    Function and regulation of the glutathione peroxidase homologous gene GPXH

    When exposed to strong sunlight, photosynthetic organisms encounter photooxidative stress by the increased production of reactive oxygen species causing harmful damages to proteins and membranes. Consequently, a fast and specific induction of defense mechanisms is required to protect the organism from cell death. In Chlamydomonas reinhardtii, the glutathione peroxidase homologous gene GPXH/GPX5 was shown to be specifically upregulated by singlet oxygen formed during high light conditions presumably to prevent the accumulation of lipid hydroperoxides and membrane damage. We now showed that the GPXH protein is a thioredoxin-dependent peroxidase catalyzing the reduction of hydrogen peroxide and organic hydroperoxides. Furthermore, the GPXH gene seems to encode a dual-targeted protein, predicted to be localized both in the chloroplast and the cytoplasm, which is active with either plastidic TRXy or cytosolic TRXh1. Putative dual-targeting is achieved by alternative transcription and translation start sites expressed independently from either a TATA-box or an Initiator core promoter. Expression of both transcripts was upregulated by photooxidative stress even though with different strengths. The induction required the presence of the core promoter sequences and multiple upstream regulatory elements including a Sp1-like element and an earlier identified CRE/AP-1 homologous sequence. This element was further characterized by mutation analysis but could not be confirmed to be a consensus CRE or AP1 element. Instead, it rather seems to be another member of the large group of TGAC-transcription factor binding sites found to be involved in the response of different genes to oxidative stres

    The HOX Code as a “biological fingerprint” to distinguish functionally distinct stem cell populations derived from cord blood

    AbstractMesenchymal stem cells (MSC) have been isolated from almost every adult tissue. In cord blood (CB), different non-hematopoietic CD45-, CD34− adherent cell populations can be generated: the cord blood derived MSC (CB-MSC), that behave almost like MSC from bone marrow (BM-MSC), and unrestricted somatic stem cells (USSC) which show a distinct differentiation potential into all three germ layers. However, distinguishing these populations easily by molecular markers is still a concern. In this study we were able to present the HOX expression pattern of USSC, CB-MSC and BM-MSC, which in fact allows a discrimination of these populations.Briefly, RT-PCR analysis of the HOX code revealed a high similarity between BM-MSC and CB-MSC, which are both HOX-positive, whereas USSC resembled H9 embryonic stem cells HOX-negative.Especially HOXA9, HOXB7, HOXC10 and HOXD8 are good candidate markers to discriminate MSC from USSC. Thus, our data suggest that the "biological fingerprint" based on the HOX code can be used to distinguish functionally distinct MSC populations derived from bone marrow and cord blood

    Gonadal Malformations in Whitefish from Lake Thun: Defining the Case and Evaluating the Role of EDCs

    The objectives of this project were to evaluate i) whether the gonad alterations of whitefish (Coregonus lavaretus spp.) in Lake Thun represent abnormal morphological variations specific to this lake, and, if so, ii) whether the malformations are related to chemical exposure, in particular to exposure to endocrine-disrupting compounds (EDCs). Large-scale monitoring data revealed that, although whitefish in other lakes display some background variation of gonad morphology, the situation in Lake Thun, is unique because of the significantly higher prevalence of gonad malformations. The abnormal variations of whitefish gonad morphology include aplasias, compartmentations, fusions, and intersex. In the search for the factor(s) causing the gonad malformations, coregonids were exposed from fertilization up to maturity to Lake Thun water and plankton or to contaminants possibly being present in the lake, including trinitrotoluenes, and naphtalene sulfonates. Since these experiments are still ongoing, a conclusive answer cannot be given yet, but initial observations point to a role of the lake plankton. The possible presence of EDCs in Lake Thun was assessed using bioanalytics and biomarkers. The bioanalytical studies found estrogenic activities in concentrated plankton extracts of Lake Thun, however, estrogenic activities occurred also in plankton extracts of reference lakes. Bioassay-directed fractionation of the plankton samples points to degradation products of natural substances as a cause of the estrogenic activity. Examination of Lake Thun whitefish for EDC biomarkers such as vitellogenin, sex steroid levels or intersex frequency yielded no indications of exposure to EDCs, neither in fish with normal nor in fish with abnormal gonad morphology. Long-term laboratory exposure of developing coregonids to the prototype estrogenic compound, 17?-estradiol, resulted in an increased frequency of intersex gonads, but did not induce the other gonad malformations typical for Lake Thun coregonids. In summing up, the currently available evidence does not support an EDC or chemical etiology of the gonad malformations, however, this preliminary conclusion needs to be substantiated in the ongoing investigations. The project also highlights the need for more detailed knowledge of natural variation in wildlife populations to be able to recognize anthropogenically caused variation

    Investigation of the change of the shopping mall's space organization in Istanbul

    Due to copyright restrictions, the access to the full text of this article is only available via subscription.Within the scope of this article shopping malls were discussed as a building type and the evolution of a building type was tried to be witnessed within a twenty-five year period by comparing the early and late examples of shopping malls in Istanbul. The change of Istanbul’s shopping culture / spaces and the inclusion of shopping centers into the practice of architecture as a building type are briefly mentioned. Each of the selected malls were separately examined in terms of location, concept, space organization and other functions included, and in the fourth section both early and late examples were evaluated together in the same terms with a holistic perspective. The effect of shopping malls, the number of which constantly increases in Istanbul, on the city and its architecture is highly important. These massive bodies of buildings play a significant role in the formation and even definition of the new settlement areas of the city, and generate new areas of attraction. Due to these reasons, from location to architectural characteristic, from schedule to interior quality, they need to be designed punctiliously and with the consideration of the effects at all levels

    Knowledge as transformative energy : on linking models and experiments in the energy transition in buildings

    The transformation processes towards a sustainable development are complex. How can science contribute towards new solutions and ideas leading to change in practice? The authors of this book discuss these questions along the energy transition in the building sector. A transformative research that leaves the neutral observer position needs appropriate concepts and methods: how can knowledge from different disciplines and from practice be integrated in order to be able to explain and understand complex circumstances and interrelations? What role do complex (agent-based) models and experiments play in this respect? Which mix of methods is required in transformative science in order to actively support the actors in transformation processes? Theses questions are illustrated by the example of the BMBF funded project "EnerTransRuhr"
