    On the applicability of the moving line source theory to thermal response test under groundwater flow: considerations from real case studies

    The classical methodology to perform and analyze thermal response test (TRT) is unsuccessful when advection contributes to heat transfer in the ground, due to the presence of a groundwater flow. In this study, the applicability, the advantages, and the limitations of the moving line source model to interpret TRT data are discussed. Two real TRT case studies from the Italian Alpine area are reported and analyzed, with both the standard infinite line source approach and the moving line source one. It is shown that the inverse heat transfer problem is ill-posed, leading to multiple solutions. However, besides minimization of the error between measurements and modeling, physical considerations help to discriminate among solutions the most plausible ones. In this regard, the MLS approach proves to be effective in the advection-dominated case. The original time criterion proposed here to disregard initial data from the fitting, based on a resistance–capacitance model of the borehole embedded in a groundwater flow, is validated in terms of convergence of the solution. In turn, in the case when advection and conduction are competitive, the MLS approach results more sensitive to ground thermal conductivity than to Darcy velocity. Although in this case a limited impact of the uncertainty in the groundwater velocity on the boreholes sizing is expected, future studies should focus on the development of a successful TRT methodology for this condition

    El objetivo de este estudio es conocer el estado de la gestión con enfoque de responsabilidad social empresarial (RSE) en las empresas hoteleras de la región Piura en el año 2015, utilizando como herramienta de evaluación el cuestionario de indicadores Ethos- Perú 2021. En la actualidad, en Perú y Latinoamérica, son pocos los hoteles que implementan la responsabilidad social en sus modelos de gestión, por lo que el presente estudio es un medio que permitirá alcanzar un fin concreto, como es la aplicación de la responsabilidad social para la sostenibilidad de las empresas del referido sector en la región. Esta investigación cuantitativa tiene un diseño no experimental con alcance transversal basado en la creación de vínculos entre las variables de estudio, la población constituida por los gerentes de cinco hoteles de la región Piura cuya identidad se mantuvo en el anonimato durante de la investigación. Para el estudio, se aplicaron las preguntas de profundidad mediante el cuestionario de indicadores de RSE de Ethos-Perú 2021, según el modelo virtual en forma auto evaluativa respondida por los informantes, y se contrastó la información con las preguntas binarias. Los resultados obtenidos y presentados en gráficas han demostrado que el cumplimiento de los indicadores de RSE Ethos-Perú 2021, según las conclusiones de las respuestas binarias, tienen una tendencia cercana a la media de cumplimiento y, según las preguntas de profundidad, en una etapa dos (02), para las cinco empresas del sector hoteleroThe aim of this study is to determine the status of management with a focus on corporate social responsibility (CSR) in hotel companies in the Piura region in 2015, using the questionnaire Ethos-Peru 2021 indicators, as an evaluation tool.     Genetic susceptibility to aspergillosis in allogeneic stem-cell transplantation

    Invasive aspergillosis (IA) is a major threat to positive outcomes for allogeneic stem-cell transplantation (allo-SCT) patients. Despite presenting similar degrees of immunosuppression, not all individuals at-risk ultimately develop infection. Therefore, the traditional view of neutropenia as a key risk factor for aspergillosis needs to be accommodated within new conceptual advances on host immunity and its relationship to infection. Polymorphisms in innate immune genes, such as those encoding TLRs, cytokines and cytokine receptors, have recently been associated with susceptibility to IA in allo-SCT recipients.     Primary Postnatal Dorsal Root Ganglion Culture from Conventionally Slaughtered Calves

    Neurological disorders in ruminants have an important impact on veterinary health, but very few host-specific in vitro models have been established to study diseases affecting the nervous system. Here we describe a primary neuronal dorsal root ganglia (DRG) culture derived from calves after being conventionally slaughtered for food consumption. The study focuses on the in vitro characterization of bovine DRG cell populations by immunofluorescence analysis. The effects of various growth factors on neuron viability, neurite outgrowth and arborisation were evaluated by morphological analysis. Bovine DRG neurons are able to survive for more than 4 weeks in culture. GF supplementation is not required for neuronal survival and neurite outgrowth. However, exogenously added growth factors promote neurite outgrowth. DRG cultures from regularly slaughtered calves represent a promising and sustainable host specific model for the investigation of pain and neurological diseases in bovines

    Identifying low density lipoprotein cholesterol associated variants in the Annexin A2 (ANXA2) gene.

    BACKGROUND AND AIMS: Annexin-A2 (AnxA2) is an endogenous inhibitor of proprotein convertase subtilisin/kexin type-9 (PCSK9). The repeat-one (R1) domain of AnxA2 binds to PCSK9, blocking its ability to promote degradation of low-density lipoprotein cholesterol-receptors (LDL-R) and thereby regulate low-density lipoprotein cholesterol (LDL-C) levels. Here we identify variants in ANXA2 influencing LDL-C levels and we determine the molecular mechanisms of their effects. RESULTS: The ANXA2 single nucleotide polymorphism (SNP) genotype-phenotype association was examined using the Second-Northwick-Park Heart Study (NPHSII) (n∼2700) and the UCL-LSHTM-Edinburgh-Bristol (UCLEB) consortium (n∼14,600). The ANXA2-R1 domain coding-SNP rs17845226 (V98L) associated with LDL-C, homozygotes for the minor allele having ≈18.8% higher levels of LDL-C (p = 0.004), and higher risk of coronary heart disease (CHD) (p = 0.04). The SNP is in modest linkage disequilibrium (r(2) > 0.5) with two intergenic SNPs, rs17191344 and rs11633032. Both SNPs showed allele-specific protein binding, and the minor alleles caused significant reduction in reporter gene expression (≈18%, p < 0.001). In the expression quantitative trait loci (eQTL) study, minor allele homozygotes have significantly lower levels of ANXA2-mRNA expression (p = 1.36 × 10(-05)). CONCLUSIONS: Both rs11633032 and rs17191344 SNPs are functional variants, where the minor alleles create repressor-binding protein sites for transcription factors that contribute to reduced ANXA2 gene expression. Lower AnxA2 levels could increase plasma levels of PCSK9 and thus increase LDL-C levels and risk of CHD. This supports, for the first time in humans, previous observations in mouse models that changes in the levels of AnxA2 directly influence plasma LDL-C levels, and thus implicate this protein as a potential therapeutic target for LDL-C lowering

    Genetically-Determined Hyperfunction of the S100B/RAGE Axis Is a Risk Factor for Aspergillosis in Stem Cell Transplant Recipients

    Invasive aspergillosis (IA) is a major threat to the successful outcome of hematopoietic stem cell transplantation (HSCT), although individual risk varies considerably. Recent evidence has established a pivotal role for a danger sensing mechanism implicating the S100B/receptor for advanced glycation end products (RAGE) axis in antifungal immunity. The association of selected genetic variants in the S100B/RAGE axis with susceptibility to IA was investigated in 223 consecutive patients undergoing HSCT. Furthermore, studies addressing the functional consequences of these variants were performed. Susceptibility to IA was significantly associated with two distinct polymorphisms in RAGE (-374T/A) and S100B (+427C/T) genes, the relative contribution of each depended on their presence in both transplantation counterparts [patient SNPRAGE, adjusted hazard ratio (HR), 1.97; P = 0.042 and donor SNPRAGE, HR, 2.03; P = 0.047] or in donors (SNPS100B, HR, 3.15; P = 7.8e-4) only, respectively. Functional assays demonstrated a gain-of-function phenotype of both variants, as shown by the enhanced expression of inflammatory cytokines in RAGE polymorphic cells and increased S100B secretion in vitro and in vivo in the presence of the S100B polymorphism. These findings point to a relevant role of the danger sensing signaling in human antifungal immunity and highlight a possible contribution of a genetically-determined hyperfunction of the S100B/RAGE axis to susceptibility to IA in the HSCT setting

    MyD88 and STING Signaling Pathways Are Required for IRF3-Mediated IFN-β Induction in Response to Brucella abortus Infection

    Type I interferons (IFNs) are cytokines that orchestrate diverse immune responses to viral and bacterial infections. Although typically considered to be most important molecules in response to viruses, type I IFNs are also induced by most, if not all, bacterial pathogens. In this study, we addressed the role of type I IFN signaling during Brucella abortus infection, a facultative intracellular bacterial pathogen that causes abortion in domestic animals and undulant fever in humans. Herein, we have shown that B. abortus induced IFN-β in macrophages and splenocytes. Further, IFN-β induction by Brucella was mediated by IRF3 signaling pathway and activates IFN-stimulated genes via STAT1 phosphorylation. In addition, IFN-β expression induced by Brucella is independent of TLRs and TRIF signaling but MyD88-dependent, a pathway not yet described for Gram-negative bacteria. Furthermore, we have identified Brucella DNA as the major bacterial component to induce IFN-β and our study revealed that this molecule operates through a mechanism dependent on RNA polymerase III to be sensed probably by an unknown receptor via the adaptor molecule STING. Finally, we have demonstrated that IFN-αβR KO mice are more resistant to infection suggesting that type I IFN signaling is detrimental to host control of Brucella. This resistance phenotype is accompanied by increased IFN-γ and NO production by IFN-αβR KO spleen cells and reduced apoptosis

    Background: Paracoccidioides brasiliensis causes paracoccidioidomycosis, one of the most prevalent systemic mycosis in Latin America. Thus, understanding the characteristics of the protective immune response to P. brasiliensis is of interest, as it may reveal targets for disease control. The initiation of the immune response relies on the activation of pattern recognition receptors, among which are TLRs. Both TLR2 and TLR4 have been implicated in the recognition of P. brasiliensis and regulation of the immune response. However, the role of TLR9 during the infection by this fungus remains unclear.J.F.
