    A biogeographic framework of octopod species diversification: the role of the Isthmus of Panama

    The uplift of the Isthmus of Panama (IP) created a land bridge between Central and South America and caused the separation of the Western Atlantic and Eastern Pacific oceans, resulting in profound changes in the environmental and oceanographic conditions. To evaluate how these changes have influenced speciation processes in octopods, fragments of two mitochondrial (Cytochrome oxidase subunit I, COI and 16S rDNA) and two nuclear (Rhodopsin and Elongation Factor-1 alpha, EF-1 alpha) genes were amplified from samples from the Atlantic and Pacific oceans. One biogeographical and four fossil calibration priors were used within a relaxed Bayesian phylogenetic analysis framework to estimate divergence times among dadogenic events. Reconstruction of the ancestral states in phylogenies was used to infer historical biogeography of the lineages and species dispersal routes. The results revealed three well-supported Glades of transisthmian octopus sister species pair/complex (TSSP/TSSC) and two additional clades showing a low probability of species diversification, having been influenced by the IP. Divergence times estimated in the present study revealed that octopod TSSP/TSSC from the Atlantic and Pacific diverged between the Middle Miocene and Early Pliocene (mean range = 5-18 Ma). Given that oceanographic changes caused by the uplift of the IP were so strong as to affect the global climate, we suggest that octopod TSSP/TSSC diverged because of these physical and environmental barriers, even before the complete uplift of the IP 3 Ma, proposed by the Late Pliocene model. The results obtained in this phylogenetic reconstruction also indicate that the octopus species pairs in each ocean share a recent common ancestor from the Pacific Ocean

    Geographic variability of Octopus insularis diet: from oceanic island to continental populations

    A predator’s choice of prey can be affected by many factors. We evaluated various influences on population dietary composition, individual specialization and size of prey in Octopus insularis populations from 2 continental and 4 insular locations. We expected that habitat diversity would lead to diet heterogeneity. Furthermore, in keeping with MacArthur & Wilson’s (1967) theory of island biogeography, we expected that diet diversity would be lower around islands than on the coast of the mainland. Both predictions were confirmed when prey remains from octopus middens were examined. The 2 continental areas exhibited a richer habitat diversity and a wider variety of prey. Niche widths in the continental areas were 2.42 and 2.03, with the lowest niche widths exhibited by the population from the most distant oceanic islands (1.30, 0.85). We found variation in the proportion of specialist relative to generalist individuals across areas based on the proportional similarity index. The correlation between habitat diversity and niche width (R2 = 0.84) was highly significant, as was distance from the continental shelf and niche width (R2 = 0.89). This study reaffirms the central position of octopuses in the nearshore benthic ecosystem, and supports MacArthur & Wilson’s (1967) prediction of a lower diversity of species on islands—which applies not only to the species themselves, but also indirectly for the diet of their predators

    Individual prey choices of octopuses: are they generalist or specialist?

Here, we analyzed the diet of octopuses of different populations with the aim to assess the importance of individual feeding habits as a factor affecting prey choice. Two methods were used, an assessment of the extent to which an individual octopus made choices of species representative of those population (PSi and IS) and 25% cutoff values for number of choices and percentage intake of individual on their prey. In one population of Octopus cf vulgaris in Bermuda individuals were generalist by IS=0.77, but most chose many prey of the same species, and were specialists on it by >75% intake. Another population had a wider prey selection, still generalist with PSi=0.66, but two individuals specialized by choices. In Bonaire, there was a wide range of prey species chosen, and the population was specialists by IS= 0.42. Individual choices revealed seven specialists and four generalists. A population of Octopus cyanea in Hawaii all had similar choices of crustaceans, so the population was generalist by IS with 0.74. But by individual choices, three were considered a specialist. A population of Enteroctopus dofleini from Puget Sound had a wide range of preferences, in which seven were also specialists, IS=0.53. By individual choices, thirteen were also specialists. Given the octopus specialty of learning during foraging, we hypothesize that both localized prey availability and individual personality differences could influence the exploration for prey and this translates into different prey choices across individuals and populations showed in this studyYe

    MKP-1 mediates glucocorticoid-induced ERK1/2 dephosphorylation and reduction in pancreatic beta-cell proliferation in islets from early lactating mothers

    Maternal pancreatic islets undergo a robust increase of mass and proliferation during pregnancy, which allows a compensation of gestational insulin resistance. Studies have described that this adaptation switches to a low proliferative status after the delivery. the mechanisms underlying this reversal are unknown, but the action of glucocorticoids (GCs) is believed to play an important role because GCs counteract the pregnancy-like effects of PRL on isolated pancreatic islets maintained in cell culture. Here, we demonstrate that ERK1/2 phosphorylation (phospho-ERK1/2) is increased in maternal rat islets isolated on the 19th day of pregnancy. Phospho-ERK1/2 status on the 3rd day after delivery (L3) rapidly turns to values lower than that found in virgin control rats (CTL). MKP-1, a protein phosphatase able to dephosphorylate ERK1/2, is increased in islets from L3 rats. Chromatin immunoprecipitation assay revealed that binding of glucocorticoid receptor (GR) to MKP-1 promoter is also increased in islets from L3 rats. in addition, dexamethasone (DEX) reduced phospho-ERK1/2 and increased MKP-1 expression in RINm5F and MIN-6 cells. Inhibition of transduction with cycloheximide and inhibition of phosphatases with orthovanadate efficiently blocked DEX-induced downregulation of phospho-ERK1/2. in addition, specific knockdown of MKP-1 with siRNA suppressed the downregulation of phosphoERK1/2 and the reduction of proliferation induced by DEX. Altogether, our results indicate that downregulation of phospho-ERK1/2 is associated with reduction in proliferation found in islets of early lactating mothers.     Hypertension is nearly universal in kidney transplant and several factors are associated with post transplant hypertension, including immunosuppressive medications and genetic predisposition. Objective: The aims were to evaluate the effects of spontaneously hypertensive rats (SHR) kidney transplantation in Wistar rats and the possible transference of 80/90-kDa N-domain ACE. Methods: To do so, the data from Wistar recipients of kidney from SHR were compared to data from transplanted Wistar submitted to CsA treatment and, to Wistar Sham. Results and Discussion: Despite the unaltered blood pressure observed at early stages, 80/90-kDa ACE was found expressed in the urine of rats 7 and 15 days after transplantation, which was intense when rats became hypertensive 30 days post-surgery. Conclusion: Our data show that this enzyme is associated with the development of hypertension, and this marker appears in the urine before any substantial blood pressure alteration.391111

    Phylloseptin-1 is leishmanicidal for amastigotes of Leishmania amazonensis inside infected macrophages

    Leishmania protozoans are the causal agents of neglected diseases that represent an important public health issue worldwide. The growing occurrence of drug-resistant strains of Leishmania and severe side effects of available treatments represent an important challenge for the leishmaniases treatment. We have previously reported the leishmanicidal activity of phylloseptin-1 (PSN-1), a peptide found in the skin secretion of Phyllomedusa azurea (=Pithecopus azureus), against Leishmania amazonensis promastigotes. However, its impact on the amastigote form of L. amazonensis and its impact on infected macrophages are unknown. In this work, we evaluated the effects of PSN-1 on amastigotes of L. amazonensis inside macrophages infected in vitro. We assessed the production of hydrogen peroxide and nitric oxide, as well as the levels of inflammatory and immunomodulatory markers (TGF-β, TNF-α and IL-12), in infected and non-infected macrophages treated with PSN-1. Treatment with PSN-1 decreased the number of infected cells and the number of ingested amastigotes per cell when compared with the untreated cells. At 32 µM (64 µg/mL), PSN-1 reduced hydrogen peroxide levels in both infected and uninfected macrophages, whereas it had little effect on NO production or TGF-β release. The effect of PSN-1 on IL-12 and TNF-α secretion depended on its concentration, but, in general, their levels tended to increase as PSN-1 concentration increased. Further in vitro and in vivo studies are needed to clarify the mechanisms of action of PSN-1 and its interaction with the immune system aiming to develop pharmacological applications
