    First-principles study of lattice instabilities in the ferromagnetic martensite Ni2_2MnGa

    The phonon dispersion relations and elastic constants for ferromagnetic Ni2_2MnGa in the cubic and tetragonally distorted Heusler structures are computed using density-functional and density-functional perturbation theory within the spin-polarized generalized-gradient approximation. For 0.9<c/a<1.060.9<c/a<1.06, the TA2_2 tranverse acoustic branch along [110][110] and symmetry-related directions displays a dynamical instability at a wavevector that depends on c/ac/a. Through examination of the Fermi-surface nesting and electron-phonon coupling, this is identified as a Kohn anomaly. In the parent cubic phase the computed tetragonal shear elastic constant, C^\prime=(C11_{11}-C12_{12})/2, is close to zero, indicating a marginal elastic instability towards a uniform tetragonal distortion. We conclude that the cubic Heusler structure is unstable against a family of energy-lowering distortions produced by the coupling between a uniform tetragonal distortion and the corresponding [110][110] modulation. The computed relation between the c/ac/a ratio and the modulation wavevector is in excellent agreement with structural data on the premartensitic (c/ac/a = 1) and martensitic (c/ac/a = 0.94) phases of Ni2_2MnGa.Comment: submitted to Phys.     Approximation Algorithms for Scheduling with Reservations

    We study the problem of non-preemptively scheduling n independent sequential jobs on a system of m identical parallel machines in the presence of reservations. This setting is practically relevant because for various reasons, some machines may not be available during specified time intervals. The objective is to minimize the makespan Cmax, which is the maximum completion time. The general case of the problem is inapproximable unless P = NP; hence we study a suitable strongly NP-hard restriction, namely the case where at least one machine is always available. For this setting we contribute approximation schemes, complemeted by inapproximability results. The approach is based on algorithms for multiple subset sum problems; our technique yields a PTAS which is best possible in the sense that an FPTAS is ruled out unless P = NP. The PTAS presented here is the rst one for the problem under consideration; so far, not even for well-know special cases approximation schemes have been proposed. Furthermore we derive a low cost algorithm with a vonstant approximation ratio and discuss FPTAes for special cases as well as the complexity of the problem if m is part of the input

    Development and external validation of a deep learning algorithm for prognostication of cardiovascular outcomes

    Background and Objectives: We aim to explore the additional discriminative accuracy of a deep learning (DL) algorithm using repeated-measures data for identifying people at high risk for cardiovascular disease (CVD), compared to Cox hazard regression. Methods: Two CVD prediction models were developed from National Health Insurance Service-Health Screening Cohort (NHIS-HEALS): A Cox regression model and a DL model. Performance of each model was assessed in the internal and 2 external validation cohorts in Koreans (National Health Insurance Service-National Sample Cohort; NHIS-NSC) and in Europeans (Rotterdam Study). A total of 412,030 adults in the NHIS-HEALS; 178,875 adults in the NHIS-NSC; and the 4,296 adults in Rotterdam Study were included. Results: Mean ages was 52 years (46% women) and there were 25,777 events (6.3%) in NHIS-HEALS during the follow-up. In internal validation, the DL approach demonstrated a C-statistic of 0.896 (95% confidence interval, 0.886-0.907) in men and 0.921 (0.908-0.934) in women and improved reclassification compared with Cox regression (net reclassification index [NRI], 24.8% in men, 29.0% in women). In external validation with NHIS-NSC, DL demonstrated a C-statistic of 0.868 (0.860-0.876) in men and 0.889 (0.876-0.898) in women, and improved reclassification compared with Cox regression (NRI, 24.9% in men, 26.2% in women). In external validation applied to the Rotterdam Study, DL demonstrated a C-statistic of 0.860 (0.824-0.897) in men and 0.867 (0.830-0.903) in women, and improved reclassification compared with Cox regression (NRI, 36.9% in men, 31.8% in women). Conclusions: A DL algorithm exhibited greater discriminative accuracy than Cox model approaches

    Universal Sequencing on a Single Machine

    We consider scheduling on an unreliable machine that may experience unexpected changes in processing speed or even full breakdowns. We aim for a universal solution that performs well without adaptation for any possible machine behavior. For the objective of minimizing the total weighted completion time, we design a polynomial time deterministic algorithm that finds a universal scheduling sequence with a solution value within 4 times the value of an optimal clairvoyant algorithm that knows the disruptions in advance. A randomized version of this algorithm attains in expectation a ratio of e. We also show that both results are best possible among all universal solutions. As a direct consequence of our results, we answer affirmatively the question of whether a constant approximation algorithm exists for the offline version of the problem when machine unavailability periods are known in advance. When jobs have individual release dates, the situation changes drastically. Even if all weights are equal, there are instances for which any universal solution is a factor of Ω(log n/ log log n) worse than an optimal sequence. Motivated by this hardness, we study the special case when the processing time of each job is proportional to its weight. We present a non-trivial algorithm with a small constant performance guarantee. © 2010 Springer-Verlag

    Management and outcomes of patients with left atrial appendage thrombus prior to percutaneous closure

Data regarding its management are very limited. The aim of this study was to analyse the medical and invasive treatment of patients referred for LAAC in the presence of LAA thrombus. Methods: This multicentre observational registry included 126 consecutive patients referred for LAAC with LAA thrombus on preprocedural imaging. Treatment strategies included intensification of antithrombotic therapy (IAT) or direct LAAC. The primary and secondary endpoints were a composite of bleeding, stroke and death at 18 months, and procedural success, respectively. Results: IAT was the preferred strategy in 57.9% of patients, with total thrombus resolution observed in 60.3% and 75.3% after initial and subsequent IAT, respectively. Bleeding complications and stroke during IAT occurred in 9.6% and 2.9%, respectively, compared with 3.8% bleeding and no embolic events in the direct LAAC group before the procedure. Procedural success was 90.5% (96.2% vs 86.3% in direct LAAC and IAT group, respectively, p=0.072), without cases of in-hospital thromboembolic complications. The primary endpoint occurred in 29.3% and device-related thrombosis was found in 12.8%, without significant difference according to treatment strategy. Bleeding complications at 18 months occurred in 22.5% vs 10.5% in the IAT and direct LAAC group, respectively (p=0.102). Conclusion: In the presence of LAA thrombus, IAT was the initial management strategy in half of our cohort, with initial thrombus resolution in 60% of these, but with a relatively high bleeding rate (∼10%). Direct LAAC was feasible, with high procedural success and absence of periprocedural embolic complications. However, a high rate of device-related thrombosis was detected during follow-up

    Genomic analysis of diet composition finds novel loci and associations with health and lifestyle

    We conducted genome-wide association studies (GWAS) of relative intake from the macronutrients fat, protein, carbohydrates, and sugar in over 235,000 individuals of European ancestries. We identified 21 unique, approximately independent lead SNPs. Fourteen lead SNPs are uniquely associated with one macronutrient at genome-wide significance (P < 5 × 10−8), while five of the 21 lead SNPs reach suggestive significance (P < 1 × 10−5) for at least one other macronutrient. While the phenotypes are genetically correlated, each phenotype carries a partially unique genetic architecture. Relative protein intake exhibits the strongest relationships with poor health, including positive genetic associations with obesity, type 2 diabetes, and heart disease (rg ≈ 0.15–0.5). In contrast, relative carbohydrate and sugar intake have negative genetic correlations with waist circumference, waist-hip ratio, and neighborhood deprivation (|rg| ≈ 0.1–0.3) and positive genetic correlations with physical activity (rg ≈ 0.1 and 0.2). Relative fat intake has no consistent pattern of genetic correlations with poor health but has a negative genetic correlation with educational attainment (rg ≈−0.1). Although our analyses do not allow us to draw causal conclusions, we find no evidence of negative health consequences associated with relative carbohydrate, sugar, or fat intake. However, our results are consistent with the hypothesis that relative protein intake plays a role in the etiology of metabolic dysfunction

    Trans-ancestry genome-wide association study identifies 12 genetic loci influencing blood pressure and implicates a role for DNA methylation

    We carried out a trans-ancestry genome-wide association and replication study of blood pressure phenotypes among up to 320,251 individuals of East Asian, European and South Asian ancestry. We find genetic variants at 12 new loci to be associated with blood pressure (P = 3.9 &times; 10-11 to 5.0 &times; 10-21). The sentinel blood pressure SNPs are enriched for association with DNA methylation at multiple nearby CpG sites, suggesting that, at some of the loci identified, DNA methylation may lie on the regulatory pathway linking sequence variation to blood pressure. The sentinel SNPs at the 12 new loci point to genes involved in vascular smooth muscle (IGFBP3, KCNK3, PDE3A and PRDM6) and renal (ARHGAP24, OSR1, SLC22A7 and TBX2) function. The new and known genetic variants predict increased left ventricular mass, circulating levels of NT-proBNP, and cardiovascular and all-cause mortality (P = 0.04 to 8.6 &times; 10-6). Our results provide new evidence for the role of DNA methylation in blood pressure regulation
