    Vascular Smooth Muscle Cell-Specific Progerin Expression Provokes Contractile Impairment in a Mouse Model of Hutchinson-Gilford Progeria Syndrome that Is Ameliorated by Nitrite Treatment

    Cardiovascular disease (CVD) is the main cause of death worldwide, and aging is its leading risk factor. Aging is much accelerated in Hutchinson-Gilford progeria syndrome (HGPS), an ultra-rare genetic disorder provoked by the ubiquitous expression of a mutant protein called progerin. HGPS patients die in their teens, primarily due to cardiovascular complications. The primary causes of age-associated CVD are endothelial dysfunction and dysregulated vascular tone; however, their contribution to progerin-induced CVD remains poorly characterized. In the present study, we found that progeroid LmnaG609G/G609G mice with ubiquitous progerin expression show both endothelial dysfunction and severe contractile impairment. To assess the relative contribution of specific vascular cell types to these anomalies, we examined LmnaLCS/LCSTie2Cretg/+ and LmnaLCS/LCSSm22αCretg/+ mice, which express progerin specifically in endothelial cells (ECs) and vascular smooth muscle cells (VSMCs), respectively. Whereas vessel contraction was impaired in mice with VSMC-specific progerin expression, we observed no endothelial dysfunction in mice with progerin expression restricted to VSMCs or ECs. Vascular tone regulation in progeroid mice was ameliorated by dietary sodium nitrite supplementation.     The psychological impact of Covid-19 on front line nurses: a synthesis of qualitative evidence

    Caring for people with COVID-19 on the front line has psychological impacts for healthcare professionals. Despite the important psychological impacts of the pandemic on nurses, the qualitative evidence on this topic has not been synthesized. Our objective: To analyze and synthesize qualitative studies that investigate the perceptions of nurses about the psychological impacts of treating hospitalized people with COVID-19 on the front line. A systematic review of qualitative studies published in English or Spanish up to March 2021 was carried out in the following databases: The Cochrane Library, Medline (Pubmed), PsycINFO, Web of Science (WOS), Scopus, and CINHAL. The PRISMA statement and the Cochrane recommendations for qualitative evidence synthesis were followed. Results: The main psychological impacts of caring for people with COVID-19 perceived by nurses working on the front line were fear, anxiety, stress, social isolation, depressive symptoms, uncertainty, and frustration. The fear of infecting family members or being infected was the main repercussion perceived by the nurses. Other negative impacts that this review added and that nurses suffer as the COVID-19 pandemic progress were anger, obsessive thoughts, compulsivity, introversion, apprehension, impotence, alteration of space-time perception, somatization, and feeling of betrayal. Resilience was a coping tool used by nurses. Conclusions: Front line care for people with COVID-19 causes fear, anxiety, stress, social isolation, depressive symptoms, uncertainty, frustration, anger, obsessive thoughts, compulsivity, introversion, apprehension, impotence, alteration of space-time perception, somatization, and feeling of betrayal in nurses. It is necessary to provide front line nurses with the necessary support to reduce the psychological impact derived from caring for people with COVID-19, improve training programs for future pandemics, and analyze the long-term impacts.info:eu-repo/semantics/publishedVersio

    The results of different labour induction approaches: A Cross sectional study

    Background and Purpose: To evaluate the use of prostaglandins and oxytocin in labour induction according to different indications. Perinatal outcomes, rate of vaginal delivery and complation of labour were studied and compared. Methods: Cross-sectional descriptive study from January 2012 to December 2012. 530 women who required labour induction were included. Seven groups were created according to the methods of induction. Women with twin pregnancies, induction of dead foetus, two previous caesarean sections or an incomplete clinical history were excluded. Results: The rate of vaginal deliveries in women that only received prostaglandins the first day was 84.6%; similar in women with prolonged pregnancies, 85.2%. The induction with oxytocin directly showed the highest rate of caesarean section. The rate of vaginal deliveries was 50% in women with previous caesarean section. Conclusions: A high rate of vaginal deliveries with a single dose of prostaglandin and within 24 hours of beginning induction. Administration of prostaglandins must be used when cervix is unfavorable and previous to oxytocin stymulation

    Información climática asociada a estaciones productivas para el ajuste de modelos estadísticos de sistemas bovinos bajo condiciones extensivas

    Artículo de investigación publicado en revista internacional indizada.El objeto de este estudio fue desarrollar una metodología para clasificar épocas de nacimiento (EN) y evaluar su impacto en indicadores productivos al compararlas con un método tradicional de clasificación. Con información meteorológica, se generó un índice de aridez para clasificar las EN. Las EN propuesta y tradicional se compararon, incluyéndolas como grupo contemporáneo (GC= hato, sexo, año y EN) a modelos de evaluación genética de caracteres de peso vivo. Fueron estimados los componentes de varianza y valores genéticos (DEP) con sus exactitudes. La clasifica- ción propuesta explicó mayor variabilidad que la clasificación tradicional (≥9,8%). Los parámetros genéticos mostraron cambios importantes, siendo los más evidentes en peso al destete. Los modelos fueron diferentes de acuerdo a la prueba de razón de verosimilitudes (P<0,01). Se observó una mejora de estructura en los GC. Para los dos caracteres evaluados las correlaciones entre los valores genéticos de los animales fueron diferentes, lo que indica cambio en la jerarquización.     U1A is a positive regulator of the expression of heterologous and cellular genes involved in cell proliferation and migration

    Here, we show that direct recruitment of U1A to target transcripts can increase gene expression. This is a new regulatory role, in addition to previous knowledge showing that U1A decreases the levels of U1A mRNA and other specific targets. In fact, genome-wide, U1A more often increases rather than represses gene expression and many U1A-upregulated transcripts are directly bound by U1A according to individual nucleotide resolution crosslinking and immunoprecipitation (iCLIP) studies. Interestingly, U1A-mediated positive regulation can be transferred to a heterologous system for biotechnological purposes. Finally, U1A-bound genes are enriched for those involved in cell cycle and adhesion. In agreement with this, higher U1A mRNA expression associates with lower disease-free survival and overall survival in many cancer types, and U1A mRNA levels positively correlate with those of some oncogenes involved in cell proliferation. Accordingly, U1A depletion leads to decreased expression of these genes and the migration-related gene CCN2/CTGF, which shows the strongest regulation by U1A. A decrease in U1A causes a strong drop in CCN2 expression and CTGF secretion and defects in the expression of CTGF EMT targets, cell migration, and proliferation. These results support U1A as a putative therapeutic target for cancer treatment. In addition, U1A-binding sequences should be considered in biotechnological applications

    Randomized Clinical Trials of obesity treatments in Mexican population. Systematic Review and Meta-Analysis

    Background: Mexicans and Mexican Americans share similar culture, genetic background, and predisposition for obesity and diabetes. Randomized clinical trials (RCT) assessing obesity treatments (ObT) are reliable to assess efficacy. To date, there is no systematic review to investigate ObT tested by RCT in Mexican adults. Methods: We conducted systematic searches in Pubmed, Scopus, and Web of Science to retrieve ObT RCT from 1990 to 2019. The ObT included alternative medicine, pharmacological, nutritional, behavioral, and surgical interventions. The analyzed RCT were at least three months of duration, and reported: BMI, weight, waist circumference, triglycerides, glucose and blood pressure. Results: We found 634 entries; after removal of duplicates and exclusions based on eligibility criteria, we analyzed 43 and 2 multinational-collaborative studies. Most of the national studies had small sample sizes, and did not have replications from other studies. The nutrition/behavioral interventions were difficult to blind, and most studies had medium to high risk of bias. Random effects meta-analysis of nutritional/behavioral interventions and medications showed effects on BMI, waist circumference, and blood pressure. Simple measures like plain water instead of sweet beverages decreased triglycerides and systolic blood pressure. Participants with obesity and hypertension had beneficial effects with antioxidants, and the treatment with insulin increased weight in those with T2D. Conclusions: The RCT’s in Mexico reported effects on metabolic components despite small sample sizes and lack of replication. In the future we should analyze ObT in population living on the U.S.-Mexico border; therefore, bi-national collaboration is desirable to disentangle cultural effects on ObT response

    Épocas de nacimiento basadas en un índice climático para el ajuste de modelos estadísticos para peso vivo de ganado bovino en México

    Artículo de investigación científica publicado en revista indizada.El objetivo fue comparar los efectos de una metodología para la clasificación de épocas climáticas, que tradicionalmente son utilizadas para establecer épocas de nacimiento (EN) en estudios y experimentos estadísticos. Esta metodología se basa en un índice de aridez (IA) para clasificar las EN utilizando información meteorológica histórica. Se trabajó una base de datos con un pedigrí de 7,460 animales, se ajustaron dos modelos para peso al nacimiento y peso al destete en bovinos Charolais manejados en pastoreo extensivo. Los modelos incluyeron el efecto fijo de grupo contemporáneo (GC= subclase de hato, sexo, año y EN) y edad de la madre. Los mismos modelos se compararon utilizando una clasificación tradicional de estaciones del año para clasificar las EN. Al estimar los componentes de varianza y valores genéticos (DEP) con sus exactitudes, los modelos fueron diferentes de acuerdo a la prueba de razón de verosimilitudes (P<0.01). El número de GC se redujo en 25 % para la EN basada en el IA, con GC con mayor número de individuos. El principal efecto observado en los modelos analizados fue el cambio en la jerarquización de las DEP para ambas características. Este método de clasificación de épocas de nacimiento, puede ayudar a mejorar el ajuste de modelos estadísticos en los sistemas ganaderos manejados en pastoreo extensivo

    Iron bioaccessibility and sensory analysis of extruded cereals fortified with different Fe sources

    To increase iron (Fe) intake in Fe deficiency-risk groups the combination of Fe source and food-vehicle must be chosen in order to minimize inhibitory effects of food matrix. Fe dialyzability and sensory properties were tested in six model systems (MS) made with extruded cereals fortified with different Fe sources such as FeNaEDTA, FeSO4 and EDTA/FeSO4 among others and with or without the addition of milk. Proximate composition and phytate content were also evaluated. Results showed that Fe dialyzability from samples fortified with FeNaEDTA was less affected by the presence of inhibitory factors such as phytates and milk. The addition of FeSO4 to the extrudates showed sensory differences. Furthermore, fortification with EDTA/FeSO4 or FeNaEDTA showed no sensory differences compared with unfortified or Feº (elemental iron) fortified matrix, with the advantage of increased iron bioaccessibility.Fil: Cagnasso, Carolina Elisa.
