    Human subjective response to steering wheel vibration caused by diesel engine idle

    This study investigated the human subjective response to steering wheel vibration of the type caused by a four-cylinder diesel engine idle in passenger cars. Vibrotactile perception was assessed using sinusoidal amplitude-modulated vibratory stimuli of constant energy level (r.m.s. acceleration, 0.41 m/s(2)) having a carrier frequency of 26 Hz (i.e. engine firing frequency) and modulation frequency of 6.5 Hz (half-order engine harmonic). Evaluations of seven levels of modulation depth parameter m (0.0, 0.1, 0.2, 0.4, 0.6, 0.8, and 1.0) were performed in order to define the growth function of human perceived disturbance as a function of amplitude modulation depth. Two semantic descriptors were used (unpleasantness and roughness) and two test methods (the Thurstone paired-comparison method and the Borg CR-10 direct evaluation scale) for a total of four tests. Each test was performed using an independent group of 25 individuals. The results suggest that there is a critical value of modulation depth m = 0.2 below which human subjects do not perceive differences in amplitude modulation and above which the stimulus-response relationship increases monotonically with a power function. The Stevens power exponents suggest that the perceived unpleasantness is non-linearly dependent on modulation depth m with an exponent greater than 1 and that the perceived roughness is dependent with an exponent close to unity

    Effects of White Space in Learning via the Web

    This study measured the effect of specific white space features on learning from instructional Web materials. The study also measured learners' beliefs regarding Web-based instruction. Prior research indicated that small changes in the handling of presentation elements can affect learning. Achievement results from this study indicated that in on-line materials, when content and overall structure are sound, minor differences regarding table borders and vertical spacing in text do not hinder learning. Beliefs regarding Web-based instruction and instructors who use it did not differ significantly between treatment groups.     Towards a Rigorous Network of Protein-Protein Interactions of the Model Sulfate Reducer Desulfovibrio vulgaris Hildenborough

    Protein–protein interactions offer an insight into cellular processes beyond what may be obtained by the quantitative functional genomics tools of proteomics and transcriptomics. The aforementioned tools have been extensively applied to study Escherichia coli and other aerobes and more recently to study the stress response behavior of Desulfovibrio vulgaris Hildenborough, a model obligate anaerobe and sulfate reducer and the subject of this study. Here we carried out affinity purification followed by mass spectrometry to reconstruct an interaction network among 12 chromosomally encoded bait and 90 prey proteins based on 134 bait-prey interactions identified to be of high confidence. Protein-protein interaction data are often plagued by the lack of adequate controls and replication analyses necessary to assess confidence in the results, including identification of potential false positives. We addressed these issues through the use of biological replication, exponentially modified protein abundance indices, results from an experimental negative control, and a statistical test to assign confidence to each putative interacting pair applicable to small interaction data studies. We discuss the biological significance of metabolic features of D. vulgaris revealed by these protein-protein interaction data and the observed protein modifications. These include the distinct role of the putative carbon monoxide-induced hydrogenase, unique electron transfer routes associated with different oxidoreductases, and the possible role of methylation in regulating sulfate reduction

    Rhabdovirus Matrix Protein Structures Reveal a Novel Mode of Self-Association

    The matrix (M) proteins of rhabdoviruses are multifunctional proteins essential for virus maturation and budding that also regulate the expression of viral and host proteins. We have solved the structures of M from the vesicular stomatitis virus serotype New Jersey (genus: Vesiculovirus) and from Lagos bat virus (genus: Lyssavirus), revealing that both share a common fold despite sharing no identifiable sequence homology. Strikingly, in both structures a stretch of residues from the otherwise-disordered N terminus of a crystallographically adjacent molecule is observed binding to a hydrophobic cavity on the surface of the protein, thereby forming non-covalent linear polymers of M in the crystals. While the overall topology of the interaction is conserved between the two structures, the molecular details of the interactions are completely different. The observed interactions provide a compelling model for the flexible self-assembly of the matrix protein during virion morphogenesis and may also modulate interactions with host proteins

    An AP-MS- and BioID-compatible MAC-tag enables comprehensive mapping of protein interactions and subcellular localizations

    Protein-protein interactions govern almost all cellular functions. These complex networks of stable and transient associations can be mapped by affinity purification mass spectrometry (AP-MS) and complementary proximity-based labeling methods such as BioID. To exploit the advantages of both strategies, we here design and optimize an integrated approach combining AP-MS and BioID in a single construct, which we term MAC-tag. We systematically apply the MAC-tag approach to 18 subcellular and 3 sub-organelle localization markers, generating a molecular context database, which can be used to define a protein's molecular location. In addition, we show that combining the AP-MS and BioID results makes it possible to obtain interaction distances within a protein complex. Taken together, our integrated strategy enables the comprehensive mapping of the physical and functional interactions of proteins, defining their molecular context and improving our understanding of the cellular interactome.Peer reviewe

    A model of management academics' intentions to influence values

    Business schools face increased criticism for failing in the teaching of management studies to nurture their students’ values. Assuming that individual academics play an important role in shaping the value-related influence of business schools, I model management academics’ intentions to influence values. The suggested model encompasses academics’ economic and social values as internal variables, as well as perceived support for attempting to influence values and academic tenure as social and structural variables. A test with empirical data from 1,254 management academics worldwide reveals that perceived external support is most relevant for explaining intentions. Moreover, academics’ social values, but not their economic ones, contribute to an explanation of their intentions to influence values. The results reveal how important it is for academics to believe that their colleagues, higher education institutions, and other stakeholders support their value-related behavioral intentions
