    Ein Neues Verfahren zur Messung der Bakteriziden Fähigkeit des Vollblutes

    Das oben erwahnte Verfahren hat vor den anderen Methoden besonders die Vorzuge, 1) da&#946; man dadurch zu einem sicheren Resultat gelangen und gleichzeitig auch jedes Datum mit exakten Ziffern zum Ausdruck bringen kann, 2) da&#946; bei diesem Verfahren keineswegs erforderlich ist, eine bestimmte Anzahl von Keimen einschlie&#946;ende Bakterienaufschwemmung herzusteHen und auch Kontrollversuch anzustellen, 3) da&#946; es von den Fehlern des Mischverhaltnisses zwischen der Bakterienlosung und dem Blut nicht so erheblich beeinflu&#946;t wird, und 4) da&#946; man durch dieses Verfahren gleichzeitig mehrere bakterientotende Faktoren untersuchen kann. Ferner hat dieses Verfahren auch den Vorzug, da&#946; es praktisch sehr einfach auszufuhren ist und nur 6 Stunden nach der Blutentnahme bereits das Ergebnis liefert. Es gestattet ferner, die bakterizide Kraft des Blutes gleichzeitig bei 6 - 8 Menschen zu untersuchen, was mich zur Uberzeugung fuhrt, da&#946; es in der Klinik hochgeschatzt werden wird. Auch das Verfahren und die ebenfalls vom mir aufgestellte Formel zur zusammenfassenden Beurteilung kann man nach meinem Erachten durch entsprechende Veranderungen einiger Faktoren ohne jede Schwierigkeiten auch fur andere Bakterienarten anwenden. Man wird wohl gegen eine einzige Lucke dieses Verfahrens, da&#946; die mikroskopische Untersuchung und die Berechnung allzu verwickelt zu sein scheint, Einwand erheben, eine Lucke, zu deren Schlu&#946; jedoch nur eine kurzfristige Ubung erfordert wird, durch welche die mikroskopische Untersuchung innerhalb 30 Minuten, die Berechnung nur in 5 Minuten vollendet werden kann. (Zur Berechnung bedarf es einer Gauss'schen Logarithmentafel.) Obgleich das geschilderte Verfahren noch viele, genauere Prufungen erheischende Punkte in sich einschlie&#946;t, mu&#946; es hier, wenn auch in Grundzugen, jetzt schon angefuhrt werden,. da ich der festen Uberzeugung bin, da&#946; es im Vergleich zu den bisherigen Methoden ein dem wirklichen Wert der Bakterizidie des Vollblutes im lebenden Organismus viel naheres Resultat liefert.</p

    Occurrence of 2n microspore production in diploid interspecific hybrids between the wild parental species of peanut (Arachis hypogaea L., Leguminosae) and its relevance in the genetic origin of the cultigen.

    Unreduced (2n) gametes are the driving force that leads to polyploidization of plants in nature and an important tool for ploidy breeding. Peanut (Arachis hypogaea L., Leguminosae) seems most likely to have originated by sexual polyploidization through interspecific hybridization between the wild diploid relatives -A. duranensis and A. ipaënsis-, and subsequent binding of the unreduced gametes in the hybrid. Therefore, aiming to contribute to the understanding the event of polyploid origin of this important crop, we made diploid hybrids between A. duranensis and A. ipaënsis, and then investigated the production of unreduced gametes in the hybrids by analyzing the size range of pollen, the constitution of the sporads, and the microsporogenesis process. The meiotic abnormalities found, such as persistent bridges, tripolar spindles and cytomixis processes, would lead to the formation of restitution nuclei in both the first and second meiotic division. These failures in the regularityof meiosis lead to the formation of dyads and triads, and consequently to 2n pollen grains. These findings constitute the first evidence of the formation of unreduced gametes in intergenomic hybrids obtained between the wild progenitors of A. hypogaea and enable us to propose a model of origin of the crop through sexual polyploidization.Fil: García, Alejandra Vanina.     Toward highly potent cancer agents by modulating the C-2 group of the arylthioindole class of tubulin polymerization inhibitors

    New arylthioindole derivatives having different cyclic substituents at position 2 of the indole were synthesized as anticancer agents. Several compounds inhibited tubulin polymerization at submicromolar concentration and inhibited cell growth at low nanomolar concentrations. Compounds 18 and 57 were superior to the previously synthesized 5. Compound 18 was exceptionally potent as an inhibitor of cell growth: it showed IC₅₀ = 1.0 nM in MCF-7 cells, and it was uniformly active in the whole panel of cancer cells and superior to colchicine and combretastatin A-4. Compounds 18, 20, 55, and 57 were notably more potent than vinorelbine, vinblastine, and paclitaxel in the NCI/ADR-RES and Messa/Dx5 cell lines, which overexpress P-glycoprotein. Compounds 18 and 57 showed initial vascular disrupting effects in a tumor model of liver rhabdomyosarcomas at 15 mg/kg intravenous dosage. Derivative 18 showed water solubility and higher metabolic stability than 5 in human liver microsomes

    La educación inclusiva frente a las desigualdades sociales: un estado de la cuestion y algunas reflexiones geograficas

    Este artículo establece un estado de la cuestión e la educación inclusiva en el mundo y sugiere algunas reflexiones al respecto. El primer apartado recuerda las conexiones ineludibles entre las preocupaciones educativas por la educación inclusiva y las preocupaciones más generales por la desigualdad. El segundo consigna los criterios de búsqueda de las publicaciones académicas, y observa dos grandes temas en sus contenidos: sobre todo, el cambio interno de las escuelas atrae las miradas, pero en segundo plano también el entorno territorial despierta algunas inquietudes. El tercero anota los criterios de búsqueda de la documentación del Banco Mundial, la OCDE y la UNESCO. En este ámbito los simposios de la Oficina Internacional de la Educación de UNESCO revelan una interpretación dispar, aunque convergente, del concepto de educación inclusiva en las distintas regiones mundiales. Asimismo, todas las publicaciones oficiales muestran una atención prioritaria a las dinámicas internas de las escuelas, puesto que apenas algunas esbozan ciertas relaciones entre la educación inclusiva y las políticas públicas. El último apartado adelanta varios argumentos a favor de una mayor consideración de las escalas local y estatal de la educación inclusiva. Las principales razones para atender a la dimensión local provienen de la causalidad acumulativa de las privaciones sociales, de la necesidad de articular la acción de las escuelas y de la posibilidad de abrir un espacio significativo para la participación ciudadana. Asimismo, las principales razones para atender a la dimensión estatal surgen de las posibles sinergias entre la educación inclusiva y la expansión educativa (p. ej. ¿es correlativo el avance de la escolarización en los distintos ciclos escolares?) como también entre la educación inclusiva y la protección social (p. ej. ¿tienen una implicación pedagógica consistente las abundantes condiciones educativas de las transferencias sociales?

    The Mitotic Arrest Deficient Protein MAD2B Interacts with the Small GTPase RAN throughout the Cell Cycle

    Contains fulltext : 81260.pdf (publisher's version ) (Open Access)BACKGROUND: Previously, we identified the mitotic arrest deficient protein MAD2B (MAD2L2) as a bona fide interactor of the renal cell carcinoma (RCC)-associated protein PRCC. In addition, we found that fusion of PRCC with the transcription factor TFE3 in t(X;1)(p11;q21)-positive RCCs results in an impairment of this interaction and, concomitantly, an abrogation of cell cycle progression. Although MAD2B is thought to inhibit the anaphase promoting complex (APC) by binding to CDC20 and/or CDH1(FZR1), its exact role in cell cycle control still remains to be established. METHODOLOGY/PRINCIPAL FINDINGS: Using a yeast two-hybrid interaction trap we identified the small GTPase RAN, a well-known cell cycle regulator, as a novel MAD2B binding protein. Endogenous interaction was established in mammalian cells via co-localization and co-immunoprecipitation of the respective proteins. The interaction domain of RAN could be assigned to a C-terminal moiety of 60 amino acids, whereas MAD2B had to be present in its full-length conformation. The MAD2B-RAN interaction was found to persist throughout the cell cycle. During mitosis, co-localization at the spindle was observed. CONCLUSIONS/SIGNIFICANCE: The small GTPase RAN is a novel MAD2B binding protein. This novel protein-protein interaction may play a role in (i) the control over the spindle checkpoint during mitosis and (ii) the regulation of nucleocytoplasmic trafficking during interphase

    Human Papillomavirus-16 E7 Interacts with Glutathione S-Transferase P1 and Enhances Its Role in Cell Survival

    Background:Human Papillomavirus (HPV)-16 is a paradigm for "high-risk" HPVs, the causative agents of virtually all cervical carcinomas. HPV E6 and E7 viral genes are usually expressed in these tumors, suggesting key roles for their gene products, the E6 and E7 oncoproteins, in inducing malignant transformation.Methodology/Principal Findings:By protein-protein interaction analysis, using mass spectrometry, we identified glutathione S-transferase P1-1 (GSTP1) as a novel cellular partner of the HPV-16 E7 oncoprotein. Following mapping of the region in the HPV-16 E7 sequence that is involved in the interaction, we generated a three-dimensional molecular model of the complex between HPV-16 E7 and GSTP1, and used this to engineer a mutant molecule of HPV-16 E7 with strongly reduced affinity for GSTP1.When expressed in HaCaT human keratinocytes, HPV-16 E7 modified the equilibrium between the oxidized and reduced forms of GSTP1, thereby inhibiting JNK phosphorylation and its ability to induce apoptosis. Using GSTP1-deficient MCF-7 cancer cells and siRNA interference targeting GSTP1 in HaCaT keratinocytes expressing either wild-type or mutant HPV-16 E7, we uncovered a pivotal role for GSTP1 in the pro-survival program elicited by its binding with HPV-16 E7.Conclusions/Significance:This study provides further evidence of the transforming abilities of this oncoprotein, setting the groundwork for devising unique molecular tools that can both interfere with the interaction between HPV-16 E7 and GSTP1 and minimize the survival of HPV-16 E7-expressing cancer cells. © 2009 Mileo et al

    Is season of birth related to disordered eating and personality in women with eating disorders?

    We assessed the relation between season of birth and eating disorder symptoms and personality characteristics in a sample of 880 women with eating disorders and 580 controls from two Price Foundation Studies. Eating disorder symptoms were assessed using Structured Interview of Anorexic and Bulimic Disorders and the Structured Clinical Interview for DSM-IV. Personality traits were assessed using the Temperament and Character Inventory and the Frost Multidimensional Perfectionism Scale. Date of birth was obtained from a sociodemographic questionnaire
