    Polymorphism and association of microsatellite SJ01 with birth weight and early growth traits in pigs

    Myostatin is a negative regulator of animal skeletal muscle development and SJ01 is a microsatellite locus flanking porcine myostatin gene. In the present study, the polymorphism of microsatellite SJ01 in three pig populations, that is, Duroc (n = 420), Yorkshire (n = 414) and Landrace (n = 119), were investigated, and its associations with birth weight and early growth traits were analyzed. The results indicated that genotype BB was predominant in Yorkshire pigs and allele B was predominant in both Yorkshire and Landrace pigs, whereas allele A was more prevalent in Duroc pigs. Among the three pig breeds, Duroc population had the highest heterozygosity and deviated from Hardy-Weinberg equilibrium (P < 0.05). The average daily gain from 28 d to 70 d in Yorkshire pigs and the body weight at 70 d in Landrace pigs were significantly different between SJ01 genotypes (P < 0.05). These resultssuggest that microsatellite SJ01 is a potential DNA marker for early growth trait selection in Yorkshire and Landrace pigs as well as for refining QTL for early growth traits on SSC15

    Progenitor cells are considered an important cell of origin of human malignancies. However, there has not been any single gene that can define mammary bipotential progenitor cells, and as such it has not been possible to use genetic methods to introduce oncogenic alterations into these cells in vivo to study tumorigenesis from them. Keratin 6a is expressed in a subset of mammary luminal epithelial cells and body cells of terminal end buds. By generating transgenic mice using the Keratin 6a (K6a) gene promoter to express tumor virus A (tva), which encodes the receptor for avian leukosis virus subgroup A (ALV/A), we provide direct evidence that K6a(+) cells are bipotential progenitor cells, and the first demonstration of a non-basal location for some biopotential progenitor cells. These K6a(+) cells were readily induced to form mammary tumors by intraductal injection of RCAS (an ALV/A-derived vector) carrying the gene encoding the polyoma middle T antigen. Tumors in this K6a-tva line were papillary and resembled the normal breast-like subtype of human breast cancer. This is the first model of this subtype of human tumors and thus may be useful for preclinical testing of targeted therapy for patients with normal-like breast cancer. These observations also provide direct in vivo evidence for the hypothesis that the cell of origin affects mammary tumor phenotypes

    Spin Hall effect in the kagome lattice with Rashba spin-orbit interaction

    We study the spin Hall effect in the kagom\'{e} lattice with Rashba spin-orbit coupling. The conserved spin Hall conductance σxys\sigma_{xy}^{s} (see text) and its two components, i.e., the conventional term σxys0\sigma_{xy}^{s0} and the spin-torque-dipole term σxysτ\sigma_{xy}^{s\tau}, are numerically calculated, which show a series of plateaus as a function of the electron Fermi energy ϵF\epsilon_{F}. A consistent two-band analysis, as well as a Berry-phase interpretation, is also given. We show that these plateaus are a consequence of the various Fermi-surface topologies when tuning ϵF\epsilon_{F}. In particular, we predict that compared to the case with the Fermi surface encircling the Γ\mathbf{\Gamma} point in the Brillouin zone, the amplitude of the spin Hall conductance with the Fermi surface encircling the K\mathbf{K} points is twice enhanced, which makes it highly meaningful in the future to systematically carry out studies of the K\mathbf{K}-valley spintronics.Comment: 7 pages, 3 figures.     Antiproliferative activity of recombinant human interferon-λ2 expressed in stably transformed BmN cells

    This study aimed at the generation of a stable transformed silkworm BmN cell line which can continuously express human interferon-λ2 (IFN-λ2) gene, and investigated the antiproliferative activity of this recombinant human IFN-λ2. Silkworm BmN cells were transfected with the recombinant vector pIZT/V5-His harboring the human IFN-λ2 gene. After the BmN cells were transfected with the pIZT/V5- His- hIFN-λ2 vector, the stably transformed BmN cells expressing hIFN-λ2 gene were selected using Zeocin. Following two months of screening, the transformed BmN cell line was obtained. Stable transformed BmN cell line can be maintained at a lower Zeocin concentration. The representing 26 kDa protein band of IFN-λ2 was detected by SDS-PAGE and Western blotting. The expression level of hIFN- λ2, determined by ELISA, was about 8.142 ng in 4 × 105 cells. The antiproliferative activity of hIFN-λ2 was determined by MTT assay. The 50% inhibitory concentrations (IC50) of the recombinant hIFN-λ2 on A549 (lung cancer cells), HL60 (acute promyelocytic leukemia cells), BEL-7402 (liver cancer cells) and M231 cells (breast cancer cells) were approximately 3.21, 2.84, 6.29 and 9.32 ng/ml, respectively. In summary, Human IFN-λ2 can be stably expressed in the transformed BmN cell line, and the expressed recombinant hIFN-λ2 demonstrated antiproliferative activity to tumor cells in vitro.Key words: Human interferon-λ2 protein, gene expression, antiproliferative activity

    Intersecting black branes in strong gravitational waves

    We consider intersecting black branes with strong gravitational waves propagating along their worldvolume in the context of supergravity theories. Both near-horizon and space-filling gravitational wave modes are included in our ansatz. The equations of motion (originally, partial differential equations) are shown to reduce to ordinary differential equations, which include a Toda-like system.     EM23, a natural sesquiterpene lactone, targets thioredoxin reductase to activate JNK and cell death pathways in human cervical cancer cells

    Sesquiterpene lactones (SLs) are the active constituents of a variety of medicinal plants and found to have potential anticancer activities. However, the intracellular molecular targets of SLs and the underlying molecular mechanisms have not been well elucidated. In this study, we observed that EM23, a natural SL, exhibited anti-cancer activity in human cervical cancer cell lines by inducing apoptosis as indicated by caspase 3 activation, XIAP downregulation and mitochondrial dysfunction. Mechanistic studies indicated that EM23-induced apoptosis was mediated by reactive oxygen species (ROS) and the knockdown of thioredoxin (Trx) or thioredoxin reductase (TrxR) resulted in a reduction in apoptosis. EM23 attenuated TrxR activity by alkylation of C-terminal redox-active site Sec498 of TrxR and inhibited the expression levels of Trx/TrxR to facilitate ROS accumulation. Furthermore, inhibition of Trx/TrxR system resulted in the dissociation of ASK1 from Trx and the downstream activation of JNK. Pretreatment with ASK1/JNK inhibitors partially rescued cells from EM23-induced apoptosis. Additionally, EM23 inhibited Akt/mTOR pathway and induced autophagy, which was observed to be proapoptotic and mediated by ROS. Together, these results reveal a potential molecular mechanism for the apoptotic induction observed with SL compound EM23, and emphasize its putative role as a therapeutic agent for human cervical cancer.published_or_final_versio

    This article describes the development and formal verification (proof of semantic preservation) of a compiler back-end from Cminor (a simple imperative intermediate language) to PowerPC assembly code, using the Coq proof assistant both for programming the compiler and for proving its correctness. Such a verified compiler is useful in the context of formal methods applied to the certification of critical software: the verification of the compiler guarantees that the safety properties proved on the source code hold for the executable compiled code as well
