Biology of the red mullet mullus surmuletus (mullidae) off the Canary Islands, Central-East Atlantic
The biology of red mullet Mullus surmuletus was studied from collections taken off the Canary Islandsbetween January 1991 and September 1993. Total length ranged from 12 to 33 cm, mainly between 15 and 21cm. Males ranged from 14 to 26 cm and females from 14 to 33 cm. Females dominated the larger size-classes (>18 cm). The overall ratio of males to females was 1:2.3. The reproductive period extended from February toMay, spawning peaking in March and April. The total length at 50% maturity was 16.6 cm for the whole population.The length-mass relationship for all individuals can be described by the parameters a = 0.0074 and b = 3.1826. Fish aged 0.8 years were present in the samples. The parameters of the Von Bertalanffy growth equation obtained for all individuals were: L‡ = 35.71 cm and k = 0.22.year-1. Significant differences were found in the growth parameters between males and females. The rates of total mortality Z, natural mortality M and fishing mortality F were 1.25, 0.55 and 0.70.year-1 respectively. The estimated total length at first capture was 15.74 cm
Particle Production near an AdS Crunch
We numerically study the dual field theory evolution of five-dimensional
asymptotically anti-de Sitter solutions of supergravity that develop
cosmological singularities. The dual theory is an unstable deformation of the N
= 4 gauge theory on R S3, and the big crunch singularity in the bulk
occurs when a boundary scalar field runs to infinity. Consistent quantum
evolution requires one imposes boundary conditions at infinity. Modeling these
by a steep regularization of the scalar potential, we find that when an
initially nearly homogeneous wavepacket rolls down the potential, most of the
potential energy of the initial configuration is converted into gradient energy
during the first oscillation of the field. This indicates there is no
transition from a big crunch to a big bang in the bulk for dual boundary

A GFP-lacZ Bicistronic Reporter System for Promoter Analysis in Environmental Gram-Negative Bacteria
Here, we describe a bicistronic reporter system for the analysis of promoter activity in a variety of Gram-negative bacteria at both the population and single-cell levels. This synthetic genetic tool utilizes an artificial operon comprising the gfp and lacZ genes that are assembled in a suicide vector, which is integrated at specific sites within the chromosome of the target bacterium, thereby creating a monocopy reporter system. This tool was instrumental for the complete in vivo characterization of two promoters, Pb and Pc, that drive the expression of the benzoate and catechol degradation pathways, respectively, of the soil bacterium Pseudomonas putida KT2440. The parameterization of these promoters in a population (using β-galactosidase assays) and in single cells (using flow cytometry) was necessary to examine the basic numerical features of these systems, such as the basal and maximal levels and the induction kinetics in response to an inducer (benzoate). Remarkably, GFP afforded a view of the process at a much higher resolution compared with standard lacZ tests; changes in fluorescence faithfully reflected variations in the transcriptional regimes of individual bacteria. The broad host range of the vector/reporter platform is an asset for the characterization of promoters in different bacteria, thereby expanding the diversity of genomic chasses amenable to Synthetic Biology methods
Poloxomer 188 Has a Deleterious Effect on Dystrophic Skeletal Muscle Function
Duchenne muscular dystrophy (DMD) is an X-linked, fatal muscle wasting disease for which there is currently no cure and limited palliative treatments. Poloxomer 188 (P188) is a tri-block copolymer that has been proposed as a potential treatment for cardiomyopathy in DMD patients. Despite the reported beneficial effects of P188 on dystrophic cardiac muscle function, the effects of P188 on dystrophic skeletal muscle function are relatively unknown. Mdx mice were injected intraperitoneally with 460 mg/kg or 30 mg/kg P188 dissolved in saline, or saline alone (control). The effect of single-dose and 2-week daily treatment was assessed using a muscle function test on the Tibialis Anterior (TA) muscle in situ in anaesthetised mice. The test comprises a warm up, measurement of the force-frequency relationship and a series of eccentric contractions with a 10% stretch that have previously been shown to cause a drop in maximum force in mdx mice. After 2 weeks of P188 treatment at either 30 or 460 mg/kg/day the drop in maximum force produced following eccentric contractions was significantly greater than that seen in saline treated control mice (P = 0.0001). Two week P188 treatment at either dose did not significantly change the force-frequency relationship or maximum isometric specific force produced by the TA muscle. In conclusion P188 treatment increases susceptibility to contraction-induced injury following eccentric contractions in dystrophic skeletal muscle and hence its suitability as a potential therapeutic for DMD should be reconsidered
Simulation of an SEIR infectious disease model on the dynamic contact network of conference attendees
The spread of infectious diseases crucially depends on the pattern of
contacts among individuals. Knowledge of these patterns is thus essential to
inform models and computational efforts. Few empirical studies are however
available that provide estimates of the number and duration of contacts among
social groups. Moreover, their space and time resolution are limited, so that
data is not explicit at the person-to-person level, and the dynamical aspect of
the contacts is disregarded. Here, we want to assess the role of data-driven
dynamic contact patterns among individuals, and in particular of their temporal
aspects, in shaping the spread of a simulated epidemic in the population.
We consider high resolution data of face-to-face interactions between the
attendees of a conference, obtained from the deployment of an infrastructure
based on Radio Frequency Identification (RFID) devices that assess mutual
face-to-face proximity. The spread of epidemics along these interactions is
simulated through an SEIR model, using both the dynamical network of contacts
defined by the collected data, and two aggregated versions of such network, in
order to assess the role of the data temporal aspects.
We show that, on the timescales considered, an aggregated network taking into
account the daily duration of contacts is a good approximation to the full
resolution network, whereas a homogeneous representation which retains only the
topology of the contact network fails in reproducing the size of the epidemic.
These results have important implications in understanding the level of
detail needed to correctly inform computational models for the study and
management of real epidemics
FATTY ACID COMPOSITION OF LAMB MEAT FROM ITALIAN AND GERMAN LOCAL BREEDS
The aim of this study was to evaluate and compare the quality characteristics, chemical
composition and lipid profile of lamb meat from Italian (Biellese and Sambucana) and
German (Texel-Merino-Blackhead-Charollais [TMBC]) breeds reared in extensive and
semi-extensive production systems. Meat samples from 89 animals were analysed.
The meat of the lambs from semi-intensively reared Biellese, and extensively reared
Sambucana and TMBC breeds produced lean meat, with slightly higher intramuscular
fat content in TMBC. The latter also produced meat of darker colour (P<0.05) and
higher protein content (P<0.05). The meat of Sambucana lambs presented the lowest
total cholesterol content (P<0.05). The fatty acid profile of the meat showed a clear
advantage of both extensively reared breeds, which had substantially lower proportion
of saturated (SFA) but higher of polyunsaturated fatty acids (PUFA), particularly n-3
(P<0.05). The beneficial effect of the extensive rearing conditions was associated with
lower n-6/n3 ratio, and atherogenic (AI) and thrombogenic (TI) indices, thereby
suggesting that production system can be used successfully to modify the fatty acid
profile to achieve a positive effect for the human health
Associations Between Methylation of Paternally Expressed Gene 3 (PEG3), Cervical Intraepithelial Neoplasia and Invasive Cervical Cancer.
Cytology-based screening for invasive cervical cancer (ICC) lacks sensitivity and specificity to discriminate between cervical intraepithelial neoplasia (CIN) likely to persist or progress from cases likely to resolve. Genome-wide approaches have been used to identify DNA methylation marks associated with CIN persistence or progression. However, associations between DNA methylation marks and CIN or ICC remain weak and inconsistent. Between 2008-2009, we conducted a hospital-based, case-control study among 213 Tanzania women with CIN 1/2/3 or ICC. We collected questionnaire data, biopsies, peripheral blood, cervical scrapes, Human papillomavirus (HPV) and HIV-1 infection status. We assessed PEG3 methylation status by bisulfite pyrosequencing. Multinomial logistic regression was used to estimate odds ratios (OR) and confidence intervals (CI 95%) for associations between PEG3 methylation status and CIN or ICC. After adjusting for age, gravidity, hormonal contraceptive use and HPV infection, a 5% increase in PEG3 DNA methylation was associated with increased risk for ICC (OR = 1.6; 95% CI 1.2-2.1). HPV infection was associated with a higher risk of CIN1-3 (OR = 15.7; 95% CI 5.7-48.6) and ICC (OR = 29.5, 95% CI 6.3-38.4). Infection with high risk HPV was correlated with mean PEG3 differentially methylated regions (DMRs) methylation (r = 0.34 p<0.0001), while the correlation with low risk HPV infection was weaker (r = 0.16 p = 0.047). Although small sample size limits inference, these data support that PEG3 methylation status has potential as a molecular target for inclusion in CIN screening to improve prediction of progression. Impact statement: We present the first evidence that aberrant methylation of the PEG3 DMR is an important co-factor in the development of Invasive cervical carcinoma (ICC), especially among women infected with high risk HPV. Our results show that a five percent increase in DNA methylation of PEG3 is associated with a 1.6-fold increase ICC risk. Suggesting PEG3 methylation status may be useful as a molecular marker for CIN screening to improve prediction of cases likely to progress
