Nanoparticle-encapsulated chlorhexidine against oral bacterial biofilms
Background: Chlorhexidine (CHX) is a widely used antimicrobial agent in dentistry. Herein, we report the synthesis of a novel mesoporous silica nanoparticle-encapsulated pure CHX (Nano-CHX), and its mechanical profile and antimicrobial properties against oral biofilms.
Methodology/Principal Findings: The release of CHX from the Nano-CHX was characterized by UV/visible absorption spectroscopy. The antimicrobial properties of Nano-CHX were evaluated in both planktonic and biofilm modes of representative oral pathogenic bacteria. The Nano-CHX demonstrated potent antibacterial effects on planktonic bacteria and mono-species biofilms at the concentrations of 50-200 mu g/mL against Streptococcus mutans, Streptococcus sobrinus, Fusobacterium nucleatum, Aggregatibacter actinomycetemcomitans and Enterococccus faecalis. Moreover, Nano-CHX effectively suppressed multi-species biofilms such as S. mutans, F. nucleatum, A. actinomycetemcomitans and Porphyromonas gingivalis up to 72 h.
Conclusions/Significance: This pioneering study demonstrates the potent antibacterial effects of the Nano-CHX on oral biofilms, and it may be developed as a novel and promising anti-biofilm agent for clinical use.published_or_final_versio
Malignant melanoma and bone resorption
The cellular and humoral mechanisms accounting for osteolysis in skeletal metastases of malignant melanoma are uncertain. Osteoclasts, the specialised multinucleated cells that carry out bone resorption, are derived from monocyte/macrophage precursors. We isolated tumour-associated macrophages (TAMs) from metastatic (lymph node/skin) melanomas and cultured them in the presence and absence of osteoclastogenic cytokines and growth factors. The effect of tumour-derived fibroblasts and melanoma cells on osteoclast formation and resorption was also analysed. Melanoma TAMs (CD14+/CD51−) differentiated into osteoclasts (CD14−/CD51+) in the presence of receptor activator for nuclear factor κB ligand (RANKL) and macrophage-colony stimulating factor. Tumour-associated macrophage-osteoclast differentiation also occurred via a RANKL-independent pathway when TAMs were cultured with tumour necrosis factor-α and interleukin (IL)-1α. RT–PCR showed that fibroblasts isolated from metastatic melanomas expressed RANKL messenger RNA and the conditioned medium of cultured melanoma fibroblasts was found to be capable of inducing osteoclast formation in the absence of RANKL; this effect was inhibited by the addition of osteoprotegerin (OPG). We also found that cultured human SK-Mel-29 melanoma cells produce a soluble factor that induces osteoclast differentiation; this effect was not inhibited by OPG. Our findings indicate that TAMs in metastatic melanomas can differentiate into osteoclasts and that melanoma fibroblasts and melanoma tumour cells can induce osteoclast formation by RANKL-dependent and RANKL-independent mechanisms, respectively
Characterization of the artisanal fishing communities in Nepal and potential implications for the conservation and management of Ganges River Dolphin (Platanista gangetica gangetica)
The Ganges River dolphin (Platanista gangetica gangetica) (GRD) is classified as one
of the most endangered of all cetaceans in the world and the second scarcest
freshwater cetacean. The population is estimated to be less than 2,000 individuals.
In Nepal’s Narayani, Sapta Koshi, and Karnali river systems, survival of GRD
continues to be threatened by various anthropogenic activities, such as dam
construction and interactions with artisanal fisheries. A basic description of the
geographic scope, economics, and types of gear used in these fisheries would help
managers understand the fishery-dolphin interaction conflict and assist with
developing potential solutions. The main goal was to provide new information on
the artisanal fishing communities in Nepal. The specific objectives were to identify,
compile, and investigate the demographics, economics, fishing characteristics, and
perception of fishermen about GRD conservation in the Narayani, Sapta Koshi, and
Karnali rivers so conservation managers can develop and implement a potential
solution to the GRD-fishery interaction problem in Nepal. Based on 169 interviews,
79% of Nepalese fishermen indicated fishing was their primary form of income.
Fishermen reported fishing effort was greater in summer than winter; greatest in the
afternoon (14:30 hrs ± 0:27) and during low water level conditions; and gear was set
4.8 ± 0.2 days/week. Fishermen reported using eight different types of monofilament
nets (gillnets and cast nets). Sixty percent used gillnets less than 10 m long, and
nearly 30% preferred gillnets between 10 and 100 m long; a few used gillnets longer
than 100 m. Most fishermen reported they believed education, awareness, and
changing occupations were important for GRD conservation, but they indicated that
alternative occupational options were currently limited in Nepal. Nepalese
fishermen acknowledged that fisheries posed a risk to GRD, but they believed water
pollution, and dam/irrigation developments were the greatest threats.Versión del edito

An outcomes-based approach to curriculum development in pharmacy
Objective. To implement an outcomes-based approach to pharmacy curriculum development. Design. Desired learning outcomes were identified; course content, learning activities, and assessment instruments were designed; and evaluation was conducted and feedback obtained to refine the curriculum. Assessment. All professional skills, 4 generic capabilities, and the coherence of the curriculum scales showed significant improvement compared to graduates' performance under both the old and transitional curriculum. Conclusion. An outcomes-based approach to pharmacy curriculum development provided convincing evidence of enhancement to the curriculum.
