    Rheological Characterization of Ethylcellulose-Based Melts for Pharmaceutical Applications

    Rheological characterization of ethylcellulose (EC)-based melts intended for the production, via micro-injection moulding (ÎĽIM), of oral capsular devices for prolonged release was carried out. Neat EC, plasticized EC and plasticized EC containing solid particles of a release modifier (filler volume content in the melt around 30%) were examined by capillary and rotational rheometry tests. Two release modifiers, differing in both chemical nature and particle geometry, were investigated. When studied by capillary rheometry, neat EC appeared at process temperatures as a highly viscous melt with a shear-thinning characteristic that progressively diminished as the apparent shear rate increased. Thus, EC as such could not successfully be processed via ÎĽIM. Plasticization, which induces changes in the material microstructure, enhanced the shear-thinning characteristic of the melt and reduced considerably its elastic properties. Marked wall slip effects were noticed in the capillary flow of the plasticized EC-based melts, with or without release modifier particles. The presence of these particles brought about an increase in viscosity, clearly highlighted by the dynamic experiments at the rotational rheometer. However, it did not impair the material processability. The thermal and rheological study undertaken would turn out a valid guideline for the development of polymeric materials based on pharma-grade polymers with potential for new pharmaceutical applications of ÎĽIM

    Effect of rootstocks and harvesting time on the nutritional quality of peel and flesh of peach fruits

    The influence was evaluated of four rootstocks (Ishtara, Mr. S 2/5, GF 677 and Barrier 1) and of harvesting time (early, middle, late) on the quality characteristics and nutritional value (vitamin C, phenols, carotenoids, total antioxidant capacity) of 'Flavorcrest' peach. The better rootstocks were Mr. S 2/5 (low-vigour) and Barrier 1 (high-vigour). In particular, Flavorcrest fruit on Mr. S 2/5 and on Barrier 1 rootstocks had higher antioxidant capacities and also higher phytochemical content, although fruits on Mr. S 2/5 were less firm. Flesh firmness was best for fruits at mid-harvest (H2, 7 July 2006), whereas phytochemical contents were best at late harvest (H3, 13 July 2006), when, for all rootstocks, the best nutritional characteristics were also recorded. Total antioxidant capacity and phytochemical content were determined for the peel and flesh. The results show that removal of peel from peach results in a significant loss of total antioxidant capacity

    Arabidopsis thaliana MYB75/PAP1 transcription factor induces anthocyanin production in transgenic tomato plants

    Tomato (Solanum lycopersicum L.) cv. Micro-Tom plants were transformed with the Arabidopsis thaliana (L.) Heyhn. MYB75/PAP1 (PRODUCTION OF ANTHOCYANIN PIGMENT 1) gene. This gene encodes for a well known transcription factor, which is involved in anthocyanin production and is modulated by light and sucrose. Transgenic tomato plants expressing AtMYB75 were characterised by a significantly higher anthocyanin production in leaves, stems, roots and flowers under normal growth conditions. Further, they also exhibited anthocyanins in fruits. Anthocyanin accumulation was not widespread but took place in specific groups of cells located in epidermal or cortical regions or in proximity of vascular bundles. In all the organs of the transgenic plants, where AtMYB75 overexpression was determined, a clear increase in the accumulation of DFR (DIHYDROFLAVONOL 4-REDUCTASE) transcript was also detected. The expression of the tomato MYB-gene ANT1 (ANTHOCYANIN1), which had previously been identified as a transcriptional endogenous regulator of anthocyanin biosynthesis, was not altered. The higher basal content of anthocyanins in the leaves of the transgenic plants could be further increased in the presence of high light conditions and contributed to mitigate photobleaching damages under high irradiance

    Intimate Partner Homicide Suicide: a Mini-Review of the Literature (2012–2018)

    Recent Findings In homicide-suicide (HS), a perpetrator kills at least one victim and then commits suicide within a time frame, which is not consistently described in the literature. Most HS happen in an intimate partner relationship (HS-IP), but data about this phenomenon are still scant and poorly systematized. Purpose of Review To assess the research papers published about HS-IP from 2012 to 2018 in Pubmed and Scopus. Article selection followed the PRISMA flow diagram. Information was extracted from the selected articles and tabulated. Summary The 22 eligible articles focusing on different types of HS, including HS-IP, suggest that HS-IPs are predominantly committed by men, usually married, cohabiting, or recently separated from their partner, with a medium-low employment status; the victim is usually the current or former female partner. Heterogenity of HS makes it difficult to generalize the results. Implications emerge for the need to target domestic violence and firearm regulation

    An Inter-Laboratory Comparative Study on the Influence of Reagents to Perform the Identification of the <i>Xylella fastidiosa</i> Subspecies Using Tetraplex Real Time PCR

    In 2022, a test performance study (TPS) assessing the influence of different master mixes on the performance of the tetraplex real-time PCR (TqPCR) assay was organized. TqPCR allows for the specific detection and identification of Xylella fastidiosa (Xf) subspecies in a single reaction. Eighteen official laboratories of the Italian National Plant Protection Organization received a panel of 12 blind samples, controls, primers, probes, and different master mixes to participate in the TPS. Furthermore, the Research Centre for Plant Protection and Certification of the Council for Agricultural Research and Economics performed an intra-laboratory study (ITS) on spiked plant matrices to evaluate the analytical sensitivity of TqPCR employing the selected master mixes with the best performance. Naturally infected samples were analyzed for subspecies identification via TqPCR compared with the official multilocus-sequence-typing (MLST) method. The best results in this comparative study were obtained using Fast Universal PCR Master Mix (Applied Biosystems) and Brilliant multiplex QPCR Master Mix (Agilent), and they confirmed that the TqPCR test is reliable, offering the advantage of identifying this subspecies at the same time, thus saving time and resources. The TqPCR assay is suggested among the tests to be used by laboratories performing the official diagnosis of Xf to support the activities of official monitoring
