    The Coombs

    The Coombs Building at The Australian National University is a Canberra icon. Named after one of Australia’s greatest administrators and public intellectuals—‘Nugget’ Herbert Cole Coombs—for more than forty years the building has housed two of the University’s four foundational Schools: the Research School of Pacific and Asian Studies and the Research School of Social Sciences. This volume of recollections is about the former. It looks at life in the building through the prism of personal experience and happenstance.     Designing Technologies to Support Professional and Workplace Learning for Situated Practice

    The papers in this special section focus on designing technologies to support professional and workplace learning in situated practices. In an era of global, organizational, and technological change, all of which are transforming the world of work, professional and workplace learning are critical for employability and organizational competitiveness. A range of fundamental transformations is changing how people work. Digital technologies are replacing human labor and, at the same time, are accelerating the expansion of job roles and work practices. Work is becoming increasingly specialized, which means that professionals in collaborative and networked ways across discipline and organization boundaries. In parallel, labor is increasingly decentralized, making decisionmaking more distributed and raising the need for remote communication and collaboration. Subsequently, work is becoming more independent from time and place, as people connect, collaborate, and work via digital technologies. These changes come with a need for substantial and continuous workplace learning, and with the need for changes in how workplace learning happens. Of course, digital technologies are already used to provide learning and training in workplaces. However, most of these learning technologies have been developed for formal education (e.g., K- 12 and higher education) rather than in workplace contexts. There is a need to understand and evidence workplace learning needs and to further develop technologies that can support and scale workplace learning

    Global Service-Learning: A Systematic Review of Principles and Practices

In global service-learning, practitioners act at the intersection of two fields: service-learning and international development. Critical scholarship in all service-learning and international development has highlighted the tensions inherent in defining and tracking “success” in community development. In response, service-learning and international development have turned considerable attention to documenting project characteristics, also known as best practices or success factors, that support equitable, sustainable community development. This article presents a systematic synthesis of these fields’ best practices in the context of global service-learning. The authors propose 18 guiding principles for project design to support practitioners in creating and maintaining justice-oriented, stakeholder-driven projects.     Genomic sequencing in clinical trials

    Human genome sequencing is the process by which the exact order of nucleic acid base pairs in the 24 human chromosomes is determined. Since the completion of the Human Genome Project in 2003, genomic sequencing is rapidly becoming a major part of our translational research efforts to understand and improve human health and disease. This article reviews the current and future directions of clinical research with respect to genomic sequencing, a technology that is just beginning to find its way into clinical trials both nationally and worldwide. We highlight the currently available types of genomic sequencing platforms, outline the advantages and disadvantages of each, and compare first- and next-generation techniques with respect to capabilities, quality, and cost. We describe the current geographical distributions and types of disease conditions in which these technologies are used, and how next-generation sequencing is strategically being incorporated into new and existing studies. Lastly, recent major breakthroughs and the ongoing challenges of using genomic sequencing in clinical research are discussed

    The Canine Oral Microbiome

    Determining the bacterial composition of the canine oral microbiome is of interest for two primary reasons. First, while the human oral microbiome has been well studied using molecular techniques, the oral microbiomes of other mammals have not been studied in equal depth using culture independent methods. This study allows a comparison of the number of bacterial taxa, based on 16S rRNA-gene sequence comparison, shared between humans and dogs, two divergent mammalian species. Second, canine oral bacteria are of interest to veterinary and human medical communities for understanding their roles in health and infectious diseases. The bacteria involved are mostly unnamed and not linked by 16S rRNA-gene sequence identity to a taxonomic scheme. This manuscript describes the analysis of 5,958 16S rRNA-gene sequences from 65 clone libraries. Full length 16S rRNA reference sequences have been obtained for 353 canine bacterial taxa, which were placed in 14 bacterial phyla, 23 classes, 37 orders, 66 families, and 148 genera. Eighty percent of the taxa are currently unnamed. The bacterial taxa identified in dogs are markedly different from those of humans with only 16.4% of oral taxa are shared between dogs and humans based on a 98.5% 16S rRNA sequence similarity cutoff. This indicates that there is a large divergence in the bacteria comprising the oral microbiomes of divergent mammalian species. The historic practice of identifying animal associated bacteria based on phenotypic similarities to human bacteria is generally invalid.     Genome Wide Analysis of Acute Myeloid Leukemia Reveal Leukemia Specific Methylome and Subtype Specific Hypomethylation of Repeats

    Methylated DNA immunoprecipitation followed by high-throughput sequencing (MeDIP-seq) has the potential to identify changes in DNA methylation important in cancer development. In order to understand the role of epigenetic modulation in the development of acute myeloid leukemia (AML) we have applied MeDIP-seq to the DNA of 12 AML patients and 4 normal bone marrows. This analysis revealed leukemia-associated differentially methylated regions that included gene promoters, gene bodies, CpG islands and CpG island shores. Two genes (SPHKAP and DPP6) with significantly methylated promoters were of interest and further analysis of their expression showed them to be repressed in AML. We also demonstrated considerable cytogenetic subtype specificity in the methylomes affecting different genomic features. Significantly distinct patterns of hypomethylation of certain interspersed repeat elements were associated with cytogenetic subtypes. The methylation patterns of members of the SINE family tightly clustered all leukemic patients with an enrichment of Alu repeats with a high CpG density (P<0.0001). We were able to demonstrate significant inverse correlation between intragenic interspersed repeat sequence methylation and gene expression with SINEs showing the strongest inverse correlation (R2 = 0.7). We conclude that the alterations in DNA methylation that accompany the development of AML affect not only the promoters, but also the non-promoter genomic features, with significant demethylation of certain interspersed repeat DNA elements being associated with AML cytogenetic subtypes.     Consumption of pasteurized human lysozyme transgenic goats’ milk alters serum metabolite profile in young pigs

    Nutrition, bacterial composition of the gastrointestinal tract, and general health status can all influence the metabolic profile of an organism. We previously demonstrated that feeding pasteurized transgenic goats’ milk expressing human lysozyme (hLZ) can positively impact intestinal morphology and modulate intestinal microbiota composition in young pigs. The objective of this study was to further examine the effect of consuming hLZ-containing milk on young pigs by profiling serum metabolites. Pigs were placed into two groups and fed a diet of solid food and either control (non-transgenic) goats’ milk or milk from hLZ-transgenic goats for 6 weeks. Serum samples were collected at the end of the feeding period and global metabolite profiling was performed. For a total of 225 metabolites (160 known, 65 unknown) semi-quantitative data was obtained. Levels of 18 known and 4 unknown metabolites differed significantly between the two groups with the direction of change in 13 of the 18 known metabolites being almost entirely congruent with improved health status, particularly in terms of the gastrointestinal tract health and immune response, with the effects of the other five being neutral or unknown. These results further support our hypothesis that consumption of hLZ-containing milk is beneficial to health
