    BEYOND PRESCRIPTIVE APPROACHES AND TOWARDS LOCALIZED KNOWLEDGE: THE EXPERIENCES OF ALTERNATIVELY CERTIFIED FOREIGN LANGUAGE TEACHERS AS THEY ATTEMPT TO IMPLEMENT COMMUNICATIVE LANGUAGE TEACHING IN U.S. URBAN SCHOOLS

    The acknowledgment of the role that diverse sociocultural realities have on teachers’ pedagogical choices have led to the emergence of context-sensitive pedagogies demanding the abandonment of hegemonic discourses such as Communicative Language Teaching (CLT). Studies on CLT’s exportability for non-Western countries have recently proliferated in foreign language literature. Critical studies on the suitability of CLT within disenfranchised communities in the U.S. are however non-existent. This qualitative multicase study addressed a gap in the literature by exploring the situated practices of alternatively certified FL teachers practicing in urban schools in the U.S. Specifically, this study examined a) FL teachers understanding of CLT and b) the contextual factors hindering or promoting the implementation of CLT in urban schools. Using Activity Theory (AT) as an analytical tool, the study unveiled a plethora of tensions between CLT and the pedagogical approaches dominant in urban schools. Finding from this study suggest that a strong culture of rules and assessments, lack of culturally relevant materials and major philosophical incongruence amongst those in charge of participants’ evaluation and development had a strong effect on teachers’ understanding, perception and implementation of CLT. Moreover, lacking the language and professional support they needed to gain agency within the accountability discourse dominant in their urban placements, participants in this study found themselves often voiceless and in need to assimilate. Finally, this study provides recommendations for the future training and support of alternatively certified urban language teachers and pushes for the recognition and promotion of teachers’ localized experiences as valid and important sources of knowledge and expertise

    Dipteran chromosomes: a simple method for obtaining high quality chromosomal preparations

    For cytogenetic studies it is necessary to develop an efficient method of chromosome preparation with wellpreserved and dispersed chromosomes and little or no cell-wall debris. In this study, we describe a method for obtaining high quality chromosomal preparations of dipteran cells. This procedure combines drop method and air-dry technique and produces a cell suspension with separated and mixed cells. Suspended mitotic cells were then dropped onto a glass slide in order to distribute the cytoplasm and spread the chromosomes. We compare and evaluate different chromosome preparation methods to develop a more reliable procedure for the resolution of chromosome characteristics and chromosome bandings in blow fly mitotic chromosomes. The combined drop and air-dry technique described here is convenient for identification of sex chromosomes. Using this protocol, acceptable high-quality chromosomal spreads with flattened cells and no cell debris, and without damage and/or loss of chromosomes were obtained.Fil: Chirino, Monica Gabriela.     Lessons Learned from Implementing Unconscious Bias Training at an Academic Medical Center

    Introduction. In 2018, our Midwestern university medical center began offering unconscious bias training. Each session concluded with a standard evaluation. We analyzed two years of data that focused on three areas: 1) whether demographic differences or amount of prior knowledge on the topic influenced the training experience; 2) what participants liked best about the training; and 3) whether participants’ stated intentions to apply their learning at the end of the training aligned with institutional goals of the training. Methods. Participants attended sessions open to the campus community pre-scheduled by the Office for Diversity, Equity, and Inclusion and posted on its website. Chi-square tests were utilized to test associations between outcomes and questionnaire responses. Outcome measures included race/ethnicity, prior knowledge level, and overall rating of the training. Thematic analysis was used to code comments and establish themes from two open-ended survey questions. Results. Significant differences were found by race and ethnicity for all questionnaire responses; each were p < 0.001. Those who reported they had advanced/expert knowledge on the topic were less likely to report the training increased their knowledge, and those who reported their race as White/Caucasian tended to give the training the highest overall rating, as did heterosexuals. Through thematic analysis, participants valued the interactive nature of the training sessions, the use of storytelling, and the safety of the learning environment. Participants’ intention to apply their learning indicated they had gained general awareness of bias and settings where it might influence their work. Conclusions. In an effort to foster a better working and learning environment for those who are underrepresented in the health professions, training was provide that may not have met the expectations of all participants. At the same time, participants who identified as White clearly increased their awareness of bias. Therefore, it is recommended to move away from one-size-fits-all unconscious bias training and develop a robust training continuum to provide ongoing advancement for diverse audiences

    SIRT1 Mediates Melatonin’s Effects on Microglial Activation in Hypoxia: In Vitro and In Vivo Evidence

    Melatonin exerts direct neuroprotection against cerebral hypoxic damage, but the mechanisms of its action on microglia have been less characterized. Using both in vitro and in vivo models of hypoxia, we here focused on the role played by silent mating type information regulation 2 homolog 1 (SIRT1) in melatonin’s effects on microglia. Viability of rat primary microglia or microglial BV2 cells and SH-SY5Y neurons was significantly reduced after chemical hypoxia with CoCl2 (250 µM for 24 h). Melatonin (1 µM) significantly attenuated CoCl2 toxicity on microglia, an effect prevented by selective SIRT1 inhibitor EX527 (5 µM) and AMP-activated protein kinase (AMPK) inhibitor BML-275 (2 µM). CoCl2 did not modify SIRT1 expression, but prevented nuclear localization, while melatonin appeared to restore it. CoCl2 induced nuclear localization of hypoxia-inducible factor-1α (HIF-1α) and nuclear factor-kappa B (NF-kB), an effect contrasted by melatonin in an EX527-dependent fashion. Treatment of microglia with melatonin attenuated potentiation of neurotoxicity. Common carotid occlusion was performed in p7 rats, followed by intraperitoneal injection of melatonin (10 mg/kg). After 24 h, the number of Iba1+ microglia in the hippocampus of hypoxic rats was significantly increased, an effect not prevented by melatonin. At this time, SIRT1 was only detectable in the amoeboid, Iba1+ microglial population selectively localized in the corpus callosum. In these cells, nuclear localization of SIRT1 was significantly lower in hypoxic animals, an effect prevented by melatonin. NF-kB showed an opposite expression pattern, where nuclear localization in Iba1+ cells was significantly higher in hypoxic, but not in melatonin-treated animals. Our findings provide new evidence for a direct effect of melatonin on hypoxic microglia through SIRT1, which appears as a potential pharmacological target against hypoxic-derived neuronal damage.Fil: Merlo, Sara.     Mechanisms of Action of Kefir in Chronic Cardiovascular and Metabolic Diseases

    The gut microbiota maintains a complex mutual interaction with different organs of the host. Whereas in normal conditions this natural community of trillions of microorganisms greatly contributes to the human health, gut dysbiosis is related with onset or worsening of diverse chronic systemic diseases. Thus, the reestablishment of gut microbiota homeostasis with consumption of prebiotics and probiotics may be a relevant strategy to prevent or attenuate several cardiovascular and metabolic complications. Among these functional foods, the synbiotic kefir, which is a fermented milk composed of a mixture of bacteria and yeasts, is currently the most used and has attracted the attention of health care professionals. The present review is focused on reports describing the feasibility of kefir consumption to provide benefits in cardiometabolic diseases, including hypertension, vascular endothelial dysfunction, dyslipidemia and insulin resistance. Interestingly, recent studies show that mechanisms of actions of kefir in cardiometabolic diseases include recruitment of endothelial progenitor cells, improvement of the balance vagal/sympathetic nervous system, diminution of excessive generation of reactive oxygen species, angiotensin converting enzyme inhibition, anti-inflammatory cytokines profile and alteration of the intestinal microbiota. These findings provide a better understanding about the mechanisms of the beneficial actions of kefir and motivate further investigations to determine whether the use of this synbiotic could also be translated into clinical improvements in cardiometabolic diseases

    Pediatric televisits and telephone triage: impact on use of a hospital emergency department

    Objetivos: analizar las características de las teleconsultas y triajes telefónicos pediátricos atendidos por CatSalut Respon y describir su impacto sobre la actitud y la decisión final de los padres-usuarios de acudir o no a urgencias. Método: estudio observacional transversal. Durante la teleconsulta los pacientes se clasificaron según los niveles del sistema español de triaje. Aquellos que fueron derivados a urgencias se volvieron a clasificar en el hospital, y se compararon los niveles de triaje. Posteriormente, se realizó una llamada de verificación. Se recogieron variables sociodemográficas y clínicas. Resultados: se analizaron 370 teleconsultas, fundamentalmente no urgentes (n = 300; 81%). Un 20,3% (n = 75) fueron derivadas a urgencias. La fiebre (p = 0,002) y las dudas de medicación (p < 0,001) fueron motivos significativos de teleconsulta no urgente. Casi un 46% de los casos con niveles de gravedad altos en el triaje de la llamada también fueron clasificados con niveles de gravedad altos en el triaje posterior realizado en el servicio de urgencias hospitalario, mostrando una concordancia moderada. Más del 50% de los padres tenían intención de acudir a urgencias antes de la teleconsulta y un 46% cambiaron de actitud tras realizar esta llamada. Conclusiones: fiebre y dudas de medicación fueron motivos estadísticamente significativos de teleconsulta no urgente. La consulta telefónica produjo un cambio de actitud en casi la mitad de los padres
