Increased gene copy number of DEFA1/DEFA3 worsens sepsis by inducing endothelial pyroptosis.
Sepsis claims an estimated 30 million episodes and 6 million deaths per year, and treatment options are rather limited. Human neutrophil peptides 1-3 (HNP1-3) are the most abundant neutrophil granule proteins but their neutrophil content varies because of unusually extensive gene copy number polymorphism. A genetic association study found that increased copy number of the HNP-encoding gene DEFA1/DEFA3 is a risk factor for organ dysfunction during sepsis development. However, direct experimental evidence demonstrating that these risk alleles are pathogenic for sepsis is lacking because the genes are present only in some primates and humans. Here, we generate DEFA1/DEFA3 transgenic mice with neutrophil-specific expression of the peptides. We show that mice with high copy number of DEFA1/DEFA3 genes have more severe sepsis-related vital organ damage and mortality than mice with low copy number of DEFA1/DEFA3 or wild-type mice, resulting from more severe endothelial barrier dysfunction and endothelial cell pyroptosis after sepsis challenge. Mechanistically, HNP-1 induces endothelial cell pyroptosis via P2X7 receptor-mediating canonical caspase-1 activation in a NLRP3 inflammasome-dependent manner. Based on these findings, we engineered a monoclonal antibody against HNP-1 to block the interaction with P2X7 and found that the blocking antibody protected mice carrying high copy number of DEFA1/DEFA3 from lethal sepsis. We thus demonstrate that DEFA1/DEFA3 copy number variation strongly modulates sepsis development in vivo and explore a paradigm for the precision treatment of sepsis tailored by individual genetic information

The Promise of Justice: Are Public and Private Defense Equal?
This thesis attempts to uncover whether or not differences in unclassified felony and first-degree felony case outcomes arise as a result of the type of counsel representing the defendant. The types of defense counsel addressed in this project include public defenders, appointed counsel, and private counsel. This research project specifically focuses on Butler, Greene, and Montgomery Counties in Ohio, in order to cultivate large enough samples for statistical testing. The felony cases that were examined include aggravated murder, murder, rape, voluntary manslaughter, and kidnapping. Existing research (Cohen, 2014; Levine, 1975; Hartley, Miller, and Spohn, 2010; and Williams, 2002) largely shows that public defenders perform better than appointed counsel, and roughly equal to private counsel. In order to discover whether or not discrepancies between the three categories of defense counsel exist in southwestern Ohio, county-level crime and court data encompassing type of counsel, charge(s) filed, and conviction and post-conviction outcomes including guilty pleas, convictions, incarceration, and sentence lengths will be analyzed. The results of this analysis show that statistically significant differences in outcomes between appointed counsel and private counsel occur for conviction and incarceration. No significant differences occurred for guilty pleas or sentence length. Additionally, public defenders were not associated with statistically better or worse outcomes than either of the other types of counsel
First species of Laselva Furth from Brazil (Chrysomelidae: Galerucinae: Alticini)
Laselva cleidae sp. nov., the first species of Laselva Furth, 2007 found in Brazil is described and illustrated. Species of Laselva are compared with those of Deciplatus Linzmeier & Konstantinov, 2009 and Andersonaltica Linzmeier & Konstantinov, 2012. This is the first species of Laselva recorded to South America
Moss inhabiting flea beetles (Coleoptera: Chrysomelidae: Galerucinae: Alticini) of the West Indies II: Menudos, a new genus from Puerto Rico and description of methods to collect moss inhabiting flea beetles
Menudos Linzmeier & Konstantinov, a new genus of moss inhabiting flea beetles, containing five species, three of them are new (M. illariosus, M. maricao, and M. toronegro—the type species of the genus), from Puerto Rico is described and illustrated. The new genus is compared to Aedmon Clark, Apleuraltica Bechyne, Andersonaltica Linzmeier & Konstantinov, Distigmoptera Blake and Ulrica Scherer. Aedmon barberi (Blake, 1943b) and Distigmoptera chamorrae Konstantinov & Konstantinova, 2011 are transferred to Menudos. A key to Menudos species identification is provided. Methods for collecting moss inhabiting flea beetles and other arthropods are described in detail for the first time
