    Modelling of Natural Convective Heating of a Standard Wet Brick for Oven Energy Consumption Tests

    The environmental impact of buildings is directly influenced by low-efficiency appliances like electric ovens. Their energy class is estimated through a test prescribed by the EN 60350 European standard, where a wet clay brick is heated under set conditions and the energy consumption is measured; throughout the test, particular attention is devoted to the control of air temperature. In this work, a transient model of the oven suitable for control design was realized including the heat and mass transfer within the brick. A lumped-parameter approach was used to ensure good predictive properties and low computational cost. The dynamics of the cavity air and the Pt500, which is used in normal operation to maintain the desired set point, was also included in the model. Model parameters were determined through an optimization procedure based on the experimental data collected during energy consumption tests with the oven in natural convective heating mode. The model was then validated. Good results were obtained in the prediction of both temperature and heating time

    Antiepileptogenic effects of trilostane in the kainic acid model of temporal lobe epilepsy

    Objective: Epileptogenesis after status epilepticus (SE) has a faster onset in rats treated to reduce brain levels of the anticonvulsant neurosteroid allopregnanolone with the 5α-reductase inhibitor finasteride; however, it still has to be evaluated whether treatments aimed at increasing allopregnanolone levels could result in the opposite effect of delaying epileptogenesis. This possibility could be tested using the peripherally active inhibitor of 3β-hydroxysteroid dehydrogenase/Δ5-4 isomerase trilostane, which has been shown repeatedly to increase allopregnanolone levels in the brain. Methods: Trilostane (50 mg/kg) was administered subcutaneously once daily for up to six consecutive days, starting 10 min after intraperitoneal administration of kainic acid (15 mg/kg). Seizures were evaluated by video-electrocorticographic recordings for 70 days maximum, and endogenous neurosteroid levels were assessed by liquid chromatography–electrospray tandem mass spectrometry. Immunohistochemical staining was performed to evaluate the presence of brain lesions. Results: Trilostane did not alter the latency of kainic acid-induced SE onset or its overall duration. When compared to the vehicle-treated group, rats receiving six daily trilostane injections presented a remarkable delay of the first spontaneous electrocorticographic seizure and subsequent tonic–clonic spontaneous recurrent seizures (SRSs). Conversely, rats treated with only the first trilostane injection during SE did not differ from vehicle-treated rats in developing the SRSs. Notably, trilostane did not modify neuronal cell densities or the overall damage in the hippocampus. In comparison to the vehicle group, repeated administration of trilostane significantly decreased the activated microglia morphology in the subiculum. As expected, allopregnanolone and other neurosteroid levels were remarkably increased in the hippocampus and neocortex of rats treated for 6 days with trilostane, but pregnanolone was barely detectable. Neurosteroids returned to basal levels after a week of trilostane washout.. Significance: Overall, these results suggest that trilostane led to a remarkable increase in allopregnanolone brain levels, which was associated with protracted effects on epileptogenesis

    Immunologic activation of human syncytiotrophoblast by Plasmodium falciparum

    <p>Abstract</p> <p>Background</p> <p>Malaria during pregnancy is characterized by the sequestration of malaria-infected red blood cells (iRBC) in the intervillous spaces of the placenta, often accompanied by the infiltration of maternal mononuclear cells, causing substantial maternal and foetal/infant morbidity. The iRBC bind to receptors expressed by the syncytiotrophoblast (ST). How ST responds to this interaction remains poorly understood. Because it is known that ST is immunoactive and can respond to infectious agents, the consequences of this ST-iRBC interaction should be investigated.</p> <p>Methods</p> <p>An in vitro system was used to assess the biochemical and immunological changes induced in ST by ST-adherent iRBCs. Changes in ST mitogen-activated protein kinase (MAPK) activation were assessed by immunoblotting and mRNA expression levels of selected cytokine and chemokines in primary ST bound by iRBC were determined using real-time, reverse transcription PCR. In addition, secreted cytokine and chemokine proteins were assayed by standard ELISA, and chemotaxis of PBMC was assessed using a two-chamber assay system.</p> <p>Results</p> <p>Following iRBC/ST interaction, ST C-Jun N-terminal kinase 1 (JNK1) was activated and modest increases in the mRNA expression of TGF-β and IL-8/CXCL8 were observed. In addition, this interaction increased secretion of MIF and MIP-1α/CCL3 by ST and induced migration of PBMC towards iRBC-stimulated ST.</p> <p>Conclusion</p> <p>Results from this study provide the first evidence that ST participates in shaping the local immunological milieu and in the recruitment of maternal immune cells to the maternal blood space during placental malaria infection.</p

    Decreasing time consumption of microscopy image segmentation through parallel processing on the GPU

    The computational performance of graphical processing units (GPUs) has improved significantly. Achieving speedup factors of more than 50x compared to single-threaded CPU execution are not uncommon due to parallel processing. This makes their use for high throughput microscopy image analysis very appealing. Unfortunately, GPU programming is not straightforward and requires a lot of programming skills and effort. Additionally, the attainable speedup factor is hard to predict, since it depends on the type of algorithm, input data and the way in which the algorithm is implemented. In this paper, we identify the characteristic algorithm and data-dependent properties that significantly relate to the achievable GPU speedup. We find that the overall GPU speedup depends on three major factors: (1) the coarse-grained parallelism of the algorithm, (2) the size of the data and (3) the computation/memory transfer ratio. This is illustrated on two types of well-known segmentation methods that are extensively used in microscopy image analysis: SLIC superpixels and high-level geometric active contours. In particular, we find that our used geometric active contour segmentation algorithm is very suitable for parallel processing, resulting in acceleration factors of 50x for 0.1 megapixel images and 100x for 10 megapixel images

    Design and construction of a new metering hot box for the in situ hygrothermal measurement in dynamic conditions of historic masonries

    The main purpose of the HeLLo project is to contribute to data available on the literature on the real hygrothermal behavior of historic walls and the suitability of insulation technologies. Furthermore, it also aims at minimizing the energy simulation errors at the design phase and at improving their conservation features. In this framework, one of the preliminary activities of the study is the creation of a real in situ hot box to measure and analyze different insulation technologies applied to a real historic wall, to quantify the hygrothermal performance of a masonry building. Inside this box, 'traditional' experiments can be carried out: recording heat flux, surface temperature, and air temperatures, as well as relative humidity values through the use of a new sensing system (composed of thermocouples and temperature/relative humidity combined sensors). Within this paper, the process of development, construction, and validation of this new metering box is exhibited. The new hot box, specifically studied for historic case studies, when compared to other boxes, presents other advantages compared to previous examples, widely exemplified

    Development of a compatible, low cost and high accurate conservation remote sensing technology for the hygrothermal assessment of historic walls

    This article aims to properly assess the hygrothermal properties of walls located in historic buildings, this study discloses the development of a remote sensing technology compatible with an in-situ measurement implemented in Palazzo Tassoni (Italy). As required by the international recommendations adapted to cultural heritage (CH), this monitoring system balances CH conservation, performance aspects and economic costs using an integrated multidisciplinary approach. Electronics for measurement of environmental parameters is composed of sensor measurements, data acquisition system and data storage and communication system. Data acquisition system, equipped with standard modbus-rtu interface, is designed to run standalone and it is based on two cloned single board PCs to reduce the possibility of data loss. In order to reduce the costs, RaspberryPI single board PCs were chosen. These run a C/C++ software based on standard modbus library and designed to implement multi-client server TCP/IP to allow communication with other devices. Storage and communication systems are based on an industrial PC; it communicates with sensor measurements' system through a modbus-TCPIP bridge. PC runs a Labview software to provide data storage on a local database and graphical user interface to properly see all acquired data. Herein, some sensing options and approaches of measurement are described, unveiling different possible ways of enhancing the retrofit of CH with adapted technology

    Augmentation of endogenous neurosteroid synthesis alters experimental status epilepticus dynamics

    Neurosteroids can modulate γ-aminobutyric acid type A receptor-mediated inhibitory currents. Recently, we discovered that the neurosteroids progesterone, 5α-dihydroprogesterone, allopregnanolone, and pregnanolone are reduced in the cerebrospinal fluid of patients with status epilepticus (SE). However, it is undetermined whether neurosteroids influence SE. For this reason, first we evaluated whether the inhibitor of adrenocortical steroid production trilostane (50 mg/kg) could modify the levels of neurosteroids in the hippocampus and neocortex, and we found a remarkable increase in pregnenolone, progesterone, 5α-dihydroprogesterone, and allopregnanolone levels using liquid chromatography tandem mass spectrometry. Second, we characterized the dynamics of SE in the presence of the varied neurosteroidal milieu by a single intraperitoneal kainic acid (KA; 15 mg/kg) injection in trilostane-treated rats and their controls. Convulsions started in advance in the trilostane group, already appearing 90 minutes after the KA injection. In contrast to controls, convulsions prevalently developed as generalized seizures with loss of posture in the trilostane group. However, this effect was transient, and convulsions waned 2 hours before the control group. Moreover, electrocorticographic traces of convulsions were shorter in trilostane-treated rats, especially at the 180-minute (P &lt; .001) and 210-minute (P &lt; .01) time points. These findings indicate that endogenous neurosteroids remarkably modulate SE dynamics
