    Stanniocalcin-1 Reduces Tumor Size in Human Hepatocellular Carcinoma

    Growing evidence has revealed high expression levels of stanniocalcin-1 (STC1) in different types of human cancers. Numerous experimental studies using cancer cell lines demonstrated the involvement of STC1 in inflammatory and apoptotic processes; however the role of STC1 in carcinogenesis remains elusive. Hepatocellular carcinoma (HCC) an exemplified model of inflammation-related cancer, represents a paradigm of studying the association between STC1 and tumor development. Therefore, we conducted a statistical analysis on the expression levels of STC1 using clinicopathological data from 216 HCC patients. We found that STC1 was upregulated in the tumor tissues and its expression levels was positively correlated with the levels of interleukin (IL)-6 and IL-8. Intriguingly tumors with greater expression levels of STC1 (tumor/normal >= 2) were significantly smaller than the lower level (tumor/normal<2) samples (p = 0.008). A pharmacological approach was implemented to reveal the functional correlation between STC1 and the ILs in the HCC cell-lines. IL-6 and IL-8 treatment of Hep3B cells induced STC1 expression. Lentiviral-based STC1 over-expression in Hep3B and MHCC-97L cells however showed inhibitory action on the pro-migratory effects of IL-6 and IL-8 and reduced size of tumor spheroids. The inhibitory effect of STC1 on tumor growth was confirmed in vivo using the stable STC1-overexpressing 97L cells on a mouse xenograft model. Genetic analysis of the xenografts derived from the STC1-overexpressing 97L cells, showed upregulation of the pro-apoptotic genes interleukin-12 and NOD-like receptor family, pyrin domain-containing 3. Collectively, the anti-inflammatory and pro-apoptotic functions of STC1 were suggested to relate its inhibitory effect on the growth of HCC cells.     Off-Street Vehicular Fog for Catering Applications in 5G/B5G: A Trust-based Task Mapping Solution and Open Research Issues

    One of the key enablers in serving the applications requiring stringent latency in 5G networks is fog computing as it is situated closer to the end users. With the technological advancement of vehicles’ on-board units, their computing capabilities are becoming robust, and considering the underutilization of the off-street vehicles, we envision that the off-street vehicles can be an enormously useful computational source for the fog computing. Additionally, clustering the vehicles would be advantageous in order to improve the service availability. As the vehicles become highly connected, trust is needed especially in distributed environments. However, vehicles are made from different manufacturers, and have different platforms, security mechanisms, and varying parking duration. These lead to the unpredictable behavior of the vehicles where quantifying trust value of vehicles would be difficult. A trust-based solution is necessary for task mapping as a task has a set of properties including expected time to complete, and trust requirements that need to be met. However, the existing metrics used for trust evaluation in the vehicular fog computing such as velocity and direction are not applicable in the off-street vehicle fog environments. In this paper, we propose a framework for quantifying the trust value of off-street vehicle fog computing facilities in 5G networks and forming logical clusters of vehicles based on the trust values. This allows tasks to be shared with multiple vehicles in the same cluster that meets the tasks’ trust requirements. Further, we propose a novel task mapping algorithm to increase the vehicle resource utilization and meet the desired trust requirements while maintaining imposed latency requirements of 5G applications. Results obtained using iFogSim simulator demonstrate that the proposed solution increases vehicle resource utilization and reduces task drop noticeably.     Proteomic analysis identifies interleukin 11 regulated plasma membrane proteins in human endometrial epithelial cells in vitro

    <p>Abstract</p> <p>Background</p> <p>During the peri-implantation period, the embryo adheres to an adequately prepared or receptive endometrial surface epithelium. Abnormal embryo adhesion to the endometrium results in embryo implantation failure and infertility. Endometrial epithelial cell plasma membrane proteins critical in regulating adhesion may potentially be infertility biomarkers or targets for treating infertility. Interleukin (IL) 11 regulates human endometrial epithelial cells (hEEC) adhesion. Its production is abnormal in women with infertility. The objective of the study was to identify IL11 regulated plasma membrane proteins in hEEC <it>in vitro </it>using a proteomic approach.</p> <p>Methods</p> <p>Using a 2D-differential in-gel electrophoresis (DIGE) electrophoresis combined with LCMS/MS mass spectrometry approach, we identified 20 unique plasma membrane proteins differentially regulated by IL11 in ECC-1 cells, a hEEC derived cell line. Two IL11 regulated proteins with known roles in cell adhesion, annexin A2 (ANXA2) and flotillin-1 (FLOT1), were validated by Western blot and immunocytochemistry in hEEC lines (ECC-1 and an additional cell line, Ishikawa) and primary hEEC. Flotilin-1 was further validated by immunohistochemistry in human endometrium throughout the menstrual cycle (<it>n = 6-8/cycle</it>).</p> <p>Results</p> <p>2D-DIGE analysis identified 4 spots that were significantly different between control and IL11 treated group. Of these 4 spots, there were 20 proteins that were identified with LCMS/MS. Two proteins; ANXA2 and FLOT1 were chosen for further analyses and have found to be significantly up-regulated following IL11 treatment. Western blot analysis showed a 2-fold and a 2.5-fold increase of ANXA2 in hEEC membrane fraction of ECC-1 and Ishikawa cells respectively. Similarly, a 1.8-fold and a 2.3/2.4-fold increase was also observed for FLOT1 in hEEC membrane fraction of ECC-1 and Ishikawa cells respectively. <it>In vitro</it>, IL11 induced stronger ANXA2 expression on cell surface of primary hEEC and ECC-1 whilst, the lipid-raft protein FLOT1 demonstrated punctate staining in the apical surface of ECC-1 plasma membranes and was upregulated in the epithelium in the receptive phase of the menstrual cycle (p lower or equal 0.05).</p> <p>Conclusions</p> <p>This is the first study to use a proteomics approach to identify hEEC plasma membrane proteins that may be useful as infertility markers or pharmacological targets for fertility regulation.</p

    We find sharp bounds for the norm inequality on a Pseudo-hermitian manifold, where the L^2 norm of all second derivatives of the function involving horizontal derivatives is controlled by the L^2 norm of the sub-Laplacian. Perturbation allows us to get a-priori bounds for solutions to sub-elliptic PDE in non-divergence form with bounded measurable coefficients. The method of proof is through a Bochner technique. The Heisenberg group is seen to be en extremal manifold for our inequality in the class of manifolds whose Ricci curvature is non-negative.Comment: 13 page

    Evaluating TCP Performance of Routing Protocols for Traffic Exchange in Street-parked Vehicles based Fog Computing Infrastructure

    As most vehicles remain parked 95% of its time, this suggests that leveraging the use of On-board Units (OBUs) in parked vehicles would provide communication and computation services to other mobile and fixed nodes for de- livery of services such as multimedia streaming, data storage and data processing. The nearby vehicles can form an infrastructure using IEEE 802.11p communication interface, facilitating communication, computation and storage services to the end users. We refer to this as a Vehicular Fog Computing (VFC) infrastructure. In this study, using NS-2 simulator, we investigate how six routing protocols consisting of two proactive routing protocols, Destination Sequence Destination Vector (DSDV) and Fisheye State Routing (FSR); two reactive routing protocols, Ad Hoc On-Demand Distance Vector (AODV) and Dynamic Source Routing (DSR); and two geographic routing protocols, Distance Routing Effect Algorithm for Mobility (DREAM) and Location Aided Routing (LAR) perform when forwarding TCP traffic among the parked vehicles that form a VFC infrastructure in an urban street parking scenario. In order to reflect an urban street parking scenario, we consider a traffic mobility traces that are generated using SUMO in our simulation. To the best of our knowledge, this work is the first effort to understand how vehicle density, vehicle speed and parking duration can influence TCP in an urban street parking scenario when packet forwarding decision is made using proactive, reactive and geographic routing protocols. In our performance evaluation, positive results are observed on the influence of parking duration in parked vehicles as TCP performance in all routing protocols increases with longer parking duration. However, variable speed in parked vehicles and moving vehicles in an urban street parking scenario may not have significant influence on TCP performance, especially in case of reactive and proactive routing protocols. Further, our findings reveal that vehicle density in a VFC infrastructure can noticeably influence TCP performance. Towards the end of the paper, we delineate some important future research issues in order to improve routing performance in a street-parked vehicle based VFC infrastructure

    Community-centred interventions for improving public mental health among adults from ethnic minority populations in the UK: A scoping review

    OBJECTIVES: Undertake a scoping review to determine the effectiveness of community-centred interventions designed to improve the mental health and well-being of adults from ethnic minority groups in the UK. METHODS: We searched six electronic academic databases for studies published between January 1990 and September 2019: Medline, Embase, PsychINFO, Scopus, CINAHL and Cochrane. For intervention description and data extraction we used the Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews checklist and Template for Intervention Description and Replication guide. Quality was assessed using Cochrane risk of bias tools. Grey literature results were deemed beyond the scope of this review due to the large number of interventions and lack of available outcomes data. RESULTS: Of 4501 studies, 7 met the eligibility criteria of UK-based community interventions targeting mental health in adults from ethnic minority populations: four randomised controlled trials, one pre/post-pilot study, one cross-sectional study and one ethnographic study. Interventions included therapy-style sessions, peer-support groups, educational materials, gym access and a family services programme. Common components included a focus on tackling social isolation, using lay health workers from within the community, signposting and overcoming structural barriers to access. Four studies reported a statistically significant positive effect on mental health outcomes and six were appraised as having a high risk of bias. Study populations were ethnically heterogeneous and targeted people mainly from South Asia. No studies examined interventions targeting men. CONCLUSIONS: There is a paucity of high-quality evidence regarding community-centred interventions focused on improving public mental health among ethnic minority groups. Decision makers need scientific evidence to inform effective approaches to mitigating health disparities. Our next steps are to map promising community activities and interventions that are currently being provided to help identify emerging evidence

    Validity and reliability of the newly developed Malay-language health belief of bloating (HB-Bloat) scale

    Copyright © 2020 by the authors. Abdominal bloating (AB), a common complaint that affects quality of life and disturbs psychological well-being, is largely a behavioral-driven disorder. We aimed to develop and validate a new health belief of bloating (HB-Bloat) scale in the Malay language. The initial item pool was developed based on the theory of planned behavior, empirical literatures, expert review and in-depth interviews. Using the population with bloating (diagnosed based on the Rome IV criteria and pictogram), exploratory and confirmatory factor analytical approaches (EFA and CFA, respectively) were utilized to explore and confirm the domains in the new scale. There were 150 and 323 respondents in the EFA and CFA, respectively. There were 45 items in the initial scale, but it was reduced to 32 items after content validity and pre-testing. In EFA, 17 items with three (3) structure factors (attitude 4 items, subjective norm 7 items, and perceived behavior control 6 items) were identified. Total variance explained by the EFA model was 40.92%. The Cronbach alpha of the three (3) factors ranged from 0.61 to 0.79. With CFA, the three factors model was further tested. Five problematic items were identified and removed. The final measurement model fit the data well (root mean square error of approximation (RMSEA (90% CI) = 0.054 (0.038, 0.070), Comparative Fit Index (CFI) = 0.941, Tucker–Lewis Fit Index (TLI) = 0.924, and standardized root mean squared residual (SRMR) = 0.044). The construct reliability of the final measurement model ranged from 0.76 to 0.84.
