    Serological Activity of Adsorbed Leptospiral Antigen

    Leptospirosis is not a single disease but a group of diseases caused by a variety of leptospiral serotypes. Investigations during the past seventy years have shown that leptospirosis occurs in humans and animals in all areas of the world. Serological diagnostic procedures have been the principle means of identifying leptospirosis in humans and domestic animals. The agglutination test is a valuable diagnostic procedure for the identification of infections, such as leptospirosis, when it is not practical to isolate the causative organism from suspected cases. One of the earliest tests for the identification of leptospirosis was the agglutination-lysis test. This test has often been used as a basis for evaluation other leptospiral serological tests. Muraschi prepared an antigen consisting of lepostoiral components absorbed onto latex particles, for a tube agglutination test to detect leptospiral antibodies in serum. This was a modification of a procedure Singer and Flotz used for the serologic diagnosis of rheumatoid arthritis. The studies described herein pertain to tube agglutination reactions with homogenized leptospiral cell antigens absorbed onto particulate organic carriers for use as antigen in antibody-antigen reactions. The high cost of leptospiral whole cell antigen suggest the need of a search for a more economical use of this expensive material. During the course of this study, eight substances or extracts of substances were tested in solution for their suitability as fixation agents for homogenized leptospiral cell particles. From these eight substances, three appear to be suitable: supernatant of washed homogenized milk, fragments of yeast cells, and lecithin

    Mandating COVID-19 Vaccines

    Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccines hold promise to control the pandemic, and help restore normal social and economic life. The Food and Drug Administration (FDA) has granted emergency use authorization (EUA) for 2 mRNA vaccines and will likely issue full biologics licenses in the coming months. Anticipating vaccine scarcity, the Advisory Committee on Immunization Practice (ACIP) published guidance on vaccine priorities.Data show 95% efficacy for vaccines granted an EUA, but even highly effective vaccines cannot curb the pandemic without high population coverage and maintenance of other mitigation strategies. Recent data from 1,676 adults surveyed November 30-December 8, 2020 found that when a COVID-19 vaccine is approved and widely available: 34% would get it as soon as possible; 39% would wait; 9% would only get it if required for work/school; 15% would definitely not get it. Black persons, at high risk of infection and hospitalization, are less likely to report vaccine intent with only 20% reporting they would get the vaccine soon and 52% intending to wait. Intent to vaccinate has changed substantially over time and is likely to evolve.     Carbon Stable Isotope Analysis of Bison Dentition

    Understanding how bison behaved in the past can provide key insights for today\u27s managers, ecologists, and anthropologists. However, the direct application of both historic documentation and modern field observations may not provide the necessary insights for understanding bison behaviors in archeological and paleontological contexts. In order to develop a better understanding of possible behavior within these contexts, we have developed individual foraging histories for 22 Bison bison from the Glenrock Buffalo Jump assemblage of the Plains Late Prehistoric period in Wyoming and four Pleistocene B. priscus from the Ukraine. Incremental stable carbon isotopic values of dental enamel were used to determine foraging histories. The progressive development of enamel allows for samples to be selected that represent distinct periods of an individual\u27s life. Comparison of these dietary patterns among members of a herd can demonstrate foraging behaviors of cohorts and, in turn, the entire assemblage. Application of this high resolution paleodietary technique provides new information on bison behaviors in a paleontological and archaeological context

    Vaccine hesitancy and healthcare providers.

    While most people vaccinate according to the recommended schedule, this success is challenged by individuals and groups who delay or refuse vaccines. The aim of this article is to review studies on vaccine hesitancy among healthcare providers (HCPs), and the influences of their own vaccine confidence and vaccination behaviour on their vaccination recommendations to others. The search strategy was developed in Medline and then adapted across several multidisciplinary mainstream databases including Embase Classic & Embase, and PschInfo. All foreign language articles were included if the abstract was available in English. A total of 185 articles were included in the literature review. 66% studied the vaccine hesitancy among HCPs, 17% analysed concerns, attitudes and/or behaviour of HCPs towards vaccinating others, and 9% were about evaluating intervention(s). Overall, knowledge about particular vaccines, their efficacy and safety, helped to build HCPs own confidence in vaccines and their willingness to recommend vaccines to others. The importance of societal endorsement and support from colleagues was also reported. In the face of emerging vaccine hesitancy, HCPs still remain the most trusted advisor and influencer of vaccination decisions. The capacity and confidence of HCPs, though, are stretched as they are faced with time constraints, increased workload and limited resources, and often have inadequate information or training support to address parents' questions. Overall, HCPs need more support to manage the quickly evolving vaccine environment as well as changing public, especially those who are reluctant or refuse vaccination. Some recommended strategies included strengthening trust between HCPs, health authorities and policymakers, through more shared involvement in the establishment of vaccine recommendations

    Fast Pixel Space Convolution for CMB Surveys with Asymmetric Beams and Complex Scan Strategies: FEBeCoP

    Precise measurement of the angular power spectrum of the Cosmic Microwave Background (CMB) temperature and polarization anisotropy can tightly constrain many cosmological models and parameters. However, accurate measurements can only be realized in practice provided all major systematic effects have been taken into account. Beam asymmetry, coupled with the scan strategy, is a major source of systematic error in scanning CMB experiments such as Planck, the focus of our current interest. We envision Monte Carlo methods to rigorously study and account for the systematic effect of beams in CMB analysis. Toward that goal, we have developed a fast pixel space convolution method that can simulate sky maps observed by a scanning instrument, taking into account real beam shapes and scan strategy. The essence is to pre-compute the "effective beams" using a computer code, "Fast Effective Beam Convolution in Pixel space" (FEBeCoP), that we have developed for the Planck mission. The code computes effective beams given the focal plane beam characteristics of the Planck instrument and the full history of actual satellite pointing, and performs very fast convolution of sky signals using the effective beams. In this paper, we describe the algorithm and the computational scheme that has been implemented. We also outline a few applications of the effective beams in the precision analysis of Planck data, for characterizing the CMB anisotropy and for detecting and measuring properties of point sources.Comment: 26 pages, 15 figures.     Retention in care, resource utilization, and costs for adults receiving antiretroviral therapy in Zambia: a retrospective cohort study

    BACKGROUND: Of the estimated 800,000 adults living with HIV in Zambia in 2011, roughly half were receiving antiretroviral therapy (ART). As treatment scale up continues, information on the care provided to patients after initiating ART can help guide decision-making. We estimated retention in care, the quantity of resources utilized, and costs for a retrospective cohort of adults initiating ART under routine clinical conditions in Zambia. METHODS: Data on resource utilization (antiretroviral [ARV] and non-ARV drugs, laboratory tests, outpatient clinic visits, and fixed resources) and retention in care were extracted from medical records for 846 patients who initiated ART at ≄15 years of age at six treatment sites between July 2007 and October 2008. Unit costs were estimated from the provider’s perspective using site- and country-level data and are reported in 2011 USD. RESULTS: Patients initiated ART at a median CD4 cell count of 145 cells/ÎŒL. Fifty-nine percent of patients initiated on a tenofovir-containing regimen, ranging from 15% to 86% depending on site. One year after ART initiation, 75% of patients were retained in care. The average cost per patient retained in care one year after ART initiation was 243(95243 (95% CI, 194-293),rangingfrom293), ranging from 184 (95% CI, 172−172-195) to 304(95304 (95% CI, 290-$319) depending on site. Patients retained in care one year after ART initiation received, on average, 11.4 months’ worth of ARV drugs, 1.5 CD4 tests, 1.3 blood chemistry tests, 1.4 full blood count tests, and 6.5 clinic visits with a doctor or clinical officer. At all sites, ARV drugs were the largest cost component, ranging from 38% to 84% of total costs, depending on site. CONCLUSIONS: Patients initiate ART late in the course of disease progression and a large proportion drop out of care after initiation. The quantity of resources utilized and costs vary widely by site, and patients utilize a different mix of resources under routine clinical conditions than if they were receiving fully guideline-concordant care. Improving retention in care and guideline concordance, including increasing the use of tenofovir in first-line ART regimens, may lead to increases in overall treatment costs

    Characterizing star formation activity in infrared dark cloud MSXDC G048.65-00.29

    Infrared Dark Clouds (IRDCs), condensed regions of the ISM with high column densities, low temperatures and high masses, are suspected sites of star formation. Thousands of IRDCs have already been identified. To date, it has not been resolved whether IRDCs always show star formation activity and, if so, if massive star formation (> 8 solar masses) is the rule or the exception in IRDCs. Previous analysis of sub-millimeter cores in the cloud MSXDC G048.65-00.29 (G48.65) indicates embedded star formation activity. To characterize this activity in detail, mid-infrared photometry (3-30 micron) has been obtained with the Spitzer Space Telescope. This paper analyzes the point sources seen in the 24 micron band, combined with counterparts or upper limits at shorter and longer wavelengths. Data points in wavelength bands ranging from 1 up to 850 micron are used to compare each 24 micron source to a set of Spectral Energy Distributions of Young Stellar Object (YSO) models. By assessing the models that fit the data, an attempt is made to identify YSOs as such and determine their evolutionary stages and stellar masses. A total of 17 sources are investigated, 13 of which are classified as YSOs, primarily - but not exclusively - in an early embedded phase of star formation. The modeled masses of the central stellar objects range from sub-solar to ~8 solar masses. Every YSO is at less than 1 pc projected distance from its nearest YSO neighbor. We conclude that IRDC G48.65 is a region of active star formation. We find YSOs in various evolutionary phases, indicating that the star formation in this cloud is not an instantaneous process. The inferred masses of the central objects suggest that this IRDC hosts only low to intermediate mass YSOs and none with masses exceeding ~8 solar masses.Comment: 10 pages, 6 figures; v2: minor editorial changes to match published versio
