    The rural context for health care systems has been experiencing substantial stress in recent times. The population, from which providers must obtain patients, is no longer growing as it had during the 1970s, but rather is likely to be either stable or decline where agriculture or energy are the primary economic sectors (Agresta, 1985, Johnson, 1989). Accompanying depopulation, in farming and energy producing communities an economic stagnation or depression has also been occurring. Both the loss of population base and restricted economies serve to limit patient use of clinics and hospitals, reducing their revenues and contributing to a further limitation of service

    The changing economic character of American agriculture is producing more than just surplus crops, it is also producing measurable increases in stress. This stress is a consequence of the faltering economic conditions in rural America (Cogner et. al,, 1986; Farmer, 1986), which has significant implications for the well-being of our rural population. Long term depression, increased suicide rates, family strain and violence are all results of this economic change (Farmer, 1986). Faraer (1986), characterized the rural sector as financially, emotionally, and socially troubled. These troubles are directly linked to the economic crisis rural America is experiencing. Social scientists and mental health professionals have expressed concern about the effects of the rural crisis on the farm population. Excessive and widespread financial difficulties have promoted increased concern which has led to psychological stress. The concept of stress is a generic term usually associated with mental tension or strain. For all its popularity, the term has only recently found its way into the medical vocabulary. One of the primary reasons for this omission has been the lack of an adequate or standardized definition of the concept (Wallis,1983). Empirical indicators of stress have ranged from migraine headaches and stomach cramps, to insomnia, lethargy, and confusion. Consequently, few studies operationalize stress the same way

    MOTIF-EM: an automated computational tool for identifying conserved regions in CryoEM structures

    We present a new, first-of-its-kind, fully automated computational tool MOTIF-EM for identifying regions or domains or motifs in cryoEM maps of large macromolecular assemblies (such as chaperonins, viruses, etc.) that remain conformationally conserved. As a by-product, regions in structures that are not conserved are revealed: this can indicate local molecular flexibility related to biological activity. MOTIF-EM takes cryoEM volumetric maps as inputs. The technique used by MOTIF-EM to detect conserved sub-structures is inspired by a recent breakthrough in 2D object recognition. The technique works by constructing rotationally invariant, low-dimensional representations of local regions in the input cryoEM maps. Correspondences are established between the reduced representations (by comparing them using a simple metric) across the input maps. The correspondences are clustered using hash tables and graph theory is used to retrieve conserved structural domains or motifs. MOTIF-EM has been used to extract conserved domains occurring in large macromolecular assembly maps, including as those of viruses P22 and epsilon 15, Ribosome 70S, GroEL, that remain structurally conserved in different functional states. Our method can also been used to build atomic models for some maps. We also used MOTIF-EM to identify the conserved folds shared among dsDNA bacteriophages HK97, Epsilon 15, and ô29, though they have low-sequence similarity

    Bioconversion of food waste to volatile fatty acids: impact of microbial community, pH and retention time

    Bio-based production of materials from waste streams is a pivotal aspect in a circular economy. This study aimed to investigate the influence of inoculum (three different sludge taken from anaerobic digestors), pH (5 & 10) and retention time on production of total volatile fatty acids (VFAs), VFA composition as well as the microbial community during anaerobic digestion of food waste. The highest VFA production was ∼22000 ± 1036 mg COD/L and 12927 ± 1029 mg COD/L on day 15 using the inoculum acclimated to food waste at pH 10 and pH 5, respectively. Acetic acid was the dominant VFA in the batch reactors with initial alkaline conditions, whereas both propionic and acetic acids were the dominant products in the acidic condition. Firmicutes, Chloroflexi and Bacteroidetes had the highest relative abundance in the reactors. VFA generation was positively correlated to the relative abundance of Firmicutes

    A new approach to high resolution, high contrast electron microscopy of macromolecular block copolymer assemblies

    Determining the structure of macromolecular samples is vital for understanding and adapting their function. Transmission electron microscopy (TEM) is widely used to achieve this, but, owing to the weak electron scattering cross-section of carbon, TEM images of macromolecular samples are generally low contrast and low resolution. Here we implement a fast and practically simple routine to achieve high-contrast imaging of macromolecular samples using exit wave reconstruction (EWR), revealing a new level of structural detail. This is only possible using ultra-low contrast supports such as the graphene oxide (GO) used here and as such represents a novel application of these substrates. We apply EWR on GO membranes to study self-assembled block copolymer structures, distinguishing not only the general morphology or nanostructure, but also evidence for the substructure (i.e. the polymer chains) which gives insight into their formation mechanisms and functional properties

    MRC2014: Extensions to the MRC format header for electron cryo-microscopy and tomography

    Open Access funded by Medical Research CouncilThe MRC binary file format is widely used in the three-dimensional electron microscopy field for storing image and volume data. Files contain a header which describes the kind of data held, together with other important metadata. In response to advances in electron microscopy techniques, a number of variants to the file format have emerged which contain useful additional data, but which limit interoperability between different software packages. Following extensive discussions, the authors, who represent leading software packages in the field, propose a set of extensions to the MRC format standard designed to accommodate these variants, while restoring interoperability. The MRC format is equivalent to the map format used in the CCP4 suite for macromolecular crystallography, and the proposal also maintains interoperability with crystallography software.     Details of ssDNA annealing revealed by an HSV-1 ICP8–ssDNA binary complex

    Infected cell protein 8 (ICP8) from herpes simplex virus 1 was first identified as a single-strand (ss) DNA-binding protein. It is essential for, and abundant during, viral replication. Studies in vitro have shown that ICP8 stimulates model replication reactions, catalyzes annealing of complementary ssDNAs and, in combination with UL12 exonuclease, will catalyze ssDNA annealing homologous recombination. DNA annealing and strand transfer occurs within large oligomeric filaments of ssDNA-bound ICP8. We present the first 3D reconstruction of a novel ICP8–ssDNA complex, which seems to be the basic unit of the DNA annealing machine. The reconstructed volume consists of two nonameric rings containing ssDNA stacked on top of each other, corresponding to a molecular weight of 2.3 MDa. Fitting of the ICP8 crystal structure suggests a mechanism for the annealing reaction catalyzed by ICP8, which is most likely a general mechanism for protein-driven DNA annealing

    Phase-plate electron microscopy: a novel imaging tool to reveal close-to-life nano-structures

    After slow progress in the efforts to develop phase plates for electron microscopes, functional phase plate using thin carbon film has been reported recently. It permits collecting high-contrast images of close-to-life biological structures with cryo-fixation and without staining. This report reviews the state of the art for phase plates and what is innovated with them in biological electron microscopy. The extension of thin-film phase plates to the material-less type using electrostatic field or magnetic field is also addressed

    Community- delivered infant- parent psychotherapy improves maternal sensitive caregiving: Evaluation of the Michigan model of infant mental health home visiting

    The current study evaluated the effectiveness of a home- based psychotherapeutic Infant Mental Health Home Visiting (IMH- HV) intervention for enhancing parenting sensitivity; a secondary aim was to evaluate whether the use of video feedback was associated with greater treatment response. Participants were N = 78 mothers and their children (age at entry ranged from prebirth to 24- month old (M = 9.8, SD = 8.4), who were initiating IMH- HV services with community mental health- based therapists (N = 51). Dyads were assessed during extended home visits via standardized interviews and observational and questionnaire methods within the first month of treatment (baseline), and again 6 and 12 months thereafter. Following each of these extended home visits, study evaluators completed a standard Q- sort to capture observations of maternal sensitivity during the visit. Therapists completed fidelity checklists used to derive the total number of IMH- HV sessions received (i.e., dosage) and frequency with which therapists provided video feedback. Results indicated a dose- response relationship between number of sessions and maternal sensitivity, and that video review with parents independently contributed to improved maternal sensitivity.
