OBJECTIVE: To quantify the concentration of sulfated glycosaminoglycans (GAGs) concentration in the synovial fluid (SF) of knees with chronic anterior cruciate ligament (ACL) rupture and to identify possible associations between GAG concentration in SF and the time elapsed since rupture and degree of chondral injury. METHOD: Fourteen adult male subjects with total unilateral ACL rupture, which had occurred between 5 and 144 months earlier, were assessed. All subjects underwent joint aspiration; it was possible to collect SF from ten individuals. The samples were quantified to determine the GAG concentration using dimethylmethylene blue (DMMB) staining. The degree of chondral injury was macroscopically evaluated using the modified Mankin histological scale. Spearman correlation test (< 0.05) was used to evaluate the association between GAG concentration and chondral injury, and Pearson correlation test (< 0.05) was used to evaluate the association between GAG concentration and the time elapsed since rupture. RESULTS: The GAG concentration in SF showed a mean variation of 73.84 ± 40.75 µg/ml, with a mean time of 40.4 ± 40.3 months since the rupture. There was no correlation between GAG concentration and time since the rupture (r= -0.09, p= 0.81). The chondral injury grades found were 0, 1, 4 and 5. There was no correlation between chondral injury grade and GAG concentration in SF (r= -0.41, p= 0.24). CONCLUSION: After at least 5 months, the GAG concentration in SF from knees with ACL rupture is independent of the time elapsed since rupture and/or the severity of chondral injury.OBJETIVO: Quantificar a concentração de glicosaminoglicanas sulfatadas (GAGs) no líquido sinovial (LS) de joelhos com ruptura crônica do ligamento cruzado anterior (LCA) e identificar uma possível correlação entre a concentração de GAGs no LS e o tempo pós-ruptura e grau de lesão condral.
Two rapid assays for screening of patulin biodegradation
Artículo sobre distintos ensayos para comprobar la biodegradación de la patulinaThe mycotoxin patulin is produced by the blue
mould pathogen Penicillium expansum in rotting apples
during postharvest storage. Patulin is toxic to a wide range
of organisms, including humans, animals, fungi and bacteria.
Wash water from apple packing and processing
houses often harbours patulin and fungal spores, which can
contaminate the environment. Ubiquitous epiphytic yeasts,
such as Rhodosporidium kratochvilovae strain LS11 which
is a biocontrol agent of P. expansum in apples, have the
capacity to resist the toxicity of patulin and to biodegrade
it. Two non-toxic products are formed. One is desoxypatulinic
acid. The aim of the work was to develop rapid,
high-throughput bioassays for monitoring patulin degradation
in multiple samples. Escherichia coli was highly
sensitive to patulin, but insensitive to desoxypatulinic acid.
This was utilized to develop a detection test for patulin,
replacing time-consuming thin layer chromatography or
high-performance liquid chromatography. Two assays for patulin degradation were developed, one in liquid medium
and the other in semi-solid medium. Both assays allow the
contemporary screening of a large number of samples. The
liquid medium assay utilizes 96-well microtiter plates and
was optimized for using a minimum of patulin. The semisolid
medium assay has the added advantage of slowing
down the biodegradation, which allows the study and isolation
of transient degradation products. The two assays are
complementary and have several areas of utilization, from
screening a bank of microorganisms for biodegradation
ability to the study of biodegradation pathways
Trauma in the elderly caused by traffic accident: integrative review
OBJECTIVE To describe the scientific knowledge produced about trauma in the elderly caused by traffic accidents in healthcare area studies. METHODS Integrative review of studies from 2003 to 2013 searched in LILACS, SciELO, PubMed and CINHAL databases. We used combination of the descriptors injuries, wounds and accidents, in English, Portuguese and Spanish languages. RESULTS 32 studies were selected. In the thematic analysis, three categories emerged: epidemiological data from traffic accidents involving elderly; traffic accidents with elderly pedestrians; and trauma care in the elderly. We observed increased incidence of trauma in most countries and pedestrians represented a large part of the victims. Among these, the elderly are the most vulnerable group. CONCLUSION Studies showed that trauma care in the elderly need protocols and professionals with training in gerontology specialized in trauma care services
Stabilizing the Complex Structure in Heterotic Calabi-Yau Vacua
In this paper, we show that the presence of gauge fields in heterotic
Calabi-Yau compacitifications causes the stabilisation of some, or all, of the
complex structure moduli of the Calabi-Yau manifold while maintaining a
Minkowski vacuum. Certain deformations of the Calabi-Yau complex structure,
with all other moduli held fixed, can lead to the gauge bundle becoming
non-holomorphic and, hence, non-supersymmetric. This leads to an F-term
potential which stabilizes the corresponding complex structure moduli. We use
10- and 4-dimensional field theory arguments as well as a derivation based
purely on algebraic geometry to show that this picture is indeed correct. An
explicit example is presented in which a large subset of complex structure
moduli is fixed. We demonstrate that this type of theory can serve as the
hidden sector in heterotic vacua and can co-exist with realistic particle
physics.Comment: 17 pages, Late
Yukawa Textures From Heterotic Stability Walls
A holomorphic vector bundle on a Calabi-Yau threefold, X, with h^{1,1}(X)>1
can have regions of its Kahler cone where it is slope-stable, that is, where
the four-dimensional theory is N=1 supersymmetric, bounded by "walls of
stability". On these walls the bundle becomes poly-stable, decomposing into a
direct sum, and the low energy gauge group is enhanced by at least one
anomalous U(1) gauge factor. In this paper, we show that these additional
symmetries can strongly constrain the superpotential in the stable region,
leading to non-trivial textures of Yukawa interactions and restrictions on
allowed masses for vector-like pairs of matter multiplets. The Yukawa textures
exhibit a hierarchy; large couplings arise on the stability wall and some
suppressed interactions "grow back" off the wall, where the extended U(1)
symmetries are spontaneously broken. A number of explicit examples are
presented involving both one and two stability walls, with different
decompositions of the bundle structure group. A three family standard-like
model with no vector-like pairs is given as an example of a class of SU(4)
bundles that has a naturally heavy third quark/lepton family. Finally, we
present the complete set of Yukawa textures that can arise for any holomorphic
bundle with one stability wall where the structure group breaks into two
factors.Comment: 53 pages, 4 figures and 13 table
Wheat-barley hybridization – the last forty years
Abstract Several useful alien gene transfers have
been reported from related species into wheat (Triticum
aestivum), but very few publications have dealt
with the development of wheat/barley (Hordeum
vulgare) introgression lines. An overview is given
here of wheat 9 barley hybridization over the last
forty years, including the development of
wheat 9 barley hybrids, and of addition and translocation
lines with various barley cultivars. A short
summary is also given of the wheat 9 barley hybrids
produced with other Hordeum species. The meiotic
pairing behaviour of wheat 9 barley hybrids is presented,
with special regard to the detection of wheat–
barley homoeologous pairing using the molecular
cytogenetic technique GISH. The effect of in vitro
multiplication on the genome composition of intergeneric
hybrids is discussed, and the production and
characterization of the latest wheat/barley translocation
lines are presented. An overview of the agronomical
traits (b-glucan content, earliness, salt tolerance,
sprouting resistance, etc.) of the newly developed
introgression lines is given. The exploitation and
possible use of wheat/barley introgression lines for
the most up-to-date molecular genetic studies
(transcriptome analysis, sequencing of flow-sorted
chromosomes) are also discussed
