Characterization and potential roles of calretinin in rodent spermatozoa
Calretinin has been detected in various excitable cells but the presence and putative roles of such a calcium-binding protein has never been characterized in sperm.Epididymal spermatozoa were collected from C57Bl6 (wild-type, WT) or calretinin knockout (CRâ/â) mice and Wistar rats. A specific staining for calretinin was detected by immunofluorescence in the principal piece of the flagellum, both in WT mouse and rat spermatozoa. Western blots confirmed the expression of calretinin in rat and WT spermatozoa as well as its absence in CRâ/â mice.No significant difference was observed in the spontaneous acrosome reaction between WT and CRâ/â sperm. The addition of the calcium-ionophore A-23187, Thapsigargin or Progesterone to WT or CRâ/â incubated spermatozoa induced increases in the acrosome reaction but the stimulatory effects were identical in both genotypes. Motility measurements assessed by computer-assisted sperm analysis indicated that, under basal non-stimulatory conditions, CR-/- sperm exhibited a lower curvilinear velocity and a smaller lateral head movement amplitude, although no difference was observed for the beat cross frequency. After incubation with 25âŻmM NH4Cl, the curvilinear velocity, the amplitude of the lateral head movement and the hyperactivation were increased, while the beat cross frequency was decreased, in both genotypes.Evaluation of the in vivo fertility potential indicated that the CRâ/â litter sizes were clearly reduced compared to the WT litter sizes. Our study describes, for the first time, the expression of calretinin in sperm. These data extend the potential implication of calcium-binding proteins in the sperm calcium-signaling cascade and bring new insights into the understanding of sperm physiology
BeatWalk: Personalized Music-Based Gait Rehabilitation in Parkinsonâs Disease
Taking regular walks when living with Parkinsonâs disease (PD) has beneficial effects on movement and quality of life. Yet, patients usually show reduced physical activity compared to healthy older adults. Using auditory stimulation such as music can facilitate walking but patients vary significantly in their response. An individualized approach adapting musical tempo to patientsâ gait cadence, and capitalizing on these individual differences, is likely to provide a rewarding experience, increasing motivation for walk-in PD. We aim to evaluate the observance, safety, tolerance, usability, and enjoyment of a new smartphone application. It was coupled with wearable sensors (BeatWalk) and delivered individualized musical stimulation for gait auto-rehabilitation at home. Forty-five patients with PD underwent a 1-month, outdoor, uncontrolled gait rehabilitation program, using the BeatWalk application (30 min/day, 5 days/week). The music tempo was being aligned in real-time to patientsâ gait cadence in a way that could foster an increase up to +10% of their spontaneous cadence. Open-label evaluation was based on BeatWalk use measures, questionnaires, and a six-minute walk test. Patients used the application 78.8% (±28.2) of the prescribed duration and enjoyed it throughout the program. The application was considered âeasy to useâ by 75% of the patients. Pain, fatigue, and falls did not increase. Fear of falling decreased and quality of life improved. After the program, patients improved their gait parameters in the six-minute walk test without musical stimulation. BeatWalk is an easy to use, safe, and enjoyable musical application for individualized gait rehabilitation in PD. It increases âwalk for exerciseâ duration thanks to high observance. This research was supported by a European grant: BeatHealth: Health and Wellness on the Beat for VC, DD, CL, AGi, VD, RV, EH, ED, ML, BB, and SB (EU FP7-ICT contract #610633)
Insomnia in Parkinson's Disease : from the study of its psychological factors of maintenance to its management
The aim of the present thesis is twofold: on the one hand, to investigate the role of cognitive and behavioral factors as processes underlying the maintenance of chronic insomnia associated with Parkinson's disease (PD); on the other hand, to evaluate the acceptability and efficacy of Cognitive Behavioral Therapy (CBT) for the management of chronic insomnia comorbid to PD. A first psychometric study (Study 1) was conducted with a large sample of 539 individuals from the general population in order to propose an adaptation and validation of a French version of the Sleep-Related Behaviors Questionnaire (SRBQ). This questionnaire measures the frequency of use of sleep-related safety behaviors, a central component in maintaining chronic insomnia. Preliminary analyses resulted in a French version consisting of 20 items (SRBQ-20) with a very good internal consistency. The exploratory factor and parallel analyses suggested a solution with 3 factors: improvement of fatigue, improvement of sleep and suppression of thoughts. Correlation analyses showed a significant relationship between dysfunctional beliefs, insomnia severity and total SRBQ-20 score. The French version of the SRBQ-20 has good psychometric properties. A second case-control study (Study 2) was conducted to determine, using Harvey's (2002) model, the relationships between psychological factors maintaining insomnia and chronic insomnia associated with PD. In addition, we studied the interaction between these factors in their contribution to chronic insomnia diagnosis associated with PD. Sixty-eight patients with PD (38 with chronic insomnia) were enrolled...
