    Transcriptomic fingerprinting of Pseudomonas putida under alternative physiological regimes

    Pseudomonas putida KT2440 is a metabolically versatile soil bacterium useful both as a model biodegradative organism and as a host of catalytic activities of biotechnological interest. In this report, we present the high-resolution transcriptome of P. putida cultured on different carbon sources as revealed by deep sequencing of the corresponding RNA pools. Examination of the data from growth on substrates that are processed through distinct pathways (glucose, fructose, succinate and glycerol) revealed that > 20% of the P. putida genome is differentially expressed depending on the ensuing physiological regime. Changes affected not only metabolic genes but also a suite of global regulators e.g. the rpoS sigma subunit of RNAP, various cold-shock proteins and the three HU histone-like proteins. Specifically, the genes encoding HU subunit variants hupA, hupB and hupN drastically altered their expression levels (and thus their ability to form heterodimeric combinations) under the diverse growth conditions. Furthermore, we found that two small RNAs, crcZ and crcY, known to inhibit the Crc protein that mediates catabolite repression in P. putida, were both down-regulated by glucose. In fact, a heavy stress concentration in the radial component appears at the edges of the contact zone being more intense in the case of the hub. Many methods have been proposed for relieving the stress peaks.     [ES] El taller sobre Tecnologías Disruptivas de la Información y la Comunicación para la Innovación y la Transformación Digital, organizado en el ámbito del proyecto DISRUPTIVE (disruptive.usal.es) y celebrado el 12 de septiembre de 2022 en Valladolid, tiene como objetivo debatir sobre los problemas, retos y beneficios del uso de tecnologías digitales disruptivas, a saber, Internet de las Cosas, Big data, computación en la nube, sistemas multiagente, aprendizaje automático, realidad virtual y aumentada y robótica colaborativa, para apoyar la transformación digital en curso en la sociedad. El programa del taller incluyó 6 papers técnicos aceptados, 2 charlas de invitados y una sesión de networking. Este volumen contiene 6 de las ponencias presentadas en el taller sobre Tecnologías Disruptivas de la Información y la Comunicación para la Innovación y la Transformación Digital.     Prognostic implications of troponin T variations in inherited cardiomyopathies using systems biology.

    The cardiac troponin T variations have often been used as an example of the application of clinical genotyping for prognostication and risk stratification measures for the management of patients with a family history of sudden cardiac death or familial cardiomyopathy. Given the disparity in patient outcomes and therapy options, we investigated the impact of variations on the intermolecular interactions across the thin filament complex as an example of an unbiased systems biology method to better define clinical prognosis to aid future management options. We present a novel unbiased dynamic model to define and analyse the functional, structural and physico-chemical consequences of genetic variations among the troponins. This was subsequently integrated with clinical data from accessible global multi-centre systematic reviews of familial cardiomyopathy cases from 106 articles of the literature: 136 disease-causing variations pertaining to 981 global clinical cases. Troponin T variations showed distinct pathogenic hotspots for dilated and hypertrophic cardiomyopathies; considering the causes of cardiovascular death separately, there was a worse survival in terms of sudden cardiac death for patients with a variation at regions 90-129 and 130-179 when compared to amino acids 1-89 and 200-288. Our data support variations among 90-130 as being a hotspot for sudden cardiac death and the region 131-179 for heart failure death/transplantation outcomes wherein the most common phenotype was dilated cardiomyopathy. Survival analysis into regions of high risk (regions 90-129 and 130-180) and low risk (regions 1-89 and 200-288) was significant for sudden cardiac death (p = 0.011) and for heart failure death/transplant (p = 0.028). Our integrative genomic, structural, model from genotype to clinical data integration has implications for enhancing clinical genomics methodologies to improve risk stratification

    Characterization of the distribution of water vapour for DInSAR studies on the volcanic island of Tenerife, Canary Islands

    Measurements of ground displacement have been carried out on the entire active volcanic island of Tenerife, Canary Islands, by means of classical and advanced DInSAR techniques. The main limiting factor on the accuracy of DInSAR measurements is the distribution of the water vapour in the lower troposphere. Hence, it is yet necessary to perform a detailed spatial and temporal characterization of water vapour to understand, and to be able to carry out a direct computation of, the effect of the tropospheric delay on DInSAR results. In this sense, satellite and balloon data have been analysed to infer the variability in the distribution of water vapour and hence, the robustness of DInSAR results on the island of Tenerife.Peer reviewe

    Physicochemical characterization of Crithmum maritimum L. and Daucus carota subsp. gummifer (Syme) Hook.fil. and their antimicrobial activity against apple tree and grapevine phytopathogens

    Crithmum maritimum and Daucus carota subsp. gummifer are two species of the Apiaceae family that share multiple characteristics: both are halophitic, live on cliffs in the same geographic habitats, and are edible. While C. maritimum is rich in essential oils and flavonoids, D. carota is rich in terpenes and a gum producer. In the work presented herein, the biomass of these two wild plants and the bioactive compounds present in their extracts have been studied by elemental and thermal analysis, infrared spectroscopy, and gas chromatography-mass spectroscopy. To explore their bioactivities, both their hydroalcoholic extracts and their major constituents (apiole in C. maritimum and geranyl acetate in D. carota), either alone or in combination with chitosan oligomers, were assayed in vitro against bacterial and fungal pathogens that affect apple trees (Malus domestica) and grapevine (Vitis vinifera). Remarkable inhibition was observed against Erwinia amylovora, the causal agent of fire blight in apple; Xylophilus ampelinus [syn. Erwinia vitivora], the causal agent of bacterial blight of grapevine; and Diplodia seriata, a virulent pathogen of grapevines that also causes canker, leaf spot and fruit rot of apple. In view of their effectiveness against these three phytopathogens, a potential application of these two medicinal plants in organic farming may be envisaged

    Self-consistent treatment of thermal effects in neutron-star post-mergers: observational implications for third-generation gravitational-wave detectors

    We assess the impact of accurate, self-consistent modelling of thermal effects in neutron-star merger remnants in the context of third-generation gravitational-wave detectors. This is done through the usage, in Bayesian model selection experiments, of numerical-relativity simulations of binary neutron star (BNS) mergers modelled through: a) nuclear, finite-temperature (or ``tabulated'') equations of state (EoSs), and b) their simplifed piecewise (or ``hybrid'') representation. These cover four different EoSs, namely SLy4, DD2, HShen and LS220. Our analyses make direct use of the Newman-Penrose scalar ψ4\psi_4 outputted by numerical simulations. Considering a detector network formed by three Cosmic Explorers, we show that differences in the gravitational-wave emission predicted by the two models are detectable with a natural logarithmic Bayes Factor logB5\log{\cal{B}}\geq 5 at average distances of dL50d_L \simeq 50Mpc, reaching dL100d_L \simeq 100Mpc for source inclinations ι0.8\iota \leq 0.8, regardless of the EoS. This impact is most pronounced for the HShen EoS. For low inclinations, only the DD2 EoS prevents the detectability of such modelling differences at dL150d_L \simeq 150Mpc. Our results suggest that the usage a self-consistent treatment of thermal effects is crucial for third-generation gravitational wave detectors.Comment: 9 pages, 3 Figure
