Looking into CALIPSO climatological products: Evaluation and suggestions from EARLINET
CALIPSO (Cloud-Aerosol Lidar and Pathfinder Satellite Observations) Level 3 (CL3) data were compared against EARLINET (European Aerosol Research Lidar Network) monthly averages obtained by profiles during satellite overpasses. Data from EARLINET stations of Évora, Granada, Leipzig, Naples and Potenza, equipped with advanced multi-wavelength Raman lidars were used for this study. Owing to spatial and temporal differences, we reproduced the CL3 filtering rubric onto the CALIPSO Level 2 data. The CALIPSO monthly mean profiles following this approach are called CALIPSO Level 3*, CL3*. This offers the possibility to achieve direct comparable datasets. In respect to CL3 data, the agreement typically improved, in particular above the areas directly affected by the anthropogenic activities within the planetary boundary layer. However in most of the cases a subtle CALIPSO underestimation was observed with an average bias of 0.03 km-1. We investigated the backscatter coefficient applying the same screening criteria, where the mean relative difference in respect to the extinction comparison improved from 15.2% to 11.4%. Dickkopf-related protein 1 (Dkk1) regulates the accumulation and function of myeloid derived suppressor cells in cancer
Tumor–stroma interactions contribute to tumorigenesis. Tumor cells can educate the stroma at primary and distant sites to facilitate the recruitment of heterogeneous populations of immature myeloid cells, known as myeloid-derived suppressor cells (MDSCs). MDSCs suppress T cell responses and promote tumor proliferation. One outstanding question is how the local and distant stroma modulate MDSCs during tumor progression. Down-regulation of β-catenin is critical for MDSC accumulation and immune suppressive functions in mice and humans. Here, we demonstrate that stroma-derived Dickkopf-1 (Dkk1) targets β-catenin in MDSCs, thus exerting immune suppressive effects during tumor progression. Mice bearing extraskeletal tumors show significantly elevated levels of Dkk1 in bone microenvironment relative to tumor site. Strikingly, Dkk1 neutralization decreases tumor growth and MDSC numbers by rescuing β-catenin in these cells and restores T cell recruitment at the tumor site. Recombinant Dkk1 suppresses β-catenin target genes in MDSCs from mice and humans and anti-Dkk1 loses its antitumor effects in mice lacking β-catenin in myeloid cells or after depletion of MDSCs, demonstrating that Dkk1 directly targets MDSCs. Furthermore, we find a correlation between CD15(+) myeloid cells and Dkk1 in pancreatic cancer patients. We establish a novel immunomodulatory role for Dkk1 in regulating tumor-induced immune suppression via targeting β-catenin in MDSCs
An automatic aerosol classification for earlinet: application and results
Aerosol typing is essential for understanding the impact of the different aerosol sources on climate, weather system and air quality. An aerosol classification method for EARLINET (European Aerosol Research Lidar Network) measurements is introduced which makes use the Mahalanobis distance classifier. The performance of the automatic classification is tested against manually classified EARLINET data. Manipulating the tumor immune microenvironment to improve cancer immunotherapy: IGF1R, a promising target
Cancer immunotherapy has made impressive advances in improving the outcome of patients affected by malignant diseases. Nonetheless, some limitations still need to be tackled to more efficiently and safely treat patients, in particular for those affected by solid tumors. One of the limitations is related to the immunosuppressive tumor microenvironment (TME), which impairs anti-tumor immunity. Efforts to identify targets able to turn the TME into a milieu more auspicious to current immuno-oncotherapy is a real challenge due to the high redundancy of the mechanisms involved. However, the insulin-like growth factor 1 receptor (IGF1R), an attractive drug target for cancer therapy, is emerging as an important immunomodulator and regulator of key immune cell functions. Here, after briefly summarizing the IGF1R signaling pathway in cancer, we review its role in regulating immune cells function and activity, and discuss IGF1R as a promising target to improve anti-cancer immunotherapy
Measuring the perception of quality physical education in Latin American professionals

educac¸ão fÃsica (PE) criou confusão entre os profissionais. O objetivo desta pesquisa é investigar
os fatores percebidos como importantes no desenvolvimento da educac¸ão fÃsica de qualidade
(QPE) por profissionais de paÃses da América Latina (AL). Um questionário composto por 24
itens com base no QPE foi respondido por 468 profissionais coletados em 6 cidades da América
Latina. Uma análise fatorial exploratória dos 24 itens usando extrac¸ão ML e rotac¸ão obliminar
direta foram aplicados, e os 17 itens retidos foram agrupados em uma soluc¸ão de três fatores
denominada Elementos de Desenvolvimento e Suporte para QPE na Escola (DSFQPE) ( = 0,935),
Valor essencial do QPE (CVPE) ( = 0,890) e Arranjo Curricular das Atividades FÃsicas (CAPA) (
= 0,850). Os itens retidos indicaram propriedades excelentes e o referencial básico percebido
pelos profissionais de EF em paÃses da América Latina como importante na investigac¸ão do PEQ.

An EARLINET early warning system for atmospheric aerosol aviation hazards
A stand-alone lidar-based method for detecting airborne hazards for aviation in near real time (NRT) is presented. A polarization lidar allows for the identification of irregular-shaped particles such as volcanic dust and desert dust. The Single Calculus Chain (SCC) of the European Aerosol Research Lidar Network (EARLINET) delivers high-resolution preprocessed data: the calibrated total attenuated backscatter and the calibrated volume linear depolarization ratio time series. From these calibrated lidar signals, the particle backscatter coefficient and the particle depolarization ratio can be derived in temporally high resolution and thus provide the basis of the NRT early warning system (EWS). In particular, an iterative method for the retrieval of the particle backscatter is implemented. This improved capability was designed as a pilot that will produce alerts for imminent threats for aviation. The method is applied to data during two diverse aerosol scenarios: first, a record breaking desert dust intrusion in March 2018 over Finokalia, Greece, and, second, an intrusion of volcanic particles originating from Mount Etna, Italy, in June 2019 over Antikythera, Greece. Additionally, a devoted observational period including several EARLINET lidar systems demonstrates the network's preparedness to offer insight into natural hazards that affect the aviation sector. © 2020 Author(s)
Sexual Dysfunction in People with Multiple Sclerosis: The Role of Disease Severity, Illness Perception, and Depression
Despite being a common issue in people with multiple sclerosis (pwMS), sexual dysfunction is still underinvestigated. This work aims to assess the potential determinants of sexual dysfunction in pwMS by considering its relationship with disease severity (in terms of global disability), illness perception, and depressive symptoms. In this multicenter study, 1010 pwMS responded to an online survey. A serial mediation model considering negative illness perception and depressive symptoms as mediators of the relationship between disease severity and sexual dysfunction was conducted using the SPSS PROCESS Macro with bias-corrected bootstrapping (5000 samples). Disease severity exerts an indirect effect on sexual dysfunction via illness perception, both independently and through depressive symptoms. However, the results indicated that illness perception plays a more crucial role in sexual dysfunction in pwMS with mild disability than in pwMS with moderate-severe disability. This study suggests that higher disability increases its magnitude by enhancing negative illness perception, that, in turn, affects sexual dysfunction both directly and through depressive symptoms, especially in pwMS with mild disability. Modulating the effect of illness perception by favoring adaptive coping strategies might represent a valid approach to mitigate sexual dysfunction symptoms in MS
Safety, feasibility, tolerability, and clinical effects of repeated psilocybin dosing combined with non-directive support in the treatment of obsessive-compulsive disorder: protocol for a randomized, waitlist-controlled trial with blinded ratings
BackgroundTo date, few randomized controlled trials of psilocybin with non-directive support exist for obsessive-compulsive disorder (OCD). Results and participant feedback from an interim analysis of an ongoing single-dose trial (NCT03356483) converged on the possibility of administering a higher fixed dose and/or more doses of psilocybin in future trials for presumably greater benefits.ObjectivesThis trial aims to evaluate the safety, feasibility, tolerability, and clinical effects of two doses of psilocybin paired with non-directive support in the treatment of OCD. This trial also seeks to examine whether two doses of psilocybin lead to greater OCD symptom reduction than a single dose, and to elucidate psychological mechanisms underlying the effects of psilocybin on OCD.DesignA randomized (1:1), waitlist-controlled design with blinded ratings will be used to examine the effects of two doses of oral psilocybin paired with non-directive support vs. waitlist control on OCD symptoms. An adaptive dose selection strategy will be implemented (i.e., first dose: 25 mg; second dose: 25 or 30 mg).Methods and analysisThis single-site trial will enroll 30 adult participants with treatment-refractory OCD. Aside from safety, feasibility, and tolerability metrics, primary outcomes include OCD symptoms assessed on the Yale-Brown Obsessive-Compulsive Scale – Second Edition (Y-BOCS-II). A blinded independent rater will assess primary outcomes at baseline and the primary endpoint at the end of the second dosing week. Participants will be followed up to 12 months post-second dosing. Participants randomized to waitlist will be rescreened after 7 weeks post-randomization, and begin their delayed treatment phase thereafter if still eligible.EthicsWritten informed consent will be obtained from participants. The unprecedented 2017–2018 stratospheric smoke event: decay phase and aerosol properties observed with the EARLINET
Six months of stratospheric aerosol observations with the European Aerosol Research Lidar Network (EARLINET) from August 2017 to January 2018 are presented. The decay phase of an unprecedented, record-breaking stratospheric perturbation caused by wildfire smoke is reported and discussed in terms of geometrical, optical, and microphysical aerosol properties. Enormous amounts of smoke were injected into the upper troposphere and lower stratosphere over fire areas in western Canada on 12 August 2017 during strong thunderstorm–pyrocumulonimbus activity. The stratospheric fire plumes spread over the entire Northern Hemisphere in the following weeks and months. Twenty-eight European lidar stations from northern Norway to southern Portugal and the eastern Mediterranean monitored the strong stratospheric perturbation on a continental scale. The main smoke layer (over central, western, southern, and eastern Europe) was found at heights between 15 and 20 km since September 2017 (about 2 weeks after entering the stratosphere). Thin layers of smoke were detected at heights of up to 22–23 km. The stratospheric aerosol optical thickness at 532 nm decreased from values > 0.25 on 21–23 August 2017 to 0.005–0.03 until 5–10 September and was mainly 0.003–0.004 from October to December 2017 and thus was still significantly above the stratospheric background (0.001–0.002). Stratospheric particle extinction coefficients (532 nm) were as high as 50–200 Mm−1 until the beginning of September and on the order of 1 Mm−1 (0.5–5 Mm−1) from October 2017 until the end of January 2018. The corresponding layer mean particle mass concentration was on the order of 0.05–0.5 µg m−3 over these months. Soot particles (light-absorbing carbonaceous particles) are efficient ice-nucleating particles (INPs) at upper tropospheric (cirrus) temperatures and available to influence cirrus formation when entering the tropopause from above. We estimated INP concentrations of 50–500 L−1 until the first days in September and afterwards 5–50 L−1 until the end of the year 2017 in the lower stratosphere for typical cirrus formation temperatures of −55 ∘C and an ice supersaturation level of 1.15. The measured profiles of the particle linear depolarization ratio indicated a predominance of nonspherical smoke particles. The 532 nm depolarization ratio decreased slowly with time in the main smoke layer from values of 0.15–0.25 (August–September) to values of 0.05–0.10 (October–November) and < 0.05 (December–January). The decrease of the depolarization ratio is consistent with aging of the smoke particles, growing of a coating around the solid black carbon core (aggregates), and thus change of the shape towards a spherical form. We found ascending aerosol layer features over the most southern European stations, especially over the eastern Mediterranean at 32–35∘ N, that ascended from heights of about 18–19 to 22–23 km from the beginning of October to the beginning of December 2017 (about 2 km per month).
