    Combined Effects of Acute Temperature Change and Elevated pCO2 on the Metabolic Rates and Hypoxia Tolerances of Clearnose Skate (Rostaraja eglanteria), Summer Flounder (Paralichthys dentatus), and Thorny Skate (Amblyraja radiata)

    Understanding how rising temperatures, ocean acidification, and hypoxia affect the performance of coastal fishes is essential to predicting species-specific responses to climate change. Although a population’s habitat influences physiological performance, little work has explicitly examined the multi-stressor responses of species from habitats differing in natural variability. Here, clearnose skate (Rostaraja eglanteria) and summer flounder (Paralichthys dentatus) from mid-Atlantic estuaries, and thorny skate (Amblyraja radiata) from the Gulf of Maine, were acutely exposed to current and projected temperatures (20, 24, or 28 °C; 22 or 30 °C; and 9, 13, or 15 °C, respectively) and acidification conditions (pH 7.8 or 7.4). We tested metabolic rates and hypoxia tolerance using intermittent-flow respirometry. All three species exhibited increases in standard metabolic rate under an 8 °C temperature increase (Q10 of 1.71, 1.07, and 2.56, respectively), although this was most pronounced in the thorny skate. At the lowest test temperature and under the low pH treatment, all three species exhibited significant increases in standard metabolic rate (44–105%; p \u3c 0.05) and decreases in hypoxia tolerance (60–84% increases in critical oxygen pressure; p \u3c 0.05). This study demonstrates the interactive effects of increasing temperature and changing ocean carbonate chemistry are species-specific, the implications of which should be considered within the context of habitat.     Systematic Functional Prioritization of Protein Posttranslational Modifications

    Protein function is often regulated by post-translational modifications (PTMs) and recent advances in mass-spectrometry have resulted in an exponential increase in PTM identification. However, the functional significance of the vast majority of these modifications remains unknown. To address this problem, we compiled nearly 200,000 phosphorylation, acetylation and ubiquitination sites from 11 eukaryotic species, including 2,500 novel ubiquitylation sites for S. cerevisiae. We developed methods to prioritize the functional relevance of these PTMs by predicting those that likely participate in cross-regulatory events, regulate domain activity or mediate protein-protein interactions. PTM conservation within domain families identifies regulatory ‘hot-spots’ that overlap with functionally important regions, a concept we experimentally validated on the HSP70 domain family. Finally, our analysis of the evolution of PTM regulation highlights potential routes for neutral drift in regulatory interactions and suggests that only a fraction of modification sites are likely to have a significant biological role

    Accelerating Medicines PartnershipÂź Schizophrenia (AMPÂź SCZ):Rationale and Study Design of the Largest Global Prospective Cohort Study of Clinical High Risk for Psychosis

    This article describes the rationale, aims, and methodology of the Accelerating Medicines PartnershipÂź Schizophrenia (AMPÂź SCZ). This is the largest international collaboration to date that will develop algorithms to predict trajectories and outcomes of individuals at clinical high risk (CHR) for psychosis and to advance the development and use of novel pharmacological interventions for CHR individuals. We present a description of the participating research networks and the data processing analysis and coordination center, their processes for data harmonization across 43 sites from 13 participating countries (recruitment across North America, Australia, Europe, Asia, and South America), data flow and quality assessment processes, data analyses, and the transfer of data to the National Institute of Mental Health (NIMH) Data Archive (NDA) for use by the research community. In an expected sample of approximately 2000 CHR individuals and 640 matched healthy controls, AMP SCZ will collect clinical, environmental, and cognitive data along with multimodal biomarkers, including neuroimaging, electrophysiology, fluid biospecimens, speech and facial expression samples, novel measures derived from digital health technologies including smartphone-based daily surveys, and passive sensing as well as actigraphy. The study will investigate a range of clinical outcomes over a 2-year period, including transition to psychosis, remission or persistence of CHR status, attenuated positive symptoms, persistent negative symptoms, mood and anxiety symptoms, and psychosocial functioning. The global reach of AMP SCZ and its harmonized innovative methods promise to catalyze the development of new treatments to address critical unmet clinical and public health needs in CHR individuals.</p

    Adolescent experiences of violence and relation to violence perpretration beyond young adulthood among an urban sample of black and African American males

    The purpose of this study is to determine if experiences of physical violence during early and late adolescence (12–21 years) places urban Black males at increased risk for interpersonal violence perpetration beyond young adulthood (30 years and older). Participants of this cross-sectional study were Black and African American men (N = 455) between the ages of 30 and 65 years, recruited from four urban clinical sites in the Northeast. Multivariate logistic regression models were used to analyze the relation of adolescent experiences of violence to: (1) past 6 month street violence involvement and (2) past year intimate partner violence perpetration. Ten percent of the sample reported that they experienced adolescent victimization. Men reporting adolescent victimization were significantly more likely to report past 6-month street violence involvement (Adjusted Odds Ratio (AOR) = 3.2, 95 % CI = 1.7–6.3) and past 6 month intimate partner violence perpetration (AOR = 2.8, 95 % CI = 1.8–5.4) compared to men who did not report such victimization. Study findings suggest that in order to prevent adulthood perpetration of violence, more work is needed to address experiences of victimization among young Black males, particularly violence experienced during adolescence

    Hearing aid processing changes tone burst onset: Effect on cortical auditory evoked potentials in individuals with normal audiometric thresholds

    Purpose: The validity of using the cortical auditory evoked potential (CAEP) as an objective measure of hearing aid outcome has been questioned in the literature due to stimulus modifications caused by hearing aid processing. This study aimed to investigate the effects of hearing aid processing on the CAEP elicited with tone bursts that may have altered onsets. Method: CAEPs to unprocessed and hearing aid-processed tone bursts were obtained from 16 individuals with normal audiometric thresholds when the onset time, level, and signalto-noise ratio (SNR) were matched between the 2 conditions. Tone bursts processed by the hearing aid were recorded in an anechoic box and were presented through insert receivers. Unprocessed tone bursts were superimposed with hearing aid noise floor to match the SNR of the hearing aid-processed tone bursts. Results: Shortening of rise time and overshoot at the onset of the tone burst were evident in the hearing aid-processed stimuli. Statistical analysis of data showed no significant effects of hearing aid processing on the latency or amplitude of CAEP peaks (p \u3e.05). Conclusion: The changes in rise time occurring in the tone bursts due to hearing aid processing may not confound CAEP measures that are used to validate hearing aid fitting. © American Speech-Language-Hearing Association
