    Shaking It Up: Challenging Veteran Teachers to Change

    My research, conducted at a 600 student middle school, explored the conditions and factors needed by veteran teachers to learn about and implement educational change, either mandated or teacher-initiated. Qualitative research methods used included in-depth interviews of veteran teachers and observations in classrooms, cluster meetings and professional development workshops. Five factors appeared to facilitate the change effort for this group of teachers. They are: An understanding that the change process is a challenge in itself. A school culture that is conducive to change. A school principal who supports, inspires and encourages teachers. Appropriate professional development is offered to teachers. Teachers are actively involved in educational change. The veteran teachers studied were stimulated by change, regularly sought ways to improve, aspired to teach their students to love learning and were committed to helping their students succeed. Recommendations are offered for the important stakeholders in the school reform effort; teachers, principals, parents, the school district and schools of education

    Early Cretaceous araucarian driftwood from hemipelagic sediments of the Puez area, South Tyrol, Italy

    AbstractWe describe a calcareously permineralised fossil tree-trunk, preserved as driftwood, within hemipelagic sediments of the Cretaceous Puez Formation near Wolkenstein, South Tyrol, Italy. Planktic foraminiferal assemblages recovered from the marls containing the fossil wood indicate a latest middle Albian age. Based on its wood anatomy, the trunk is assigned to Agathoxylon and probably has an affinity with the conifer family Araucariaceae. The wood lacks pronounced tree-rings consistent with tree growth within the broad humid tropical belt that existed at that time. The trunk contains cylindrical chambers filled within faecal pellets, demonstrating that oribatid mites infested the tree, either during life, or shortly after death. Prior to final burial, the tree-trunk drifted out into the open sea for a considerable period as indicated by extensive borings assigned to the ichnospecies Teredolites longissimus and produced by teredinid bivalves. Relatively little is known about the Cretaceous floras of Italy, so this new finding fills a gap in our knowledge of the composition and ecology of the vegetation of this region

    Analyzing M-CSF dependent monocyte/macrophage differentiation: Expression modes and meta-modes derived from an independent component analysis

    <p>Abstract</p> <p>Background</p> <p>The analysis of high-throughput gene expression data sets derived from microarray experiments still is a field of extensive investigation. Although new approaches and algorithms are published continuously, mostly conventional methods like hierarchical clustering algorithms or variance analysis tools are used. Here we take a closer look at independent component analysis (ICA) which is already discussed widely as a new analysis approach. However, deep exploration of its applicability and relevance to concrete biological problems is still missing. In this study, we investigate the relevance of ICA in gaining new insights into well characterized regulatory mechanisms of M-CSF dependent macrophage differentiation.</p> <p>Results</p> <p>Statistically independent gene expression modes (GEM) were extracted from observed gene expression signatures (GES) through ICA of different microarray experiments. From each GEM we deduced a group of genes, henceforth called <it>sub-mode</it>. These <it>sub-modes </it>were further analyzed with different database query and literature mining tools and then combined to form so called <it>meta-modes</it>. With them we performed a knowledge-based pathway analysis and reconstructed a well known signal cascade.</p> <p>Conclusion</p> <p>We show that ICA is an appropriate tool to uncover underlying biological mechanisms from microarray data. Most of the well known pathways of M-CSF dependent monocyte to macrophage differentiation can be identified by this unsupervised microarray data analysis. Moreover, recent research results like the involvement of proliferation associated cellular mechanisms during macrophage differentiation can be corroborated.</p

    Emotional valence and arousal affect reading in an interactive way: neuroimaging evidence for an approach-withdrawal framework

    A growing body of literature shows that the emotional content of verbal material affects reading, wherein emotional words are given processing priority compared to neutral words. Human emotions can be conceptualised within a two-dimensional model comprised of emotional valence and arousal (intensity). These variables are at least in part distinct, but recent studies report interactive effects during implicit emotion processing and relate these to stimulus-evoked approach-withdrawal tendencies. The aim of the present study was to explore how valence and arousal interact at the neural level, during implicit emotion word processing. The emotional attributes of written word stimuli were orthogonally manipulated based on behavioural ratings from a corpus of emotion words. Stimuli were presented during an fMRI experiment while 16 participants performed a lexical decision task, which did not require explicit evaluation of a word's emotional content. Results showed greater neural activation within right insular cortex in response to stimuli evoking conflicting approach-withdrawal tendencies (i.e., positive high-arousal and negative low-arousal words) compared to stimuli evoking congruent approach vs. withdrawal tendencies (i.e., positive low-arousal and negative high-arousal words). Further, a significant cluster of activation in the left extra-striate cortex was found in response to emotional than neutral words, suggesting enhanced perceptual processing of emotionally salient stimuli. These findings support an interactive two-dimensional approach to the study of emotion word recognition and suggest that the integration of valence and arousal dimensions recruits a brain region associated with interoception, emotional awareness and sympathetic functions

    Novel Relationships among Lampreys (Petromyzontiformes) Revealed by a Taxonomically Comprehensive Molecular Data Set

    The systematics of lampreys was investigated using complete mitochondrial cytochrome b sequences from all genera and nearly all recognized species. The families Geotriidae and Petromyzontidae are monophyletic, but the family Mordaciidae was resolved as two divergent lineages at the base of the tree. Within Petromyzontidae, the nonparasitic Lethenteron sp. S and Okkelbergia aepyptera were recognized as distinct lineages, Lethenteron morii and Lampetra zanandreai were moved to new genera, a sister species relationship was recovered between Caspiomyzon wagneri and Eudontomyzon hellenicus, and a clade was recovered inclusive of Entosphenus hubbsi and western North American Lampetra (L. ayresii and L. richardsoni). The placement of E. hellenicus as the sister species to C. wagneri reduces the number of genera comprised entirely of parasitic species to two, Geotria and Petromyzon. The recognition of distinct lineages for O. aepyptera and Lethenteron sp. S recognizes, for the first time, lineages comprised entirely of nonparasitic species. Apart from the results mentioned above, monophyly was supported for the multispecific genera Entosphenus, Eudontomyzon, Ichthyomyzon, Lampetra (restricted to European species), and Lethenteron. Intergeneric relationships within Petromyzontidae were poorly resolved, but separate clades inclusive of Entosphenus and Tetrapleurodon (subfamily Entospheninae) and one comprised of Eudontomyzon, Lampetra, and Okkelbergia were recovered

    A phase I trial to evaluate the safety and pharmacokinetics of low-dose methotrexate as an anti-malarial drug in Kenyan adult healthy volunteers

    <p>Abstract</p> <p>Background</p> <p>Previous investigations indicate that methotrexate, an old anticancer drug, could be used at low doses to treat malaria. A phase I evaluation was conducted to assess the safety and pharmacokinetic profile of this drug in healthy adult male Kenyan volunteers.</p> <p>Methods</p> <p>Twenty five healthy adult volunteers were recruited and admitted to receive a 5 mg dose of methotrexate/day/5 days. Pharmacokinetics blood sampling was carried out at 2, 4, 6, 12 and 24 hours following each dose. Nausea, vomiting, oral ulcers and other adverse events were solicited during follow up of 42 days.</p> <p>Results</p> <p>The mean age of participants was 23.9 ± 3.3 years. Adherence to protocol was 100%. No grade 3 solicited adverse events were observed. However, one case of transiently elevated liver enzymes, and one serious adverse event (not related to the product) were reported. The maximum concentration (C<sub>max</sub>) was 160-200 nM and after 6 hours, the effective concentration (C<sub>eff</sub>) was <150 nM.</p> <p>Conclusion</p> <p>Low-dose methotraxate had an acceptable safety profile. However, methotrexate blood levels did not reach the desirable C<sub>eff </sub>of 250-400-nM required to clear malaria infection <it>in vivo</it>. Further dose finding and safety studies are necessary to confirm suitability of this drug as an anti-malarial agent.</p

    Für den Erfolg eines Produktes oder einer Dienstleistung ist die umfassende Kenntnis der Bedürfnisse, Wahrnehmungs- und Entscheidungsfilter von Zielgruppen entscheidend. Nur eine zielgerichtete, zur Zielgruppe passende Produktentwicklung und -vermarktung kann Auf Dauer erfolgreich sein. Da soziodemographische Zielgruppendefinitionen heute alleine nicht valide genug sind, um Zielgruppen umfassend zu verstehen, werden im Marketing zu-nehmend psychographische Verortungen von Zielgruppen verwendet. Auch bei chronisch Kranken reichen rein soziodemographische und medizinische Kriterien nicht aus, um Produkte und Dienstleistungen passend zu gestalten. Psychographische Erkenntnisse können die klassischen Kriterien sinnvoll ergänzen. Am Beispiel von Menschen mit Diabetes wird in einer internationalen empirischen Studie gezeigt, dass sich Menschen mit Diabetes mit Hilfe eines neuropsychologisch fundierten, visuellen Testverfahrens (dem u.a. an der NORD-AKADEMIE entwickelten Visual Questionnaire, ViQ) eindeutig psychographisch verorten lassen. Die Implikationen und Anwendungsmöglichkeiten dieser psychographischen Erkenntnisse für die Produktentwicklung, Kommunikation, Vertrieb und Marketing bei Krankenkassen, medizinischem Personal und der Pharmabranche werden aufgezeigt und kritisch diskutiert. --

    Steroid metabolome analysis reveals prevalent glucocorticoid excess in primary aldosteronism

    BACKGROUND. Adrenal aldosterone excess is the most common cause of secondary hypertension and is associated with increased cardiovascular morbidity. However, adverse metabolic risk in primary aldosteronism extends beyond hypertension, with increased rates of insulin resistance, type 2 diabetes, and osteoporosis, which cannot be easily explained by aldosterone excess. METHODS. We performed mass spectrometry–based analysis of a 24-hour urine steroid metabolome in 174 newly diagnosed patients with primary aldosteronism (103 unilateral adenomas, 71 bilateral adrenal hyperplasias) in comparison to 162 healthy controls, 56 patients with endocrine inactive adrenal adenoma, 104 patients with mild subclinical, and 47 with clinically overt adrenal cortisol excess. We also analyzed the expression of cortisol-producing CYP11B1 and aldosterone-producing CYP11B2 enzymes in adenoma tissue from 57 patients with aldosterone-producing adenoma, employing immunohistochemistry with digital image analysis. RESULTS. Primary aldosteronism patients had significantly increased cortisol and total glucocorticoid metabolite excretion (all P < 0.001), only exceeded by glucocorticoid output in patients with clinically overt adrenal Cushing syndrome. Several surrogate parameters of metabolic risk correlated significantly with glucocorticoid but not mineralocorticoid output. Intratumoral CYP11B1 expression was significantly associated with the corresponding in vivo glucocorticoid excretion. Unilateral adrenalectomy resolved both mineralocorticoid and glucocorticoid excess. Postoperative evidence of adrenal insufficiency was found in 13 (29%) of 45 consecutively tested patients. CONCLUSION. Our data indicate that glucocorticoid cosecretion is frequently found in primary aldosteronism and contributes to associated metabolic risk. Mineralocorticoid receptor antagonist therapy alone may not be sufficient to counteract adverse metabolic risk in medically treated patients with primary aldosteronism

    Ultrafast all-optical switching by single photons

    An outstanding goal in quantum optics is the realization of fast optical non-linearities at the single-photon level. Such non-linearities would allow for the realization of optical devices with new functionalities such as a single-photon switch/transistor or a controlled-phase gate, which could form the basis of future quantum optical technologies. While non-linear optics effects at the single-emitter level have been demonstrated in different systems, including atoms coupled to Fabry-Perot or toroidal micro-cavities, super-conducting qubits in strip-line resonators or quantum dots (QDs) in nano-cavities, none of these experiments so far has demonstrated single-photon switching on ultrafast timescales. Here, we demonstrate that in a strongly coupled QD-cavity system the presence of a single photon on one of the fundamental polariton transitions can turn on light scattering on a transition from the first to the second Jaynes-Cummings manifold with a switching time of 20 ps. As an additional device application, we use this non-linearity to implement a single-photon pulse-correlator. Our QD-cavity system could form the building-block of future high-bandwidth photonic networks operating in the quantum regime
