    88 s., 15 s. příl. :obr., tab., grafy +CD ROMHlavním cílem diplomové práce bylo vyhodnotit úroveň pohybové aktivity a tělesné zdatnosti u adolescentů ve věku 15-17 let. Výběrový soubor tvořilo 108 adolescentů, z toho 63 dívek a 45 chlapců z Gymnázia a Střední odborné školy v Jilemnici. Úroveň pohybové aktivity byla zjišťována pomocí mezinárodních dotazníků a krokoměrů SW-700. Tělesná zdatnost byla stanovena na základě pěti motorických testů (hrudní předklony, záklon v lehu na břiše, dotyk prstů za zády, 90° kliky, vytrvalostní člunkový běh) vybraných z testové baterie FITNESSGRAM. Na základě somatických parametrů (tělesná výška, tělesná hmotnost) byl vypočítán index tělesné hmotnosti (BMI). Vztahová analýza pohybové aktivity a tělesné zdatnosti byla zpracována podle Pearsonova koeficientu korelace. Výsledky prokázaly nízkou úroveň pohybové aktivity u adolescentů, adolescenti nedosáhli doporučených hodnot počtu kroků a více než polovina adolescentů nevykonávala žádnou intenzivní či středně zatěžující pohybovou aktivitu doma, v okolí domu, při studiu či ve svém volném čase. Výsledky motorických testů spadaly do intervalů cílových zdravotních zón testové baterie FITNESSGRAM, nejlepších výsledků adolescenti dosáhli v oblastech silově-vytrvalostních schopností a pohyblivosti extenzorů trupu. Na základě výsledků korelačních koeficientů byla u adolescentů ve věku 15?17 let zjištěna velmi slabá závislost mezi pohybovou aktivitou a tělesnou zdatností (rxy = -0,20 do 0,21)

    Development of a human mitochondrial oligonucleotide microarray (h-MitoArray) and gene expression analysis of fibroblast cell lines from 13 patients with isolated F1Fo ATP synthase deficiency

    <p>Abstract</p> <p>Background</p> <p>To strengthen research and differential diagnostics of mitochondrial disorders, we constructed and validated an oligonucleotide microarray (h-MitoArray) allowing expression analysis of 1632 human genes involved in mitochondrial biology, cell cycle regulation, signal transduction and apoptosis. Using h-MitoArray we analyzed gene expression profiles in 9 control and 13 fibroblast cell lines from patients with F<sub>1</sub>F<sub>o </sub>ATP synthase deficiency consisting of 2 patients with mt9205ΔTA microdeletion and a genetically heterogeneous group of 11 patients with not yet characterized nuclear defects. Analysing gene expression profiles, we attempted to classify patients into expected defect specific subgroups, and subsequently reveal group specific compensatory changes, identify potential phenotype causing pathways and define candidate disease causing genes.</p> <p>Results</p> <p>Molecular studies, in combination with unsupervised clustering methods, defined three subgroups of patient cell lines – M group with mtDNA mutation and N1 and N2 groups with nuclear defect. Comparison of expression profiles and functional annotation, gene enrichment and pathway analyses of differentially expressed genes revealed in the M group a transcription profile suggestive of synchronized suppression of mitochondrial biogenesis and G1/S arrest. The N1 group showed elevated expression of complex I and reduced expression of complexes III, V, and V-type ATP synthase subunit genes, reduced expression of genes involved in phosphorylation dependent signaling along MAPK, Jak-STAT, JNK, and p38 MAP kinase pathways, signs of activated apoptosis and oxidative stress resembling phenotype of premature senescent fibroblasts. No specific functionally meaningful changes, except of signs of activated apoptosis, were detected in the N2 group. Evaluation of individual gene expression profiles confirmed already known <it>ATP6/ATP8 </it>defect in patients from the M group and indicated several candidate disease causing genes for nuclear defects.</p> <p>Conclusion</p> <p>Our analysis showed that deficiency in the ATP synthase protein complex amount is generally accompanied by only minor changes in expression of ATP synthase related genes. It also suggested that the site (mtDNA vs nuclear DNA) and the severity (ATP synthase content) of the underlying defect have diverse effects on cellular gene expression phenotypes, which warrants further investigation of cell cycle regulatory and signal transduction pathways in other OXPHOS disorders and related pharmacological models.</p

    Application of novel genomic techniques in studies of pathogenesis of selected rare inherited disorders

    Rare diseases are a heterogeneous group of disorders. Knowledge of their molecular basis is poor and till recently there were no appropriate methodical approaches due to a limited number of patients. Novel genomic techniques, especially the DNA array technology and the next generation sequencing emerging in last few years, enabled studies of these diseases even in small families and sporadic cases. This PhD thesis focuses on application of novel genomic techniques in studies of rare inherited diseases. It describes a use of DNA array technology in linkage analysis, analysis of differential gene expression, analysis of copy number variations and homozygous mapping, and a use of next generation sequencing technology. Combination of these methods was used for identification of molecular basis of adult neuronal ceroid lipofuscinosis, Rotor syndrome, isolated defect of ATP synthase and mucopolysaccharidosis type IIIC

    Preferences of the basic components of food by central European speciens of ants.

    This thesis has two parts: the first brings an overview of the role of various foods and nutrients in ants, the second presents the results of an experiment focused on preference of the basic components of food by central European species of ants. In the experiment we provided ants with six different baits (water, 1% sodium chlorid, olive oil, 20% amino acid glutamine, artificial honey [10% sucrose + 10% amino acid] and 20% sucrose). Total, 187 of 690 traps were visited by ants. There were 26 recorded species of ants. Preference of four most abundant species differed. F. polyctena attended most of the offered baits, while other species preferred the sugar components. In general, ants preferred artificial honey and sucrose, regardless of the season and habitats. Additional field tests with different bait concentration of salt and sucrose showed the same preferences regardless of bait type and no additive effects of the amino acid in the sucrose baits. The results of the experiment show a surprisingly consistent and strong preference of central European species of ants for sugars

    Ways of spending leisure time in Jilemnice and its neighbourhood by Youth between the ages of 11-18 with the intention of sport

    katedra: KTV; rozsah: 87 s.The main aim of my bachelor paper was to find out the ways of spending leisure time in Jilemnice and its neighbourhood by youth between the ages of 11-18 with the intention of doing sport. The bachelor paper is based on my own questionnaire. The boys and girl, I focused on, were students from secondary school, grammar school and comprehensive school. 750 students responded to the questionnaire. It was found out, that the youth between the ages of 11-18 spend the leisure time doing sport activities than watching TV, playing computer games. The youth participate in physical training at least five times a week, the most favorite sport is cross-country skiing. Jilemnice and its neighbourhood have a good sporting background for both active sportsmen and for those who like to do sports only for recreation. The youth demand only in-line skating rink and open-air swimming pool reconstruction in Jilemnice.Cílem bakalářské práce bylo zjistit využívání volného času v Jilemnici a okolí mládeží ve věku 11-18 let se zaměřením na sportovní aktivity. Pro získání potřebných dat byla použita anketa vlastní struktury. Sledovanou skupinu tvořili dívky a chlapci ze základních škol, gymnázia a střední odborné školy. Výzkumný soubor tvořilo 710 respondentů. Z výzkumného šetření vyplývá, že mládež ve věku 11-18 let se ve svém volném čase věnuje více sportu a pohybovým aktivitám než pasivním činnostem (počítač, televize, četba, poslech hudby). Mládež je ochotna věnovat se pětkrát a vícekrát týdně volnočasovým sportovním aktivitám, nejoblíbenějším sportem je lyžování. Jilemnice a okolí poskytuje mládeži dostatečné zázemí pro jejich oblíbené volnočasové pohybové aktivity, avšak mládež by v Jilemnici uvítala zrekonstruované zázemí pro kolečkové brusle a znovuotevření koupaliště

    Import 04/07/2011V mé diplomové práci je vypracováno řešení pokračování těžby bentonitu na lomu Maršov v rozšířeném dobývacím prostoru. V úvodní části je popsána stručná charakteristika lomu z hlediska geologie a hydrologie. Z výsledků geologického průzkumu byl proveden výpočet zásob v rozšířeném dobývacím prostoru. Dále je navržena otvírka lomu podle platných legislativ. Pozornost byla věnována dvěma variantám řešení navržené sanace a rekultivace. Řešení bylo posouzeno z hlediska báňsko-technických, ekonomických a ekologických parametrů. Na závěr je diplomová práce celkově zhodnocena.This thesis treats the continuation of bentonite mining at the Maršov quarry in extended allotment. At the beginning the brief quarry characterization is described in the light of geological and hydro-geological situation of the deposit. On grounds of information that was extracted from geological survey, reserves are calculated in extended allotment. In the next part of the thesis, another quarry progress is proposed according to valid legislation. Attention to two alternates of proposed sanitation and reclamation solution was aimed. Solution was reviewed in the light of technical–economical and environmental parameters. A solution was reviewed in the light of technical–economical and environmental parameters.     Film usage in foreign language teaching - on the example of methodologically processed Russian-speaking material for upper-secondary school

    The diploma thesis is focused on film usage in foreign language teaching. It presents film as a teaching aid suitable for complete language skills developmen, including the so called fifth language skill - "understanding of the seen and the heard". Strenghts and weaknesses of film in foreign language teaching are described along with tips how to avoid the latter. Practical tips are presented such as how to choose a suitable film, how to present it and process it methodologically to ensure the most efficient way of language acquisition. A typology of exercises was developed to meet the aim of efficiency including pre-, during- and post-viewing exercises. The practical part of the thesis consists of examples of lesson plans using film clips in teaching Russian language at secondary school
