engaged with the anthropology and museum theory of the time. The two current exhibits offer updated scholarship and multivocal narratives but designed for and aimed at audiences of very different backgrounds

    <p>Abstract</p> <p>Background</p> <p>Activated microglia are a feature of the host response to neurodegeneration in Parkinson's disease (PD) and are thought to contribute to disease progression. Recent evidence suggests that extracellular Ī±-synuclein (eSNCA) may play an important role in the pathogenesis of PD and that this may be mediated by a microglial response.</p> <p>Methods</p> <p>We wished to discover whether the host response to eSNCA would be sufficient to induce significant cytokine production. <it>In vitro </it>cultured BV-2 microglia were used to determine the basic inflammatory response to eSNCA. <it>In vivo</it>, 8-week old Biozzi mice were subjected to a single intranigral injection of either 3 Ī¼g SNCA, lipopolysaccharide (LPS) or serum protein (BSA) and allowed to recover for 24 hours. A second cohort of animals were peripherally challenged with LPS (0.5 mg/kg) 6 hours prior to tissue collection. Inflammation was studied by quantitative real-time PCR for a number of pro-inflammatory genes and immunohistochemistry for microglial activation, endothelial activation and cell death.</p> <p>Results</p> <p><it>In vitro </it>data showed a robust microglial response to SNCA, including a positive NFÄøB response and the production of pro-inflammatory cytokines. Direct injection of SNCA into the substantia nigra resulted in the upregulation of mRNA expression of proinflammatory cytokines, the expression of endothelial markers of inflammation and microglial activation. However, these results were significantly different to those obtained after direct injection of LPS. By contrast, when the animals were injected intracerebrally with SNCA and subsequently challenged with systemic LPS, the level of production of IL-1Ī² in the substantia nigra became comparable to that induced by the direct injection of LPS into the brain. The injection of albumin into the nigra with a peripheral LPS challenge did not provoke the production of a significant inflammatory response. Direct injection of LPS into the substantia nigra also induces cell death in a more robust manner than direct injection of either SNCA or BSA.</p> <p>Conclusion</p> <p>These results suggest that the presence of eSNCA protein 'primes' microglia, making them susceptible to environmental proinflammatory challenge. For this reason, we hypothesise that where 'inflammation' contributes to the disease progression in PD, it does so in a punctuate manner (on-off) as a result of systemic events.</p

    Mapbook of Syntactic Variation in American English: Survey Results, 2015ā€“2019

    This work presents the results of a series of acceptability judgment surveys conducted by the Yale Grammatical Diversity Project (YGDP) between 2015 and 2019. It contains over 200 maps of some 194 sentences, covering a wide range of syntactic constructions, including dative presentatives, personal datives, extended benefactives, the have yet to construction, the done my homework construction, wicked, hella, the so donā€™t I construction, the alls construction, the come with construction, fixinā€™ to, the needs washed construction, non-polarity anymore (aka ā€œpositive anymoreā€), and many others. For each sentence, we also provide some basic demographic information, such as how the sentence judgments varied by age, race, gender, education, and urban/rural classifications. We describe the goals of these surveys, as well as how they were designed, administered, processed, and mapped, along with a brief introduction to the history of the YGDP. In addition to providing a detailed look at syntactic variation in U.S. English to an extent that has previously been unavailable, we hope that this work will be useful in linguistics classrooms at all levels, and will provide the springboard for further, more detailed studies of the individual constructions, geographic regions, and linguistic and social factors connected to syntactic variation in U.S. English

    Chimpanzees demonstrate individual differences in social information use

    Studies of transmission biases in social learning have greatly informed our understanding of how behaviour patterns may diffuse through animal populations, yet within-species inter-individual variation in social information use has received little attention and remains poorly understood. We have addressed this question by examining individual performances across multiple experiments with the same population of primates. We compiled a dataset spanning 16 social learning studies (26 experimental conditions) carried out at the same study site over a 12-year period, incorporating a total of 167 chimpanzees. We applied a binary scoring system to code each participantā€™s performance in each study according to whether they demonstrated evidence of using social information from conspecifics to solve the experimental task or not (Social Information Scoreā€”ā€˜SISā€™). Bayesian binomial mixed effects models were then used to estimate the extent to which individual differences influenced SIS, together with any effects of sex, rearing history, age, prior involvement in research and task type on SIS. An estimate of repeatability found that approximately half of the variance in SIS was accounted for by individual identity, indicating that individual differences play a critical role in the social learning behaviour of chimpanzees. According to the model that best fit the data, females were, depending on their rearing history, 15ā€“24% more likely to use social information to solve experimental tasks than males. However, there was no strong evidence of an effect of age or research experience, and pedigree records indicated that SIS was not a strongly heritable trait. Our study offers a novel, transferable method for the study of individual differences in social learning

    Local-Circuit Phenotypes of Layer 5 Neurons in Motor-Frontal Cortex of YFP-H Mice

    Layer 5 pyramidal neurons comprise an important but heterogeneous group of cortical projection neurons. In motor-frontal cortex, these neurons are centrally involved in the cortical control of movement. Recent studies indicate that local excitatory networks in mouse motor-frontal cortex are dominated by descending pathways from layer 2/3 to 5. However, those pathways were identified in experiments involving unlabeled neurons in wild type mice. Here, to explore the possibility of class-specific connectivity in this descending pathway, we mapped the local sources of excitatory synaptic input to a genetically labeled population of cortical neurons: YFP-positive layer 5 neurons of YFP-H mice. We found, first, that in motor cortex, YFP-positive neurons were distributed in a double blade, consistent with the idea of layer 5B having greater thickness in frontal neocortex. Second, whereas unlabeled neurons in upper layer 5 received their strongest inputs from layer 2, YFP-positive neurons in the upper blade received prominent layer 3 inputs. Third, YFP-positive neurons exhibited distinct electrophysiological properties, including low spike frequency adaptation, as reported previously. Our results with this genetically labeled neuronal population indicate the presence of distinct local-circuit phenotypes among layer 5 pyramidal neurons in mouse motor-frontal cortex, and present a paradigm for investigating local circuit organization in other genetically labeled populations of cortical neurons

    Complexities of Digital Preservation in a Virtual Reality Environment, the Case of Virtual Bethel

    The complexity of preserving virtual reality environments combines the challenges of preserving singular digital objects, the relationships among those objects, and the processes involved in creating those relationships. A case study involving the preservation of the Virtual Bethel environment is presented. This case is active and ongoing. The paper provides a brief history of the Bethel AME Church of Indianapolis and its importance, then describes the unique preservation challenges of the Virtual Bethel project, and finally provides guidance and preservation recommendations for Virtual Bethel, using the National Digital Stewardship Alliance Levels of Preservation. Discussion of limitations of the guidance and recommendations follow
