    Peers, pirates, and persuasion : rhetoric in the peer-to-peer debates

While broadband Internet services now allow speedy transfers of complex media files, Americans face real uncertainty about whether peer-to-peer file sharing is or should be legal. John Logie analyzes the public arguments growing out of more than five years of debate sparked by the advent of Napster, the first widely adopted peer-to-peer technology. The debate continues with the second wave of peer-to-peer file transfer utilities like Limewire, KaZaA, and BitTorrent. With Peers, Pirates, and Persuasion, Logie joins the likes of Lawrence Lessig, Siva Vaidhyanathan, Jessica Litman, and James Boyle in the ongoing effort to challenge and change current copyright law so that it fulfills its purpose of fostering creativity and innovation while protecting the rights of artists in an attention economy. Logie examines metaphoric frames—warfare, theft, piracy, sharing, and hacking, for example—that dominate the peer-to-peer debates and demonstrably shape public policy on the use and exchange of digital media. Peers, Pirates, and Persuasion identifies the Napster case as a failed opportunity for a productive national discussion on intellectual property rights and responsibilities in digital environments. Logie closes by examining the U.S. Supreme Court’s ruling in the “Grokster” case, in which leading peer-to-peer companies were found to be actively inducing copyright infringement.     Mechanisms of glucocorticoid-mediated inhibition of angiogenesis

    Angiogenesis, the formation of new blood vessels from pre-existing vascular beds, is a requirement of embryonic development but only occurs in a limited number of discrete processes in the healthy adult. In contrast, abnormal angiogenesis is central to many pathological processes including: tumour growth, diabetic retinopathy and arthritis. Consequently, pharmacological manipulation of angiogenesis has great clinical potential.Angiogenesis is inhibited by glucocorticoids and this is exploited clinically for the treatment of proliferating capillary haemangiomas in children. Despite this, the exact mechanism(s) through which glucocorticoids inhibit angiogenesis is (are) unknown. Whilst glucocorticoids can act directly on the vessel wall their effects on individual cell types and on molecular signalling remain unclear. The work in this thesis explores the hypothesis that glucocorticoid-mediated inhibition of angiogenesis is the result of direct modulation of growth factor signalling within the vascular endothelial cells.A well-characterised 2-dimensional in vitro model of human endothelial tube formation was introduced. Glucocorticoids were shown to inhibit tube formation in this model via stimulation of glucocorticoid receptors and this process was not influenced by intra-cellular glucocorticoid metabolism by ll(3-hydroxysteroid dehydrogenases. This demonstration that glucocorticoids inhibit angiogenesis by acting directly on the endothelium is consistent with, and extends, observations of glucocorticoid-mediated angiostasis in rodent aortic rings and during cutaneous wound healing. Molecular and biochemical assays suggested that glucocorticoids inhibit tube formation by altering the balance of pro- and anti-angiogenic factor activity. Time-lapse imaging of tube formation, combined with assays of endothelial cell migration and proliferation, indicated that glucocorticoids reduce tube formation, rather than accelerating degradation of existing tubes, by preventing morphological changes in the cells but do not inhibit cell division or migration.In conclusion, these studies demonstrate that glucocorticoids can inhibit angiogenesis by directly inhibiting morphological changes required for tube formation by endothelial cells but without altering migration or proliferation

    Population ecology and lifetime reproductive success of dippers Cinclus cinclus

    Acidified catchments are known to hold significantly reduced dipper Cinclus cinclus populations throughout the year relative to circum-neutral rivers, although the processes leading to these declines remain unclear. This study considered the population ecology of dippers within the circumneutral River Devon catchment, Central Scotland, and focused primarily on determining the factors influencing survival, breeding probabilities and reproductive success. It aimed to examine the role of spatial variation in 'habitat quality' on the population (and meta-population) dynamics of dippers, based on measures of seasonal and lifetime reproductive success, and the balance between survival and reproduction; in particular, to assess if the reduced reproductive success of dippers on acid rivers is likely to lead to population declines. Within the Devon catchment, approximately 81% of all adults survived from spring (March/April) to autumn (September/October), with 65% of these birds surviving from autumn to the following breeding season. Overall, these estimates predicted annual adult survival rates of c.53%, with no significant differences between years. Population density had no detectable effect on adult mortality rates, although juvenile over-winter survival was significantly lower than the adult rate at between 40 and 58%, and negatively related to the total size of the autumn population. There was no evidence of sex differences in juvenile over-winter survival, or any significant influence of weather or river flows on the rates for adults or juveniles. The local post-fledging survival of females was significantly lower than for males, however, apparently reflecting sex differences in post-natal dispersal. On average, less than 6.5% of all eggs laid, or 10.4-14.5% of male and 6.3-9.2% of female fledglings raised within the Devon catchment survived locally to breeding age. Juvenile, although not adult, recapture rates in spring were significantly lower than for birds known to have bred previously and negatively related to spring river flows. This suggested that with recapture dependent on a breeding attempt that was successful at least until laying, either more first year birds failed during the initial stages of nesting or that full breeding was not achieved at age one. The birds fledging the most young, both within a season and over a lifetime, all bred at 'prime' lowland sites characterised by wide, shallow rivers of intermediate gradient, although with less than 10% of all birds attempting to raise a second brood each year, no significant habitat differences were identified in any component of reproductive output measured until fledging. River width, altitude and gradient were all significantly inter-correlated and related to laying date, however, and post-fledging survival was significantly reduced for late fledged young. On average, lowland birds laid earlier than upland breeders, and were significantly more likely to produce autumn 'recruits' due to the enhanced post-fledging survival prospects of their young. This suggested that broad measures of river structure can provide a biologically appropriate classification of habitat quality. The size of the breeding population of dippers within the Devon catchment appeared to be related to the availability of critical resources, most likely food, roost sites and ultimately breeding territories through density-dependent changes in over-winter mortality and recruitment. The relative importance of resource abundance and recruitment levels in determining autumn population densities on acid streams still remained unclear, although reference to published relationships between acidity and reproductive success suggested that with adult survival at the rate estimated for the Devon catchment, many dipper populations are unlikely to produce sufficient recruits to match all adult losses, and may only persist with continued immigration from more productive (circumneutral) catchments elsewhere

    Limits of the cylindrical absorber design for a sodium receiver

    The applicability of the cylindrical arrangement of vertical tube banks is evaluated for liquid sodium concentrating solar thermal receivers and compared with a molten salt reference case through a series of parametric studies. It is shown that sodium receivers experience less thermo-elastic stress load and can operate under higher flux which presents advantages in terms of size reduction and efficiency. While the cylindrical receiver configuration cannot reach the efficiency target of 91% in a high temperature configuration (480 °C to 640 °C), there is potential to improve efficiency by improving heliostat field optics. Flux limitations due to thermo-elastic stresses are less stringent due for sodium receivers due to the better heat transfer properties, and consequently better heliostat field optics would benefit sodium receiver concepts more than molten salts ones

However current evidence relies on either longitudinal studies, which are complex and resource-intensive, or linked data studies based on social services data, which is arguably the tip of the iceberg in terms of children who are maltreated. Reliable, linked, population-level data on children referred to services due to suspected abuse or neglect will increase our ability to examine risk factors for, and outcomes following, abuse and neglect.Objective: The objective of this project was to create a linkable population level dataset, The Edinburgh Child Protection Dataset (ECPD), comprising all children referred to the Edinburgh Child Protection Paediatric healthcare team due to a concern about their welfare between 1995 and 2015.Methods: The paper presents the process for creating the dataset. The analyses provide examples of available data from the main referrals dataset between 1995 and 2011 (where data quality was highest).Results: 19,969 referrals were captured, relating to 11,653 children. Of the 19,969 referrals, a higher proportion were girls (54%), although boys were referred for physical abuse more often than girls (41% versus 30%). Younger children were more likely to be referred for physical abuse (35% of 0-4 year olds vs. 27% 15+): older children were more likely to be referred for sexual abuse (48% of 15+ years vs. 18% of 0-4 years). Most referrals came from social workers (46%) or police (31%)).Conclusions: The ECPD offers a unique insight into the characteristics of referrals to child protection paediatric services over a key period in the history of child protection in Scotland. It is hoped that by making these data available to researchers, and able to be easily linked with both mother and child current and future health records, evidence will be created to better support maltreated children and monitor changes over time

    Multiple Aspects of ATP-Dependent Nucleosome Translocation by RSC and Mi-2 Are Directed by the Underlying DNA Sequence

    Contains fulltext : 129351.pdf (publisher's version ) (Open Access)Background Chromosome structure, DNA metabolic processes and cell type identity can all be affected by changing the positions of nucleosomes along chromosomal DNA, a reaction that is catalysed by SNF2-type ATP-driven chromatin remodelers. Recently it was suggested that in vivo, more than 50% of the nucleosome positions can be predicted simply by DNA sequence, especially within promoter regions. This seemingly contrasts with remodeler induced nucleosome mobility. The ability of remodeling enzymes to mobilise nucleosomes over short DNA distances is well documented. However, the nucleosome translocation processivity along DNA remains elusive. Furthermore, it is unknown what determines the initial direction of movement and how new nucleosome positions are adopted. Methodology/Principal Findings We have used AFM imaging and high resolution PAGE of mononucleosomes on 600 and 2500 bp DNA molecules to analyze ATP-dependent nucleosome repositioning by native and recombinant SNF2-type enzymes. We report that the underlying DNA sequence can control the initial direction of translocation, translocation distance, as well as the new positions adopted by nucleosomes upon enzymatic mobilization. Within a strong nucleosomal positioning sequence both recombinant Drosophila Mi-2 (CHD-type) and native RSC from yeast (SWI/SNF-type) repositioned the nucleosome at 10 bp intervals, which are intrinsic to the positioning sequence. Furthermore, RSC-catalyzed nucleosome translocation was noticeably more efficient when beyond the influence of this sequence. Interestingly, under limiting ATP conditions RSC preferred to position the nucleosome with 20 bp intervals within the positioning sequence, suggesting that native RSC preferentially translocates nucleosomes with 15 to 25 bp DNA steps. Conclusions/Significance Nucleosome repositioning thus appears to be influenced by both remodeler intrinsic and DNA sequence specific properties that interplay to define ATPase-catalyzed repositioning. Here we propose a successive three-step framework consisting of initiation, translocation and release steps to describe SNF2-type enzyme mediated nucleosome translocation along DNA. This conceptual framework helps resolve the apparent paradox between the high abundance of ATP-dependent remodelers per nucleus and the relative success of sequence-based predictions of nucleosome positioning in vivo

    Experimental Testing of a High-Flux Cavity Receiver

    A new tubular cavity receiver for direct steam generation, ‘SG4’, has been built and tested on-sun based on integrated optical and thermal modelling. The new receiver achieved an average thermal efficiency of 97.1±2.1% across several hours of testing, and reduced the losses by more than half, compared to the modelled performance of the previous SG3 receiver and dish.
