    Anthelmintic action of plant cysteine proteinases against the rodent stomach nematode, Protospirura muricola, in vitro and in vivo

    Cysteine proteinases from the fruit and latex of plants, including papaya, pineapple and fig, were previously shown to have a rapid detrimental effect, in vitro, against the rodent gastrointestinal nematodes, H eligmosomoides polygyrus (which is found in the anterior small intestine) and Trichuris miti,is (which resides in the caecum). Proteinases in the crude latex of papaya also showed anthelmintic efficacy against both nematodes in vivo. In this paper, we describe the in vitro and in vivo effects of these plant extracts against the rodent nematode, Protospirua muricola, which is found in the stomach. As in earlier work, all the plant cysteine proteinases examined, with the exception of actinidain from the juice of kiwi fruit, caused rapid loss of motility and digestion of the cuticle, leading to death of the nematode in vitro. In vivo, in contrast to the efficacy against H. polygyrus and T. muris, papaya latex only showed efficacy against P. muricola adult female worms when the stomach acidity had been neutralized prior to administration of papaya latex. Therefore, collectively, our studies have demonstrated that, with the appropriate formulation, plant cysteine proteinases have efficacy against nematodes residing throughout the rodent gastrointestinal tract

    We examined the mechanism of action and compared the anthelmintic efficacy of cysteine proteinases from papaya, pineapple, fig, kiwi fruit and Egyptian milkweed in vitro using the rodent gastrointestinal nematode Heligmosomoides polygyrus. Within a 2 h incubation period, all the cysteine proteinases, with the exception of the kiwi fruit extract, caused marked damage to the cuticle of H. polygyrus adult male and female worms, reflected in the loss of surface cuticular layers. Efficacy was comparable for both sexes of worms, was dependent on the presence of cysteine and was completely inhibited by the cysteine proteinase inhibitor, E-64. LD50 values indicated that the purified proteinases were more efficacious than the proteinases in the crude latex, with purified ficin, papain, chymopapain, Egyptian milkweed latex extract and pineapple fruit extract, containing fruit bromelain, having the most potent effect. The mechanism of action of these plant enzymes (i.e. an attack on the protective cuticle of the worm) suggests that resistance would be slow to develop in the field. The efficacy and mode of action make plant cysteine proteinases potential candidates for a novel class of anthelmintics urgently required for the treatment of humans and domestic livestock

    Effects of feeding corn silage, pelleted, ensiled, or pelleted and ensiled alfalfa on growth and carcass characteristics of lamb

    Two experiments were conducted to evaluate the effects of corn silage (CS) and alfalfa (pelleted (AP), haylage (AH), or combination (APH)) on lamb growth and carcass characteristics. The objective of Experiment 1 (Exp. 1) was to compare AH to CS in lamb feedlot diets on lamb growth and carcass characteristics. Eighty lambs were used in a 56 day experiment with a randomized complete block design. The objective of Experiment 2 (Exp. 2) was to determine the effects of alfalfa form, AP, AH or AHP, on animal performance and carcass characteristics. Seventy two lambs were used in an 82 day experiment with a randomized complete block design. In Exp. 1, lambs offered AH consumed 23.5% more feed on a daily basis than lambs offered CS. However, lambs fed CS gained weight 21.3% faster than lambs fed AH (259 versus 213 g/day, respectively). Additionally, lambs fed CS were 50.4% more efficient in converting feed to gain compared with lambs offered AH (0.173 versus 0.115 kg gain/kg feed, respectively). In Exp. 2, AP resulted in a greater dry matter intake (DMI) and average daily gain (ADG), and fewer days on feed than with AH. The combination of pellets + haylage resulted in a greater DMI, ADG, and fewer days on feed compared to AH alone. Therefore, there was an inverse relationship between both forage particle size and diet moisture content with DMI. There was no difference in daily efficiency of gain due to the form of alfalfa. Average daily gain, days on feed, and DMI are economically important criteria that differ due to the form of alfalfa even when feed efficiencies are similar

    An in-depth look into PTSD-depression comorbidity: A longitudinal study of chronically-exposed Detroit residents

    Background Although PTSD-major depressive disorder (MDD) co-morbidity is well-established, the vast majority of studies have examined comorbidity at the level of PTSD total severity, rather than at the level of specific PTSD symptom clusters. This study aimed to examine the long-term associations between MDD and PTSD symptom clusters (intrusion, avoidance, hyperarousal), and the moderating role of gender in these associations. Methods 942 residents of urban Detroit neighborhoods were interviewed at 3 waves, 1 year apart. At each wave, they were assessed for PTSD, depression, trauma exposure, and stressful life events. Results At all waves, hyperarousal was the PTSD cluster most strongly correlated with MDD. For the full sample, a reciprocal relationship was found between MDD and all three PTSD clusters across time. Interestingly, the relative strength of associations between MDD and specific PTSD clusters changed over time. Women showed the same bidirectional MDD-PTSD pattern as in the entire sample, while men sometimes showed non-significant associations between early MDD and subsequent PTSD clusters. Limitations First, our analyses are based on DSM-IV criteria, as this was the existing edition at the time of this study. Second, although this is a longitudinal study, inferences regarding temporal precedence of one disorder over another must be made with caution. Conclusions Early identification of either PTSD or MDD following trauma may be crucial in order to prevent the development of the other disorder over time. The PTSD cluster of hyper-arousal may require special therapeutic attention. Also, professionals are encouraged to develop more gender-specific interventions post-trauma

    Mineral reaction kinetics constrain the length scale of rock matrix diffusion

    Mass transport by aqueous fluids is a dynamic process in shallow crustal systems, redistributing nutrients as well as contaminants. Rock matrix diffusion into fractures (void space) within crystalline rock has been postulated to play an important role in the transient storage of solutes. The reacted volume of host rock involved, however, will be controlled by fluid-rock reactions. Here we present the results of a study which focusses on defining the length scale over which rock matrix diffusion operates within crystalline rock over timescales that are relevant to safety assessment of radioactive and other long-lived wastes. Through detailed chemical and structural analysis of natural specimens sampled at depth from an active system (Toki Granite, Japan), we show that, contrary to commonly proposed models, the length scale of rock matrix diffusion may be extremely small, on the order of centimetres, even over timescales of millions of years. This implies that in many cases the importance of rock matrix diffusion will be minimal. Additional analyses of a contrasting crystalline rock system (Carnmenellis Granite, UK) corroborate these results

    Learning and Matching Multi-View Descriptors for Registration of Point Clouds

    Critical to the registration of point clouds is the establishment of a set of accurate correspondences between points in 3D space. The correspondence problem is generally addressed by the design of discriminative 3D local descriptors on the one hand, and the development of robust matching strategies on the other hand. In this work, we first propose a multi-view local descriptor, which is learned from the images of multiple views, for the description of 3D keypoints. Then, we develop a robust matching approach, aiming at rejecting outlier matches based on the efficient inference via belief propagation on the defined graphical model. We have demonstrated the boost of our approaches to registration on the public scanning and multi-view stereo datasets. The superior performance has been verified by the intensive comparisons against a variety of descriptors and matching methods

    Loosely distinctive features for robust surface alignment

    Many successful feature detectors and descriptors exist for 2D intensity images. However, obtaining the same effectiveness in the domain of 3D objects has proven to be a more elusive goal. In fact, the smoothness often found in surfaces and the lack of texture information on the range images produced by conventional 3D scanners hinder both the localization of interesting points and the distinctiveness of their characterization in terms of descriptors. To overcome these limitations several approaches have been suggested, ranging from the simple enlargement of the area over which the descriptors are computed to the reliance on external texture information. In this paper we offer a change in perspective, where a game-theoretic matching technique that exploits global geometric consistency allows to obtain an extremely robust surface registration even when coupled with simple surface features exhibiting very low distinctiveness. In order to assess the performance of the whole approach we compare it with state-of-the-art alignment pipelines. Furthermore, we show that using the novel feature points with well-known alternative non-global matching techniques leads to poorer results. © 2010 Springer-Verlag

    NMR and Mossbauer study of spin dynamics and electronic structure of Fe{2+x}V{1-x}Al and Fe2VGa

    In order to assess the magnetic ordering process in Fe2VAl and the related material Fe2VGa, we have carried out nuclear magnetic resonance (NMR) and Mossbauer studies. 27Al NMR relaxation measurements covered the temperature range 4 -- 500 K in Fe(2+x)V(1-x)Al samples. We found a peak in the NMR spin-lattice relaxation rate, 27T1^-1, corresponding to the magnetic transitions in each of these samples. These peaks appear at 125 K, 17 K, and 165 K for x = 0.10, 0, and - 0.05 respectively, and we connect these features with critical slowing down of the localized antisite defects. Mossbauer measurements for Fe2VAl and Fe2VGa showed lines with no hyperfine splitting, and isomer shifts nearly identical to those of the corresponding sites in Fe3Al and Fe3Ga, respectively. We show that a model in which local band filling leads to magnetic regions in the samples, in addition to the localized antisite defects, can account for the observed magnetic ordering behavior.Comment: 5 pages, 3 figure
