Physical inactivity is a public health problem and one of the greatest evils of modern society. Biggest part of the sedentary population practice physical activity sporadically. It is well established that the excess of physical exercise in or non-conditioned people causes oxidative stress and muscle damage. The present study tested the hypothesis that a single session of physical exercise can cause oxidative stress and muscle damage in sedentary rats. Additively, the antioxidant effect of the polyphenol resveratrol (RV) was evaluated, and its ability to attenuate oxidative stress and muscle damage caused by physical activity. For this, 40 sedentary adults rats (Rattus norvegicus Albinus, Wistar) were randomly subjected or not to 90 minutes of swimming, with and without treatment with RV (100mg/kg/14days): N-RV- (n=10) group maintained at rest and not treated with RV, N-RV+ (n=10) group maintained at rest and treated with RV, N+RV- (n=10) group subjected to physical exercise of swimming not treated with RV and N+RV+ (n=10) group submitted to physical exercise of swimming and treated with RV. In sedentary rats, physical activity of swimming promoted oxidative stress (observed by increased lipid peroxidation and decreased total antioxidant capacity of plasma) and significant increase in plasma activity of creatine kinase (CK) and lactate dehydrogenase (LDH). The RV treatment decreased lipid peroxidation and activity of the markers of muscle injury (CK and LDH) in sedentary rats submitted to swimming. This is one of the first evidence that a single session of physical exercise can cause oxidative stress in sedentary individuals and that the RV can be an alternative to alleviate muscle injury caused by this stres

Abstract Among the uremic toxins proven to affect the neutrophil function in humans with chronic kidney disease (CKD), guanidine compounds stand out. To achieve a clearer understanding of the mechanisms that affect the immunity of uremic patients, the hypothesis that guanidine acetic acid (GAA) contributes to the inhibition of oxidative metabolism and an increase in neutrophil apoptosis in healthy dogs was investigated in vitro. To this end, neutrophils isolated from ten healthy dogs were incubated in pure RPMI 1640 (control) and enriched with 5 mg/L of GAA. Capillary flow cytometry was used to quantify superoxide production in neutrophils with the probe (hydroethidine), in the presence and absence of phorbol-12-myristate-13-acetate (PMA), in order to assess oxidative metabolism. Apoptotic indices were quantified using the Annexin V-PE system, with and without the inductive effect of camptothecin. Neutrophils isolated and incubated in a GAA-enriched medium produced smaller amounts of superoxide (p<0.001) when activated with PMA, however, this inhibition of oxidative metabolism occurred without significantly altering their viability or rate of apoptosis. Thus, the results show guanidine compounds contribute to immunosuppression in dogs with CKD

BACKGROUND:Different equations have been used to describe and understand the growth kinetics of undisturbed malignant solid tumors. The aim of this paper is to propose a new formulation of the Gompertz equation in terms of different parameters of a malignant tumor: the intrinsic growth rate, the deceleration factor, the apoptosis rate, the number of cells corresponding to the tumor latency time, and the fractal dimensions of the tumor and its contour. METHODS:Furthermore, different formulations of the Gompertz equation are used to fit experimental data of the Ehrlich and fibrosarcoma Sa-37 tumors that grow in male BALB/c/Cenp mice. The parameters of each equation are obtained from these fittings. RESULTS:The new formulation of the Gompertz equation reveals that the initial number of cancerous cells in the conventional Gompertz equation is not a constant but a variable that depends nonlinearly on time and the tumor deceleration factor. In turn, this deceleration factor depends on the apoptosis rate of tumor cells and the fractal dimensions of the tumor and its irregular contour. CONCLUSIONS:It is concluded that this new formulation has two parameters that are directly estimated from the experiment, describes well the growth kinetics of unperturbed Ehrlich and fibrosarcoma Sa-37 tumors, and confirms the fractal origin of the Gompertz formulation and the fractal property of tumors

This study aimed to evaluate the influence of apical periodontitis (AP) and/or periodontal disease (PD) on serum interleukin-17 (IL-17) levels in a rat model of diabetes mellitus (DM).Eighty male Wistar rats were divided into eight groups of ten animals each: normoglycemic, AP, PD, AP+PD, DM, DM+AP, DM+PD, and DM+AP+PD. DM was induced using streptozotocin, AP by dental pulp exposure to the oral environment, and PD by periodontal ligature. The animals were sacrificed after 30 days, and venous blood samples were collected via cardiac puncture to determine the serum IL-17 and neutrophil levels. The maxillae were dissected and processed for radiographic analysis. The periapical lesion areas were quantified in pixels. The total assessed values were tabulated according to each experimental group and were statistically analyzed using Spearman's correlation and Kruskal-Wallis test (p < 0.05).A significant difference in the serum IL-17 levels was observed between the groups without oral infections and the groups with AP+PD-associated lesions, regardless of the presence of DM (p < 0.05). Diabetes increased the neutrophil levels, regardless of the presence of oral infection. However, a combination of two oral infections increased the neutrophil levels in DM rats (p < 0.05). The level of bone resorption lesions was greater in DM rats than in normoglycemic rats (p < 0.05).The combination of AP and PD increased the serum IL-17 levels in DM and normoglycemic rats and increased the neutrophil levels in DM rats. Diabetes increased the neutrophil levels and bone resorption in rats.AP is capable of potentiating systemic inflammatory changes when associated with PD, and increases in blood glucose can accelerate the pathogenesis of oral infections.Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP

Understanding untreated tumour growth kinetics and its intrinsic behaviour is interesting and intriguing. The aim of this study is to propose an approximate analytical expression that allows us to simulate changes in surface charge density at the cancer-surrounding healthy tissue interface during the untreated solid tumour growth. For this, the Gompertz and Poisson equations are used. Simulations reveal that the unperturbed solid tumour growth is closely related to changes in the surface charge density over time between the tumour and the surrounding healthy tissue. Furthermore, the unperturbed solid tumour growth is governed by temporal changes in this surface charge density. It is concluded that results corroborate the correspondence between the electrical and physiological parameters in the untreated cancer, which may have an essential role in its growth, progression, metastasis and protection against immune system attack and anti-cancer therapies. In addition, the knowledge of surface charge density changes at the cancer-surrounding healthy tissue interface may be relevant when redesigning the molecules in chemotherapy and immunotherapy taking into account their polarities. This can also be true in the design of completely novel therapies
