    Identifying a molecular phenotype for bone marrow stromal cells with in vivo bone-forming capacity

    The ability of bone marrow stromal cells (BMSCs) to differentiate into osteoblasts is being exploited in cell-based therapy for repair of bone defects. However, the phenotype of ex vivo cultured BMSCs predicting their bone-forming capacity is not known. Thus we employed DNA microarrays comparing two human bone marrow stromal cell (hBMSC) populations: One is capable of in vivo heterotopic bone formation (hBMSC-TERT(+Bone)), and the other is not (hBMSC-TERT(-Bone)). Compared with hBMSC-TERT(-Bone), the hBMSC-TERT(+Bone) cells had an increased overrepresentation of extracellular matrix genes (17% versus 5%) and a larger percentage of genes with predicted SP3 transcription factor-binding sites in their promoter region (21% versus 8%). On the other hand, hBMSC-TERT(-Bone) cells expressed a larger number of immune-response-related genes (26% versus 8%). In order to test for the predictive value of these markers, we studied the correlation between their expression levels in six different hBMSC-derived clones and the ability to form bone in vivo. We found a significant correlation for decorin, lysyl oxidase-like 4, natriuretic peptide receptor C, and tetranectin. No significant positive correlation was found for canonical osteoblastic markers Runx2, alkaline phosphatase, collagen type I, osteopontin, and bone sialoprotein. Prospective isolation of four additional hBMSC clones based on their expression levels of the molecular markers correlated with their in vivo bone-formation ability. In conclusion, our data suggest an in vitro molecular signature predictive for hBMSCs' in vivo bone-formation ability. Identifying more of these predictive markers would be very useful in the quality control of osteoblastic cells before use in therapy

    Ozone exposure assessment in a southern California community.

    An ozone exposure assessment study was conducted in a Southern California community. The Harvard ozone passive sampler was used to monitor cohorts of 22 and 18 subjects for 8 weeks during the spring and fall of 1994, respectively. Ozone exposure variables included 12-hr personal O3 measurements, stationary outdoor O3 measurements from a continuous UV photometer and from 12-hr Harvard active monitors, and time-activity information. Results showed that personal O3 exposure levels averaged one-fourth of outdoor stationary O3 levels, attributable to high percentages of time spent indoors. Personal O3 levels were not predicted well by outdoor measurements. A random-effect general linear model analysis indicated that variance in personal exposure measurements was largely accounted for by random error (59-82%), followed by inter-subject (9-18%) and between-day (9-23%) random effects. The microenvironmental model performs differently by season, with the regression model for spring cohorts exhibiting two times the R2 of the fall cohorts (R2 = 0.21 vs. 0.09). When distance from the stationary monitoring site, elevation, and traffic are taken into account in the microenvironmental models, the adjusted R2 increased almost twofold for the fall personal exposure data. The low predictive power is due primarily to the apparent spatial variation of outdoor O3 and errors in O3 measurements and in time-activity records (particularly in recording the use of air conditioning). This study highlights the magnitude of O3 exposure misclassification in epidemiological settings and proposes an approach to reduce exposure uncertainties in assessing air pollution health effects

    Structure preserving schemes for mean-field equations of collective behavior

    In this paper we consider the development of numerical schemes for mean-field equations describing the collective behavior of a large group of interacting agents. The schemes are based on a generalization of the classical Chang-Cooper approach and are capable to preserve the main structural properties of the systems, namely nonnegativity of the solution, physical conservation laws, entropy dissipation and stationary solutions. In particular, the methods here derived are second order accurate in transient regimes whereas they can reach arbitrary accuracy asymptotically for large times.     Searching for globular cluster chemical anomalies on the main sequence of a young massive cluster

    The spectroscopic and photometric signals of the star-to-star abundance variations found in globular clusters seem to be correlated with global parameters like the cluster’s metallicity, mass, and age. Understanding this behaviour could bring us closer to the origin of these intriguing abundance spreads. In this work we use deep HST photometry to look for evidence of abundance variations in the main sequence of a young massive cluster NGC 419 (∼105 M⊙, ∼1.4 Gyr). Unlike previous studies, here we focus on stars in the same mass range found in old globulars (∼0.75–1 M⊙), where light elements variations are detected. We find no evidence for N abundance variations among these stars in the Un − B and U − B colour–magnitude diagrams of NGC 419. This is at odds with the N variations found in old globulars like 47 Tuc, NGC 6352, and NGC 6637 with similar metallicity to NGC 419. Although the signature of the abundance variations characteristic of old globulars appears to be significantly smaller or absent in this young cluster, we cannot conclude if this effect is mainly driven by its age or its mass

    Pulmonary Toxicity and Adjuvant Effect of Di-(2-exylhexyl) Phthalate in Ovalbumin-Immunized BALB/c Mice

    BACKGROUND: Asthma is a complex pulmonary inflammatory disease, which is characterized by airway hyperresponsiveness, variable airflow obstruction and inflammation in the airways. The majority of asthma is allergic asthma, which is a disease caused by type I hypersensitivity mediated by IgE. Exposures to a number of environmental chemicals are suspected to lead to asthma, one such pollutant is di-(2-ethylheyl) phthalate (DEHP). DEHP is a manufactured chemical that is commonly added in plastic products to make them flexible. Epidemiological studies have revealed a positive association between DEHP exposure and asthma prevalence. METHODOLOGY/PRINCIPAL FINDINGS: The present study was aimed to determine the underlying role of DEHP exposure in airway reactivity, especially when combined with allergen exposure. The biomarkers include pulmonary histopathology, airway hyperresponsiveness (lung function), IgE, IL-4, IFN-γ and eosinophils. Healthy balb/c mice were randomly divided into eight exposure groups (n = 8 each): (1) saline control, (2) 30 µg/(kg•d) DEHP, (3) 300 µg/(kg•d) DEHP, (4) 3000 µg/(kg•d) DEHP, and (5) ovalbumin (OVA)-sensitized group, (6) OVA-combined with 30 µg/(kg•d) DEHP, (7) OVA-combined with 300 µg/(kg•d) DEHP, and (8) OVA-combined with 3000 µg/(kg•d) DEHP. Experimental tests were conducted after 52-day DEHP exposure and subsequently one week of challenge with aerosolized OVA. The principal findings include: (1) Strong postive associations exist between OVA-combined DEHP exposure and serum total IgE (T-IgE), as well as histological findings. These positive associations show a dose-dependent low dose sensitive effect of DEHP. (2) IL-4, eosinophil recruitment and lung function are also indicators for adjuvant effect of DEHP. CONCLUSIONS/SIGNIFICANCE: Our results suggest that except the significant changes of immunological and inflammatory biomarkers (T-IgE, IL-4, IFN-γ and eosinophils), the pulmonary histological (histopathological examination) and physiological (lung function) data also support that DEHP may promote and aggravate allergic asthma by adjuvant effect

    Prospective blind comparative clinical study of two point fixation of zygomatic complex fracture using wire and mini plates

    <p>Abstract</p> <p>Background</p> <p>The zygomatic maxillary complex (ZMC) fractures are one of the most frequent injuries of the facial skeleton due to its position and facial contour. Assaults, road traffic accidents and falls are the principal etiologic factors that may cause fractures of zygomatic bone. The different fixation methods are applied to treat the zygomatic bone fractures, with many more classifications which have been described in the literature for the ease of management. The type of the fracture, its severity and associated facial fractures usually interferes the treatment modality.</p> <p>Purpose of study</p> <p>The aim of this paper is to show the results of 18yrs prospective blind comparative study using wire and plate osteosynthesis which needed open reduction and internal fixation involving Type II to Type IV Spissel and Schroll ZMC fractures.</p> <p>Materials and methods</p> <p>Total 80 cases included in the study out of 1780 ZMC cases which were treated using wire and plate osteosynthesis over a period of 18 yrs, involving only Type II to Type IV Spissel and Schroll ZMC fractures. Other types excluded from study to prevent observer bias. All the fixations carried out through Standard Dingman's incision using stainless steel 26 gauze wire and titanium 1.5 mm mini plate system under general anesthesia by single maxillofacial surgeon and evaluated by another maxillofacial surgeon who is blinded for surgical procedure after 2 and 4 wks of follow-up for facial symmetry, wound healing, functional assessment (mouth opening, diplopia), and sensory disturbance. All the data tabulated in Excel software (Microsoft) for statistical analysis. P-value calculated to know the Significance of treatment modality in all aspects.</p> <p>Results</p> <p>Result shows no significant p-values indicating both the operating techniques are equally efficient in the surgical management of ZMC fracture.</p> <p>Conclusion</p> <p>Osteosynthesis by mini plates is simple, logical and effective treatment compared to wire osteosynthesis in regard to stability of fracture fragments. Wire osteosynthesis will be helpful in emergency surgeries or where the mini plates are not available. Even though the wire osteosynthesis is economical compared to mini plate fixation; but the time and skill is required for fixation of wires.</p
