    Ricerca di evolved blue straggler stars in M80

    Questo elaborato di tesi si inserisce nel contesto della ricerca di stelle Evolved Blue Straggler Stars (E-BSS) in ammassi globulari Galattici. Le BSS sono ritenute essere significativamente più massicce delle stelle che tipicamente popolano un ammasso stellare. Questi oggetti sono facilmente identificabili durante la loro fase di bruciamento dell'idrogeno, al contrario è impossibile distinguerli fotometricamente, in modo certo, dalle altre stelle di minore massa del sistema che li ospita durante le fasi evolutive successive. In questo lavoro è stato applicato il metodo spettroscopico utilizzato da Ferraro et al. (2016) per identificare una E-BSS nell'ammasso globulare 47 Tuc e basato sul confronto tra le abbondanze chimiche derivate da righe spettrali neutre e ionizzate. Tale metodo permette di ricavare una stima della gravità a superficiale e, quindi, della massa stellare. In particolare, sono stati analizzati spettri ad alta risoluzione, acquisiti con lo spettrografo ad alta risoluzione UVES, di un campione 13 stelle candidate E-BSS dell'ammasso globulare M80 situate nella regione del CMD tra l'HB e l'attacco dell'AGB. I principali risultati di quest'analisi hanno mostrato che nel campione analizzato non vi sono E-BSS particolarmente massicce e, inoltre, hanno evidenziato una sottostima sistematica dell'abbondanza chimica ricavata da righe neutre in tutte le stelle di post-RGB, mentre le righe ionizzate risultano il diagnostico più u robusto ed affidabile per derivare le abbondanze chimiche

    Exploring the internal kinematics of Galactic Globular Cluster cores

    Globular clusters (GCs) are traditionally described as simple quasi-relaxed non-rotating stellar systems, characterized by spherical symmetry and isotropy in velocity space. However, recent studies have shown deviations from isotropic velocity distributions and significant internal rotation in many GCs, suggesting that their internal structure and kinematics are more complex than previously thought. The aim of this thesis is to investigate the internal kinematics of Galactic Globular Clusters (GGCs) as part of the Multi-Instrument Kinematic Survey (MIKiS), which exploits the capabilities of different ESO-VLT spectrographs to obtain comprehensive velocity dispersion (VD) and rotation profiles of GGCs. Moreover, this thesis has the particular goal of unraveling the kinematics of GC cores, which are still largely unexplored, by taking advantage of the exceptional spatial resolution of the adaptive-optics assisted integral-field spectrograph MUSE/NFM. The thesis presents a thorough kinematic study of three GGCs NGC 1904, NGC 6440, and NGC 6569. By combining the data sets acquired with four different spectrographs, we obtained the radial velocity (RV) of more than 1000 individual stars in each cluster, sampling from the innermost to the outermost regions. This allowed us to obtain the entire VD profile of each cluster and exclude the presence of an intermediate-mass black hole in the core of NGC 1904, at odds with previous findings obtained from integrated-light spectra. The studies also revealed signatures of internal rotation in each of the GCs studied. These results, supported by those of N-body simulations, prove that GCs were born with a significant initial rotation that they gradually lost through internal two-body relaxation and angular momentum loss carried away by escaping stars. Furthermore, we derived the structural parameters of NGC 6440 and NGC 6569, obtaining a comprehensive overview of the internal kinematics and structure of these GCs, which is necessary to properly reconstruct the evolutionary history of these systems

    Internal kinematics and structure of the bulge globular cluster NGC 6569

    In the context of a project aimed at characterizing the properties of star clusters in the Galactic bulge, here we present the determination of the internal kinematics and structure of the massive globular cluster NGC 6569. The kinematics has been studied by means of an unprecedented spectroscopic dataset acquired in the context of the ESO-VLT Multi-Instrument Kinematic Survey (MIKiS) of Galactic globular clusters, combining the observations from four different spectrographs. We measured the line-of-sight velocity of a sample of almost 1300 stars distributed between ~0.8" and 770" from the cluster center. From a sub-sample of high-quality measures, we determined the velocity dispersion profile of the system over its entire radial extension (from ~ 5" to ~ 200" from the center), finding the characteristic behavior usually observed in globular clusters, with a constant inner plateau and a declining trend at larger radii. The projected density profile of the cluster has been obtained from resolved star counts, by combining high-resolution photometric data in the center, and the Gaia EDR3 catalog radially extended out to ~20' for a proper sampling of the Galactic field background. The two profiles are properly reproduced by the same King model, from which we estimated updated values of the central velocity dispersion, main structural parameters (such as the King concentration, the core, half-mass, and tidal radii), total mass, and relaxation times. Our analysis also reveals a hint of ordered rotation in an intermediate region of the cluster (40"<r<90", corresponding to 2rc<r<4.5rc 2 r_c<r<4.5 r_c), but additional data are required to properly assess this possibility.Comment: Accepted for publication in The Astrophysical Journal; 21 pages, 10 figures, 4 table

    JWST uncovers helium and water abundance variations in the bulge globular cluster NGC 6440

    We used ultra-deep observations obtained with the NIRCam aboard the James Webb Space Telescope to explore the stellar population of NGC 6440: a typical massive, obscured and contaminated globular cluster formed and orbiting within the Galactic bulge. Leveraging the exceptional capabilities of this camera, we sampled the cluster down to ~5 magnitudes below the main-sequence turn-off in the (mF115W , mF115W - mF200W ) colour-magnitude diagram. After carefully accounting for differential extinction and contamination by field interlopers, we find that the main sequence splits into two branches both above and below the characteristic knee. By comparing the morphology of the colour-magnitude diagram with a suitable set of isochrones, we argue that the upper main-sequence bi-modality is likely due to the presence of a He-enriched stellar population with a helium spread of DeltaY = 0.04. The lower main-sequence bi-modality can be attributed to variations in the abundance of water (i.e., oxygen) with Delta[O/Fe] ~ -0.4. This is the first evidence of both helium and oxygen abundance variations in a globular cluster purely based on JWST observations. These results open the window for future in-depth investigations of the multiple population phenomenon in clusters located in the Galactic bulge, which were previously unfeasible with near-UV observations, due to prohibitive reddening and crowding conditions.Comment: Accepted for publication in A&

    Variability in genes regulating vitamin D metabolism is associated with vitamin D levels in type 2 diabetes

    Mortality rate is increased in type 2 diabetes (T2D). Low vitamin D levels are associated with increased mortality risk in T2D. In the general population, genetic variants affecting vitamin D metabolism (DHCR7 rs12785878, CYP2R1 rs10741657, GC rs4588) have been associated with serum vitamin D. We studied the association of these variants with serum vitamin D in 2163 patients with T2D from the "Sapienza University Mortality and Morbidity Event Rate (SUMMER) study in diabetes". Measurements of serum vitamin D were centralised. Genotypes were obtained by Eco™ Real-Time PCR. Data were adjusted for gender, age, BMI, HbA1c, T2D therapy and sampling season. DHCR7 rs12785878 (p = 1 x 10-4) and GC rs4588 (p = 1 x 10-6) but not CYP2R1 rs10741657 (p = 0.31) were significantly associated with vitamin D levels. One unit of a weighted genotype risk score (GRS) was strongly associated with vitamin D levels (p = 1.1 x 10-11) and insufficiency (&lt;30 ng/ml) (OR, 95%CI = 1.28, 1.16-1.41, p = 1.1 x 10-7). In conclusion, DHCR7 rs12785878 and GC rs4588, but not CYP2R1 rs10741657, are significantly associated with vitamin D levels. When the 3 variants were considered together as GRS, a strong association with vitamin D levels and vitamin D insufficiency was observed, thus providing robust evidence that genes involved in vitamin D metabolism modulate serum vitamin D in T2D

    Effects of Pin1 Loss in Hdh(Q111) Knock-in Mice

    Huntington's disease (HD) is a fatal, dominantly inherited, neurodegenerative disorder due to a pathological expansion of the CAG repeat in the coding region of the HTT gene. In the quest for understanding the molecular basis of neurodegeneration, we have previously demonstrated that the prolyl isomerase Pin1 plays a crucial role in mediating p53-dependent apoptosis triggered by mutant huntingtin (mHtt) in vitro. To assess the effects of the lack of Pin1 in vivo, we have bred Pin1 knock-out mice with Hdh(Q111) knock-in mice, a genetically precise model of HD. We show that Pin1 genetic ablation modifies a portion of Hdh(Q111) phenotypes in a time-dependent fashion. As an early event, Pin1 activity reduces the DNA damage response (DDR). In midlife mice, by taking advantage of next-generation sequencing technology, we show that Pin1 activity modulates a portion of the alterations triggered by mHtt, extending the role of Pin1 to two additional Hdh(Q111) phenotypes: the unbalance in the "synthesis/concentration of hormones", as well as the alteration of "Wnt/\u3b2-catenin signaling". In aging animals, Pin1 significantly increases the number of mHtt-positive nuclear inclusions while it reduces gliosis.     Promising prognostic value of Transglutaminase type 2 and its correlation with tumor-infiltrating immune cells in skin cutaneous melanoma

    Tissue Transglutaminases (TGs) are crosslinking enzymes with pleiotropic functions that have been linked to the development and progression of numerous cancers, with a recent focus on their ability to remodel the tumor microenvironment. Although several pieces of evidence demonstrated their importance in the regulation of the major signaling pathways that control oncogenesis, the correlation between TGs with clinical and pathological features remains controversial and to be further explored. Moreover, an assessment of the TGs alterations together with a functional analysis associated with clinical features and prognostic values are still lacking and would help to understand these intricacies, particularly in human cancers. In the present study, we processed data from numerous public datasets to investigate TGs distribution and prognostic signature in cancer patients. Here, we found that skin cutaneous melanoma (SKCM) shows the highest abundance of TGs mutations among the other human cancers. Interestingly, among all the TGs, TG2 is the only member whose expression is associated with a better overall survival in SKCM, although its expression increases with the worsening of the tumor phenotype. Our analysis revealed a strong positive association between TG2 expression and anti-tumoral immune response, which would explain the relationship between high mRNA levels and better overall survival. Our data suggest that TG2 may be presented as a new promising immune biomarker of prognosis in SKCM, which may contribute to identifying patients who would benefit the most from adjuvant immunotherapy

    A Meta-Analysis Study to Infer Voltage-Gated K+ Channels Prognostic Value in Different Cancer Types

    : Potassium channels are often highly expressed in cancer cells with respect to healthy ones, as they provide proliferative advantages through modulating membrane potential, calcium homeostasis, and various signaling pathways. Among potassium channels, Shaker type voltage-gated Kv channels are emerging as promising pharmacological targets in oncology. Here, we queried publicly available cancer patient databases to highlight if a correlation exists between Kv channel expression and survival rate in five different cancer types. By multiple gene comparison analysis, we found a predominant expression of KCNA2, KCNA3, and KCNA5 with respect to the other KCNA genes in skin cutaneous melanoma (SKCM), uterine corpus endometrial carcinoma (UCEC), stomach adenocarcinoma (STAD), lung adenocarcinoma (LUAD), and lung squamous cell carcinoma (LUSC). This analysis highlighted a prognostic role of KCNA3 and KCNA5 in SKCM, LUAD, LUSC, and STAD, respectively. Interestingly, KCNA3 was associated with a positive prognosis in SKCM and LUAD but not in LUSC. Results obtained by the analysis of KCNA3-related differentially expressed genes (DEGs); tumor immune cell infiltration highlighted differences that may account for such differential prognosis. A meta-analysis study was conducted to investigate the role of KCNA channels in cancer using cancer patients' datasets.
