    Compressive Loading and Modeling of Stitched Composite Stiffeners

    A series of single-frame and single-stringer compression tests were conducted at NASA Langley Research Center on specimens harvested from a large panel built using the Pultruded Rod Stitched Efficient Unitized Structure (PRSEUS) concept. Different frame and stringer designs were used in fabrication of the PRSEUS panel. In this paper, the details of the experimental testing of single-frame and single-stringer compression specimens are presented, as well as discussions on the performance of the various structural configurations included in the panel.     Full-Scale Test and Analysis of a PRSEUS Fuselage Panel to Assess Damage-Containment Features

    Stitched composite technology has the potential to substantially decrease structural weight through enhanced damage containment capabilities. The most recent generation of stitched composite technology, the Pultruded Rod Stitched Efficient Unitized Structure (PRSEUS) concept, has been shown to successfully arrest damage at the sub-component level through tension testing of a three stringer panel with damage in the form of a two-bay notch. In a joint effort undertaken by the National Aeronautics and Space Administration (NASA), the Federal Aviation Administration (FAA), and the Boeing Company, further studies are being conducted to characterize the damage containment features of the PRSEUS concept. A full-scale residual strength test will be performed on a fuselage panel to determine if the load capacity will meet strength, deformation, and damage tolerance requirements. A curved panel was designed, fabricated, and prepared for residual strength testing. A pre-test Finite Element Model (FEM) was developed using design allowables from previous test programs to predict test panel deformation characteristics and margins of safety. Three phases of testing with increasing damage severity include: (1) as manufactured; (2) barely visible impact damage (BVID) and visible impact damage (VID); and (3) discrete source damage (DSD) where the panel will be loaded to catastrophic failure. This paper presents the background information, test plan, and experimental procedure.     Pressure Testing of a Minimum Gauge PRSEUS Panel

    Advanced aircraft configurations that have been developed to increase fuel efficiency require advanced, novel structural concepts capable of handling the unique load conditions that arise. One such concept is the Pultruded Rod Stitched Efficient Unitized Structure (PRSEUS) developed by the Boeing Company. The PRSEUS concept is being investigated by NASA s Environmentally Responsible Aviation (ERA) Program for use in a hybrid-wing body (HWB) aircraft. This paper summarizes the analysis and test of a PRSEUS panel subjected to internal pressure, the first such pressure test for this structural concept. The pressure panel used minimum gauge skin, with stringer and frame configurations consistent with previous PRSEUS tests. Analysis indicated that for the minimum gauge skin panel, the stringer locations exhibit fairly linear response, but the skin bays between the stringers exhibit nonlinear response. Excellent agreement was seen between nonlinear analysis and test results in the critical portion at the center of the panel. The pristine panel was capable of withstanding the required 18.4 psi pressure load condition without exhibiting any damage. The impacted panel was capable of withstanding a pressure load in excess of 28 psi before initial failure occurred at the center stringer, and the panel was capable of sustaining increased pressure load after the initial failure. This successful PRSEUS panel pressure panel test was a critical step in the building block approach for enabling the use of this advanced structural concept on future aircraft, such as the HWB

    Structural requirements for PACSIN/Syndapin operation during zebrafish embryonic notochord development.

    PACSIN/Syndapin proteins are membrane-active scaffolds that participate in endocytosis. The structure of the Drosophila Syndapin N-terminal EFC domain reveals a crescent shaped antiparallel dimer with a high affinity for phosphoinositides and a unique membrane-inserting prong upon the concave surface. Combined structural, biochemical and reverse genetic approaches in zebrafish define an important role for Syndapin orthologue, Pacsin3, in the early formation of the notochord during embryonic development. In pacsin3-morphant embryos, midline convergence of notochord precursors is defective as axial mesodermal cells fail to polarize, migrate and differentiate properly. The pacsin3 morphant phenotype of a stunted body axis and contorted trunk is rescued by ectopic expression of Drosophila Syndapin, and depends critically on both the prong that protrudes from the surface of the bowed Syndapin EFC domain and the ability of the antiparallel dimer to bind tightly to phosphoinositides. Our data confirm linkage between directional migration, endocytosis and cell specification during embryonic morphogenesis and highlight a key role for Pacsin3 in this coupling in the notochord

    Drug interactions and pharmacogenetic factors contribute to variation in apixaban concentration in atrial fibrillation patients in routine care

    Factor Xa-inhibitor apixaban is an oral anticoagulant prescribed in atrial fibrillation (AF) for stroke prevention. Its pharmacokinetic profile is known to be affected by cytochrome P450 (CYP)3A metabolism, while it is also a substrate of the efflux transporters ATP-binding cassette (ABC)B1 (P-glycoprotein) and ABCG2 (breast cancer resistance protein, BCRP). In this study, we assessed the impact of interacting medication and pharmacogenetic variation to better explain apixaban concentration differences among 358 Caucasian AF patients. Genotyping (ABCG2, ABCB1, CYP3A4*22, CYP3A5*3) was performed by TaqMan assays, and apixaban quantified by mass spectrometry. The typical patient was on average 77.2 years old, 85.5 kg, and had a serum creatinine of 103.1 Β΅mol/L. Concomitant amiodarone, an antiarrhythmic agent and moderate CYP3A/ABCB1 inhibitor, the impaired-function variant ABCG2 c.421C \u3e A, and sex predicted higher apixaban concentrations when controlling for age, weight and serum creatinine (multivariate regression; R2 = 0.34). Our findings suggest that amiodarone and ABCG2 genotype contribute to interpatient apixaban variability beyond known clinical factors

    Analysis of Clinical Presentations of Bruton Disease: A Review of 20 Years of Accumulated Data from Pediatric Patients at Severance Hospital

    PURPOSE: X-linked agammaglobulinemia (XLA) is a humoral immunodeficiency disease caused by a mutation in the Bruton tyrosine kinase (BTK) gene resulting in defective B cell differentiation. Because it is a relatively rare disorder, it is difficult for clinicians to have a comprehensive understanding of XLA due to a lack of exposure to the disease. Clinical presentations of patients with XLA were analyzed and discussed to improve care plans. MATERIALS AND METHODS: During a 20 year period, from January 1987 to June 2006, a total of 19 patients were diagnosed as XLA in the Department of Pediatrics at Severance Hospital, Seoul, Korea. A retrospective analysis of the clinical presentations of those patients was performed. RESULTS: The mean age of the XLA patients included in the study was 4.89 years, with a range of 6 months to 13 years. Twelve patients were diagnosed before age 5, while the other 7 patients were diagnosed after age 5. Recurrent infections observed in the patients included pneumonia, acute otitis media, septic arthritis, skin infection, sepsis, sinusitis, acute gastroenteritis, cervical lymphadenitis, epididymitis, meningitis, osteomyelitis, urinary tract infection and encephalitis. Frequency of admissions was variable from 0 to 12 times, depending on the time at which immunoglobulin therapy was started. Six cases had family histories positive for XLA. BTK gene mutations were found in 8 cases. CONCLUSION: The overall prognosis of XLA is good as long as patients are diagnosed and treated early with regular intra venous gamma globulin therapy before the sequelae of recurrent infections appear.ope

    Do Consumption-Based Asset Pricing Models Explain Serial Dependence in Stock Returns?

    We show that the Bansal-Yaron, Campbell-Cochrane and Cecchetti-Lam-Mark models of asset prices cannot explain the serial correlation structure of stock returns. We show this by estimating these models and deriving expected returns from them and then testing whether the difference between observed and expected returns is a martingale difference sequence. We use variance ratio and rescaled range tests which we modify to account for the expected returns being functions of estimated parameters. We also use a weighted quantilogram test based on a bootstrap procedure robust to this estimation. The evidence against the BansalYaron and Campbell-Cochrane models is significant. While the evidence against the Cecchetti-Lam-Mark model is not in general significant, our point estimates strongly suggest its residuals are not a martingale difference sequence. Furthermore, a semi-parametric maximal predictability test suggests there is some evidence that the three models’ state variables struggle to explain the degree of predictability observed in the market return. A timing strategy designed to exploit predictability in the market can significantly outperform the market in certainty equivalent terms under the Bansal-Yaron model. The timing strategy may underperform the market by less than it ought to under the Campbell-Cochrane model
