    Nature read in tooth and fur: non-invasive sampling of the Pygmy Shrew (Sorex minutus Linnaeus, 1766) on Lundy for population genetics

    Britain’s smallest mammal, the pygmy shrew (Sorex minutus Linnaeus, 1766) is the only long established terrestrial mammal on Lundy. Little is known about this species’ mode of colonisation or its population structure on the island. Genetic analyses might provide useful insights in this respect, but DNA samples are difficult to obtain because pygmy shrews can be harmed by sampling methods. Two non invasive methods for obtaining DNA sequences were tested. DNA was extracted from hair samples and from a mandible derived from a bird-pellet. A 612 base pair fragment of the cytochrome oxidase b gene was PCR amplified and sequenced. To investigate the origin of pygmy shrew on Lundy the sequences were analysed in the context of published data. The sequences are most similar to those from the U.K. mainland which supports a British origin of this population

    Structure determination of Split-soret Cytochrome from a Desulfovibrio species isolated from a human abdominal abcess

    The determined structure of the split-soret cytochrome (SSC) isolated from Desulfovibrio desulfuricans ATCC 27774 (D.d.) revealed a new Heme arrangement, which suggests that this protein constitutes a new cytochrome class.. SSC is a 52.6kDa homodimer containing four hemes at one end of the molecule. In each monomer the two hemes have their edges overlapped within van der Waals contacts. The polypeptide chain of each monomer supplies the sixth ligand to the heme-iron of the other monomer. A similar protein was recently purified from a homologous Desulfovibrio clinical strain isolated from an abdominal wall abscess in human patient2. Crystals of this SSC were grown using vapour diffusion method in the presence of agarose gel. Diffraction data were collected using X-ray synchrotron radiation at the ESRF, beamline, ID 14-1. The structure will be solved by molecular replacement using the structure of the D.d. as a starting model

    Basement control on dyke distribution in Large Igneous Provinces: Case study of the Karoo triple junction.

    Continental flood basalts consist of vast quantities of lava, sills and giant dyke swarms that are associated with continental break-up. The commonly radiating geometry of dyke swarms in these provinces is generally interpreted as the result of the stress regime that affected the lithosphere during the initial stage of continental break-up or as the result of plume impact. On the other hand, structures in the basement may also control dyke orientations, though such control has not previously been documented. In order to test the role of pre-dyke structures, we investigated four major putative Karoo-aged dyke swarms that taken together represent a giant radiating dyke swarm (the so-called "triple-junction") ascribed to the Jurassic Karoo continental flood basalt (> 3x10 6 km2; southern Africa). One of the best tests to discriminate between neoformed and inherited dyke orientation is to detect Precambrian dykes in the Jurassic swarms. Accordingly, we efficiently distinguished between Jurassic and Precambrian dykes using abbreviated low resolution, 40Ar/39Ar incremental heating schedules. Save-Limpopo dyke swarm samples (n = 19) yield either apparent Proterozoic (728-1683 Ma) or Mesozoic (131-179 Ma) integrated ages; the Olifants River swarm (n = 20) includes only Proterozoic (851-1731 Ma) and Archaean (2470-2872 Ma) dykes. The single age obtained on one N-S striking dyke (1464 Ma) suggests that the Lebombo dyke swarm includes Proterozoic dykes in the basement as well. These dates demonstrate the existence of pre-Karoo dykes in these swarms as previously hypothesized without supporting age data.In addition, aeromagnetic and air-photo interpretations indicate that: (1) dyke emplacement was largely controlled by major discontinuities such as the Zimbabwe and Kaapvaal craton boundaries, the orientation of the Limpopo mobile belt, and other pre-dyke structures including shear zones and (2) considering its polygenetic, pre-Mesozoic origin, the Olif ants River dyke swarm cannot be considered part of the Karoo magmatic event.This study, along with previous results obtained on the Okavango dyke swarm, shows that the apparent "triple junction" formed by radiating dyke swarms is not a Jurassic structure; rather, it reflects weakened lithospheric pathways that have controlled dyke orientations over hundreds of millions of years. One consequence is that the "triple-junction" geometry can no longer be unambiguously used as a mantle plume marker as previously proposed, although it does not preclude the possible existence of a mantle plume. More generally, we suggest that most Phanerozoic dyke swarms (including triple junctions) related to continental flood basalts were probably controlled in part by pre-existing lithospheric discontinuities

    Studies on the redox centers of the terminal oxidase from Desulfovibrio gigas and evidence for its interaction with rubredoxin

    Abstract: Rubredoxin-oxygen oxidoreductase (ROO) is the final component of a soluble electron transfer chain that couples NADH oxidation to oxygen consumption in the anaerobic sulfate reducer Desulfovibrio gigas. It is an 86-kDa homodimeric flavohemeprotein containing two FAD molecules, one mesoheme IX, and one Fe-uroporphyrin I per monomer, capable of fully reducing oxygen to water. EPR studies on the native enzyme reveal two components with g values at similar to 2.46, 2.29, and 1.89, which are assigned to low spin hemes and are similar to the EPR features of P-450 hemes, suggesting that ROO hemes have a cysteinyl axial ligation. At pH 7.6, the flavin redox transitions occur at 0 +/- 15 mV for the quinone/semiquinone couple and at -130 +/- 15 mV for the semiquinone/hydroquinone couple; the hemes reduction potential is -350 +/- 15 mV. Spectroscopic studies provided unequivocal evidence that the flavins are the electron acceptor centers from rubredoxin, and that their reduction proceed through an anionic semiquinone radical. The reaction with oxygen occurs in the flavin moiety. These data are strongly corroborated by the finding that rubredoxin and ROO are located in the same polycistronic unit of D. gigas genome. For the first time, a clear role for a rubredoxin in a sulfate-reducing bacterium is presented

    The ‘strict’ anaerobe Desulfovibrio gigas contains a membrane-bound oxygen-reducing respiratory chain

    AbstractSulfate-reducing bacteria are considered as strict anaerobic microorganisms, in spite of the fact that some strains have been shown to tolerate the transient presence of dioxygen. This report shows that membranes from Desulfovibrio gigas grown in fumarate/sulfate contain a respiratory chain fully competent to reduce dioxygen to water. In particular, a membrane-bound terminal oxygen reductase, of the cytochrome bd family, was isolated, characterized, and shown to completely reduce oxygen to water. This oxidase has two subunits with apparent molecular masses of 40 and 29 kDa. Using NADH or succinate as electron donors, the oxygen respiratory rates of D. gigas membranes are comparable to those of aerobic organisms (3.2 and 29 nmol O2 min−1 mg protein−1, respectively). This ‘strict anaerobic’ bacterium contains all the necessary enzymatic complexes to live aerobically, showing that the relationships between oxygen and anaerobes are much more complex than originally thought

    Pattern of seat belt use by drivers in Trinidad and Tobago, West Indies

    <p>Abstract</p> <p>Background</p> <p>In Trinidad and Tobago, the law on the mandatory use of seat belts was passed in 1995, but this law is hardly enforced. The objective of this study was to determine the frequency and predictors of seat belt use by motor vehicle drivers in the country.</p> <p>Findings</p> <p>A cross-sectional study of 959 motor vehicle drivers using a self-administered questionnaire. Data analysis included Pearson Chi square test and multinomial logistic regression analysis in order to determine the possible predictors of seat belt use by the drivers in Trinidad and Tobago. A majority of the drivers sometimes (51.8%) or always (31.6%) use a seat belt. About 16.7%, 29% and 54.2% of the drivers perceived that the other drivers use their seat belts more frequently, with the same frequency and less frequently respectively compared to themselves. The main reason for not using seat belt by the drivers was given as frequent stops (40.7%) and the main motivation to use seat belt by the drivers was given as stiffer penalties for non-compliance with the seat belt law (44.5%). The predictors of seat belt use were male driver, no formal or lower level of education, driving for less than 10 years, and the perception that the other drivers use seat belts with the same or higher frequency compared to the respondents.</p> <p>Conclusion</p> <p>Only a small proportion of the drivers in Trinidad and Tobago always use a seat belt when driving. There is the need to enforce the seat belt legislation in the country.</p
