“PREPARE-SE DESDE JÁ”: OS IMPRESSOS DE CUNHO CIVILIZATÓRIO E AS PRESCRIÇÕES PARA O COMPORTAMENTO FEMININO (ANOS 1950)

É na esteira das pesquisas que se ocupam em compreender a sociabilidade, a civilidade e as formas de agir no ambiente privado que pesquisamos uma coleção de impressos de cunho civilizatório, denominado “Pensando em Casar”. O objetivo foi atentar para as imagens relacionadas à família, no sentido de compreender as representações sobre a condição feminina nesse contexto. Foram analisadas 22 revistas publicadas originalmente da França (1954) e traduzidas quase que simultaneamente no Brasil (1955). Na análise dos documentos, destacamos duas categorias: “Representações de casamento e família” e “Comportamento feminino: entre a espera e a conquista”. Nesse conjunto, as representações sobre o comportamento feminino evidenciam que as mulheres a quem se destinava a revista eram favorecidas economicamente. As imagens indicam que as leitoras deveriam estar sempre preparadas para conquistar um marido, formar uma família e permanecer no matrimônio

Preservar para lembrar: vestígios da cultura material da escola no Museu Histórico Visconde de São Leopoldo (1905-1940)

Objetos, mobiliários e fotografias, enquanto documentos, contam-nos histórias e nos permitem refletir sobre o que se passou, reverberando um tempo por vezes distante e respondendo a questionamentos a eles feitos. Partindo desse pressuposto, buscamos, neste estudo, analisar os processos educativos que se passaram na cidade de São Leopoldo/RS, no início do século XX, particularmente entre os anos de 1905 e 1940. Para isso, tomamos como documento os mobiliários escolares, especialmente no que tange a fotografias e objetos escolares em exposição salvaguardados no Museu Histórico Visconde São Leopoldo. Os objetos foram registrados por meio fotográfico, catalogados e analisados a partir de sua materialidade. A análise realizada tem como temática a cultura material da escola à luz dos estudos realizados por Antonio Viñao Frago (1995, 2006), Rosa Fátima de Souza (2007) e Hernández Díaz (2002). Entendemos que o conjunto de objetos aqui analisados apresenta uma cultura escolar de práticas locais, em consonância com uma cultura nacional e, até mesmo, internacional. Under this assumption, we seek, in this study, to analyze the educational processes that occurred in the city of São Leopoldo/RS, at the beginning of the 20th century, particularly between the years 1905 and 1940. In this regard, we consider as documents the school furnishings, especially concerning exposed photographs and school objects safeguarded in the Visconde de São Leopoldo Historical Museum. The objects were photographed, catalogued, and analyzed from their materiality. The conducted analysis approaches the material culture of schools considering the studies undertaken by Antonio Viñao Frago (1995, 2006), Rosa Fátima de Souza (2007), and Hernández Díaz (2002). We understand that the set of objects analyzed here presents a school culture of local practices, in accord with a national culture, and even with an international one. O Papel do ensino do português como língua estrangeira na defesa do multicultarismo

As política actuais existentes a nível oficial para a implementação e defesa do ensino da Língua Portuguesa como Língua Estrangeira (L. E.) na Europa e no resto do mundo levam-nos a pensar que são, sobretudo, os casos isolados de leitores portugueses pioneiros, inspirados e marginais que na sua missão individual e afastada lutam pela implementação e defesa desta língua nos seus países de acolhimento. Segundo Volfgram, “cabe ensinar a alguns que o multiculturalismo não está apenas na teoria e sim ao nosso redor, nos elevando realmente à condição de seres humanos” (2005), e o mesmo é dizer que o multiculturalismo começa nas suas bases pela aprendizagem desinteressada e não interesseira das crianças na sua mais tenra idade. Não é impunemente que em países multiculturais como a Bélgica, a Língua Portuguesa ensinada como segunda língua ou como língua estrangeira desempenha um papel preponderante na defesa e na preservação do Português e, em simultâneo, pugna pela defesa incontestável da necessidade incontornável que o multiculturalismo é hoje. É indubitável que a luta contra a xenofobia, a luta pela tolerância e o respeito mútuo, bem como o diálogo profícuo biunívoco não podem sobreviver actualmente sem uma consciencialização da importância das línguas minoritárias, da crioulização, da relação com as línguas maioritárias e da conquista da defesa do multiculturalismo hic et nunc. Abordando algumas opiniões avisadas, esperamos trazer à discussão temas importantes, tais como, a necessidade de articulação de políticas de difusão da língua portuguesa na Europa e no Mundo concertadamente com o Brasil e outros Países Lusófonos, a necessidade de implementação de medidas concretas no terreno para defesa da Língua de Camões fora de Portugal, a sobrevivência do Português que embora sendo minoritária na Europa é uma das línguas mais faladas no mundo, a necessidade da consciencialização para a crescente importância geo-estratégica do Português paralelamente com o recrudescimento do multiculturalismo à escala global

Relatório de estágio - Mestrado em Ensino da MúsicaRésumé-Section I - En septembre 2012, l’auteur a été engagé comme professeur au Conservatoire à Rayonnement Départemental du Choletais en France et a pris la classe de cor constituée de 25 élèves en charge. Dans un premier temps, l’auteur présente le projet pédagogique établi pour cette classe. Il répond aux exigences liées à son cadre d’emploi et tient compte des con naissances académiques acquises lors de sa formation à l’École Supérieure de Musique de Lisbonne et à l’École Supérieure d’Éducation. Dans un second temps, lors d’une description et d’une analyse de la mise en place de ce projet pédagogique pour l’ensemble de la classe, il aborde les sujets de l’enseignement adapté aux élèves déficients, l’enseignement aux adultes et de la transversalité des enseignements. Finalement est analysé, sur la base de ce projet pédagogique, le cas d'un élève de chaque cycle d'enseignement classique.Résumé-Section II - Depuis l’abaissement de l’âge d’entrée en 1er cycle dans l’enseignement spécialisé de la musique décrété par le ministère de la Culture dans les schémas d’orientation pédagogique des 20 dernières années, les cors standards doubles utilisés jusqu’à présent représentent de par leur poids un obstacle à l’apprentissage. En 1981, suite à une formation de 3 ans comme thérapeute de technique Alexander, le corniste anglais Pip Eastop conçoit un pied-support qui lui permet de jouer du cor tout en respectant "le bon usage de soi". Cette étude se concentre sur ce pied-support et cherche à définir s’il est possible de le considérer comme un outil ergonomiquement valide pour l’acquisition des compétences fondamentales présentées en 1ère section. Sont présentés un historique de l'évolution du poids du cor, les solutions envisageables pour l'enseignement du cor, une étude empirique basée sur des questionnaires effectuée au sein de la classe de cor du Conservatoire à Rayonnement départemental du Choletais et pour conclure est abordée en discussion finale les conséquences de l'ergonomie sur l'enseignement spécialisé de la musique.Abstract-Section I - In September 2012, the author was taken on as a French horn teacher at the "Conservatoire à Rayonnement Départemental du Choletais" in France, taking over the corresponding class of 25 students. Firstly, the author presents the educational project designed specifically for this class. It meets the requirements of his job description, and reflects the academic knowledge acquired during his training at the "Escola Superior de Musica de Lisboa," as well as at the "Escola Superior da Educacao." Secondly, by describing and analysing the implementation of this educational project for the whole class, the author examines issues regarding the education of "special-needs" students, adult education and the transversatily of musical teaching syllabuses. Finally, based on the educational project, the author analyses the individual case of a student from each of the classical cycle.Abstract Section II - Since the lowering of the age of entry to the first cycle in special, musical education in a decree enacted over the past twenty years by the Ministry of Culture, the double- standard French horns used so far have been a barrier to learning, simply through their sheer weight. In 1981, after three years training as a therapist using the "Alexander Method," the English French horn player, Pip Eastop, designed a hornstick which enabled him to play while maintaining "the proper use of the self." This study focuses on the hornstick, and attempts to ascertain whether it is possible for the hornstick to be considered as an ergonomically-valid tool for the acquisition of the basic skills presented in the first section. The author presents the history of the evolution of the weight of the French horn, some possible solutions for teaching the French horn, as well as an empirical study based on questionnaires completed by members of the French horn classes at the "Conservatoire à Rayonnement Département du Choletais," before concluding by examining the ergonomic consequences on teaching methods.N/

Detection and Quantification of Leptospira interrogans in Hamster and Rat Kidney Samples: Immunofluorescent Imprints versus Real-time PCR

A major limitation in the clinical management and experimental research of leptospirosis is the poor performance of the available methods for the direct detection of leptospires. In this study, we compared real-time PCR (qPCR), targeting the lipL32 gene, with the immunofluorescent imprint method (IM) for the detection and quantification of leptospires in kidney samples from the rat and hamster experimental models of leptospirosis. Using a virulent strain of Leptospira interrogans serovar Copenhageni, a chronic infection was established in the rat model, which were euthanized 28 days post-infection, while the hamster model simulated an acute infection and the hamsters were euthanized eight days after inoculation. Leptospires in the kidney samples were detected using culture isolation, qPCR and the IM, and quantified using qPCR and the IM. In both the acute and chronic infection models, the correlation between quantification by qPCR and the IM was found to be positive and statistically significant (P<0.05). Therefore, this study demonstrates that the IM is a viable alternative for not only the detection but also the quantification of leptospires, particularly when the use of qPCR is not feasible

Aplicação da biologia molecular no diagnóstico de doenças genéticas

With the advances of the so called Genomic Medicine, the molecular analysis of genetic diseases increased the needs of a full characterization of alterations shown by an individual and is becoming essential for a differential diagnosis in several cases. The purpose of the present paper is to describe the molecular diagnostic protocols used in the Molecular Genetics Laboratory of the Medical Genetics Service of the Hospital de Clínicas of Porto Alegre. Most of these protocols are aimed at the use of updated and specialized molecular biology techniques in Brazil. The objective is not only the characterization of the affected patient’s genotype, but also the improvement of genetic counseling through the use of more sensitive and trustable techniques and strategies for the detection of carriers and for prenatal diagnosis in future pregnancies.Com os avanços da chamada Medicina Genômica, a análise molecular de doenças genéticas vem sendo cada vez mais necessária para a completa caracterização das alterações apresentadas por um indivíduo ou até mesmo essencial para um diagnóstico diferencial em muitos casos. O objetivo deste artigo é descrever os protocolos de diagnóstico molecular utilizados no laboratório de genética molecular do Serviço de Genética Médica do Hospital de Clínicas de Porto Alegre, que visam, em sua maioria, a utilização de técnicas atualizadas e especializadas de biologia molecular no Brasil, para permitir não apenas a caracterização do genótipo dos pacientes afetados mas também para melhorar o aconselhamento genético através do uso de técnicas e estratégias mais sensíveis e mais confiáveis para a detecção de portadores e para o diagnóstico pré-natal em gestações futuras

Anatomical Organization of Urocortin 3-Synthesizing Neurons and Immunoreactive Terminals in the Central Nervous System of Non-Human Primates [Sapajus spp.]

Urocortin 3 (UCN3) is a neuropeptide member of the corticotropin-releasing factor (CRF) peptide family that acts as a selective endogenous ligand for the CRF, subtype 2 (CRF2) receptor. Immunohistochemistry and in situ hybridization data from rodents revealed UCN3-containing neurons in discrete regions of the central nervous system (CNS), such as the medial preoptic nucleus, the rostral perifornical area (PFA), the medial nucleus of the amygdala and the superior paraolivary nucleus. UCN3-immunoreactive (UCN3-ir) terminals are distributed throughout regions that mostly overlap with regions of CRF2 messenger RNA (mRNA) expression. Currently, no similar mapping exists for non-human primates. To better understand the role of this neuropeptide, we aimed to study the UCN3 distribution in the brains of New World monkeys of the Sapajus genus. To this end, we analyzed the gene and peptide sequences in these animals and performed immunohistochemistry and in situ hybridization to identify UCN3 synthesis sites and to determine the distribution of UCN3-ir terminals. The sequencing of the Sapajus spp. UCN3-coding gene revealed 88% and 65% identity to the human and rat counterparts, respectively. Additionally, using a probe generated from monkey cDNA and an antiserum raised against human UCN3, we found that labeled cells are mainly located in the hypothalamic and limbic regions. UCN3-ir axons and terminals are primarily distributed in the ventromedial hypothalamic nucleus (VMH) and the lateral septal nucleus (LS). Our results demonstrate that UCN3-producing neurons in the CNS of monkeys are phylogenetically conserved compared to those of the rodent brain, that the distribution of fibers agrees with the distribution of CRF2 in other primates and that there is anatomical evidence for the participation of UCN3 in neuroendocrine control in primates

Release of NO from a nitrosyl ruthenium complex through oxidation of mitochondrial NADH and effects on mitochondria

Nitrosyl ruthenium complexes are promising NO donor agents with numerous advantages for the biologic applications of NO. We have characterized the NO release from the nitrosyl ruthenium complex [Ru(NO2)(bpy)(2)(4-pic)](+) (I) and the reactive oxygen/nitrogen species (ROS/RNS)-mediated NO actions on isolated rat liver mitochondria. The results indicated that oxidation of mitochondrial NADH promotes NO release from (I) in a manner mediated by NO2 formation (at neutral pH) as in mammalian cells, followed by an oxygen atom transfer mechanism (OAT). The NO released from (I) uncoupled mitochondria at low concentrations/incubation times and inhibited the respiratory chain at high concentrations/incubation times. In the presence of ROS generated by mitochondria NO gave rise to peroxynitrite, which, in turn, inhibited the respiratory chain and oxidized membrane protein-thiols to elicit a Ca2+-independent mitochondrial permeability transition; this process was only partially inhibited by cyclosporine-A, almost fully inhibited by the thiol reagent N-ethylmaleimide (NEM) and fully inhibited by the NO scavenger 2-(4-carboxyphenyl)-4,45,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO). These actions correlated with the release of cytochrome c from isolated mitochondria as detected by Western blotting analysis. These events, typically involved in cell necrosis and/or apoptosis denote a potential specific action of (I) and analogs against tumor cells via mitochondria-mediated processes.
