    Dietary lipid level affects growth performance and nutrient utilisation of Senegalese sole (Solea senegalensis) juveniles

    Over the last few years, several aspects of Senegalese sole (Solea senegalensis) culture have been developed and optimised but the dietary lipid level for optimal growth has never been determined. Hence, five isonitrogenous diets (56 % dietary protein) with increasing dietary lipid levels (4, 8, 12, 16 and 20 % DM) were fed to satiation to triplicate groups of twenty fish (mean initial weight 10 g). Fifteen tanks were randomly assigned one of the five diets. Feed was distributed using automatic feeders, and fish were fed over a 16-week period. At the end of the experiment the fish fed on diets containing the two lowest dietary lipid levels (4 and 8 %) showed a 3-fold body-weight increase with a significantly higher daily growth index than fish fed higher lipid levels (1-2 v. 0-8). Moreover, these fish displayed a significantly lower dry feed intake (12g/kg per d) and feed conversion ratio (1-0) compared with fish fed higher lipids levels (16-19 g/kg per d; feed conversion ratio 2-0). Low dietary lipid levels (< 12 %) significantly improved nutrient retention and gain and hence growth, without major effects on whole-body composition. Despite the slight alteration in n-3 PUFA muscle content in the fish fed low-fat-diets, this fish fed low dietary lipid still remains a rich n-3 PUFA product and generally maintained its nutritional value. These results evidenced a low lipid tolerance of Senegalese sole juveniles and suggest a maximal dietary inclusion level of 8 % lipids for both optimal growth and nutrient utilisation without compromising flesh quality.IDEIA; European fund FEDERinfo:eu-repo/semantics/publishedVersio

    Growth and nutritional responses of bean and soybean genotypes to elevated CO2 in a controlled environment

    In the current situation of a constant increase in the atmospheric CO2 concentration, there is a potential risk of decreased nutritional value and food crop quality. Therefore, selecting strong-responsive varieties to elevated CO2 (eCO2) conditions in terms of yield and nutritional quality is an important decision for improving crop productivity under future CO2 conditions. Using bean and soybean varieties of contrasting responses to eCO2 and different origins, we assessed the effects of eCO2 (800 ppm) in a controlled environment on the yield performance and the concentration of protein, fat, and mineral elements in seeds. The range of seed yield responses to eCO2 was −11.0 to 32.7% (average change of 5%) in beans and −23.8 to 39.6% (average change of 7.1%) in soybeans. There was a significant correlation between seed yield enhancement and aboveground biomass, seed number, and pod number per plant. At maturity, eCO2 increased seed protein concentration in beans, while it did not affect soybean. Lipid concentration was not affected by eCO2 in either legume species. Compared with ambient CO2 (aCO2), the concentrations of manganese (Mn), iron (Fe), and potassium (K) decreased significantly, magnesium (Mg) increased, while zinc (Zn), phosphorus (P), and calcium (Ca) were not changed under eCO2 in bean seeds. However, in soybean, Mn and K concentrations decreased significantly, Ca increased, and Zn, Fe, P, and Mg concentrations were not significantly affected by eCO2 conditions.     Processed by‐products from soy beverage (Okara) as sustainable ingredients for nile tilapia (o. niloticus) juveniles: effects on nutrient utilization and muscle quality

    The apparent digestibility coefficients (ADCs) of differently processed okara meals were assessed in Nile tilapia diets: dried okara not autoclaved (FOK), dried okara autoclaved (AOK), okara hydrolyzed with Alcalase (ALOK) or Cynara cardunculus proteases (CYOK), and hydrolyzed okara fermented with lactic bacteria: Lactobacillus rhamnosus R11 (CYR11OK) or Bifidobacterium animalis ssp. lactis Bb12 (CYB12OK). Okara processing significantly affected nutrient digestibility: dry matter ADC was highest in CYR11OK (80%) and lowest in FOK (40%). The lowest protein digestibility was observed in CYR11OK (72%), and the highest in AOK (97%) and CYOK (91%), evidencing the effectiveness of the autoclave and the use of C. Cardunculus proteases to increase okara protein bioavailability. The inclusion of up to 20% of AOK or CYOK did not affect fish growth, nutrient utilization, or whole body composition of Nile tilapia. The flesh quality (color, pH, water activity, cohesiveness, elasticity and resilience) was not affected by the dietary incorporation of AOK or CYOK. Fish fed with AOK diets stand out for their high density of muscle fibers, particularly in AOK20, which can explain their high muscle firmness and may result in further hypertrophic growth.     Protein utilisation and intermediary metabolism of Senegalese sole (Solea senegalensis) as a function of protein:lipid ratio

    Previous experiments with Senegalese sole (Solea senegalensis) have demonstrated that dietary lipid levels above 8% impaired growth and did not promote protein retention. We hypothesised that this low ability to use high-lipid diets may depend on the dietary protein level. In the present study, a 2 x 2 factorial design was applied where two dietary lipid (4-17% DM) and two dietary protein (below and above the requirement levels, 48 and 54% DM) levels were tested in juveniles for 114 d. Growth performance was not improved by the increase in dietary fat, irrespectively of the dietary protein levels. Protein retention was similar among the diets, although fish fed the diets with high lipid content resulted in significantly lower protein gain. Among the enzymes involved in amino acid catabolism, only aspartate aminotransferase activity in the liver was affected by the dietary lipid levels, being stimulated in fish fed high-lipid diets. Moreover, phosphofructokinase 1 activity was significantly elevated in the muscle of Senegalese sole fed 4% lipid diets, suggesting enhanced glycolysis in the muscle when the dietary lipid supply was limited and dietary starch increased. The results confirmed that high-lipid diets do not enhance growth, and data from the selected enzymes support the assumption that lipids are not efficiently used for energy production and protein sparing, even when dietary protein is below the protein requirement of the species.     Stress response of European seabass (Dicentrarchus labrax) fed plant-based diets supplemented with swine blood hydrolysates

    To improve fish welfare, it is essential that aquafeeds are designed to help fish cope with the stressful conditions of fish farms. One effective strategy to achieve this goal is to supplement the diet with bioactive hydrolysates. Here, diet supplementation to modulate oxidative stress after air exposure was investigated in European seabass, using swine blood hydrolysates (BH), obtained either by autohydrolysis (AH) or enzymatically. The enzymatically produced BH were further submitted to a micro- (RMF) and nanofiltration (RNF). Four isolipidic, isoproteic and isoenergetic diets were developed: a plant-based diet with low (12.5%) fishmeal levels (control, CTRL) and three diets where 3% of each BH (RMF, RNF and AH) was added to the CTRL. Diets were assigned to triplicate groups of 71 European seabass juveniles (initial weight 12.3 ± 1.4 g). After 12 weeks, 9 fish per treatment were either immediately sampled or air-exposed for 1 min and let to recover in a new system for 6 h prior to sampling. Stress response increased cortisol levels, followed by an increment in plasma lactate. The challenge increased liver lipid peroxidation (LPO) due to reactive oxygen species (ROS) accumulation. Carbonyls decreased post-stress, maybe due to a possible interaction with the LPO radicals, reducing protein oxidation. None of the BH improved plasma stress response. By reducing catalase levels without increasing LPO, the RNF treatment appears to adjust European seabass' antioxidant defences, indicating its potential to supply exogenous antioxidants to combat oxidative stress induced by ROS. However, this impact was not sufficient to lower LPO levels compared to a control plant-based diet. The tested diets seemed to affect the fish oxidative stress response in the liver, possibly due to the presence of bioactive peptides, which aided in the non-enzymatic modulation of stress response, as observed by the total antioxidant capacity values in the liver.info:eu-repo/semantics/publishedVersio

    Silica Aerogels/Xerogels Modified with Nitrogen-Containing Groups for Heavy Metal Adsorption

    Heavy metals are common inorganic pollutants found in the environment that have to be removed from wastewaters and drinking waters. In this work, silica-derived aerogels and xerogels were modified via a co-precursor method to obtain functional adsorbents for metal cations. A total of six formulations based upon four different functional precursors were prepared. The materials' structural characterization revealed a decreased porosity and surface area on modified samples, more prominent in xerogel counterparts. Preliminary tests were conducted, and the prepared samples were also compared to activated carbon. Three samples were selected for in-depth studies. Isotherm studies revealed that the pre-selected samples remove well copper, lead, cadmium and nickel, and with similar types of interactions, following a Langmuir trend. The adsorption kinetics starts very fast and either equilibrium is reached quickly or slowly, in a two-stage process attributed to the existence of different types of active sites. Based on the previous tests, the best sample, prepared by mixing different functional co-precursors, was selected and its behavior was studied under different temperatures. For this material, the adsorption performance at 20 °C is dependent on the cation, ranging from 56 mg·g-1 for copper to 172 mg·g-1 for lead

    High levels of dietary lipids are incorporated in feeds for most teleost fish to promote growth and reduce nitrogen waste. However, in Senegalese sole (Solea senegalensis) previous studies revealed that increasing the level of dietary lipids above 8% negatively affect growth and nutrient utilization regardless of dietary protein content. It has been shown that glucose regulation and metabolism can be impaired by high dietary fat intake in mammals, but information in teleost fish is scarce. The aim of this study was to assess the possible effect of dietary lipids on glucose metabolism in Senegalese sole with special emphasis on the regulation of proteins involved in the muscle insulin-signalling pathway. Senegalese sole juveniles (29 g) were fed two isonitrogenous diets (53% dry matter) for 88 days. These two diets were one with a high lipid level (∼17%, HL) and a moderate starch content (∼14%, LC), and the other being devoid of fish oil (4% lipid, LL) and with high starch content (∼23%, HC). Surprisingly, feeding Senegalese sole the HL/LC diet resulted in prolonged hyperglycaemia, while fish fed on LL/HC diet restored basal glycaemia 2 h after feeding. The hyperglycaemic phenotype was associated with greater glucose-6-phosphatase activity (a key enzyme of hepatic glucose production) and lower citrate synthase activity in the liver, with significantly higher liver glycogen content. Sole fed on HL/LC diet also had significantly lower hexokinase activity in muscle, although hexokinase activity was low with both dietary treatments. The HL/LC diet was associated with significant reductions in muscle AKT, p70 ribosomal S6-K1 Kinase (S6K-1) and ribosomal protein S6 (S6) 2 h after feeding, suggesting down regulation of the AKT-mTOR nutrient signalling pathway in these fish.
