Detection of Leishmania infantum by PCR, serology and cellular immune response in a cohort study of Brazilian dogs
The sensitivity and specificity of PCR, serology (ELISA) and lymphoproliferative response to Leishmania antigen for the detection of Leishmania infantum infection were evaluated in a cohort of 126 dogs exposed to natural infection in Brazil. For PCR, Leishmania DNA from bone-marrow was amplified with both minicircle and ribosomal primers. The infection status and time of infection of each dog were estimated from longitudinal data. The sensitivity of PCR in parasite-positive samples was 98%. However, the overall sensitivity of PCR in post-infection samples, from dogs with confirmed infection, was only 68%. The sensitivity of PCR varied during the course of infection, being highest (78â88%) 0â135 days post-infection and declining to around 50% after 300 days. The sensitivity of PCR also varied between dogs, and was highest in sick dogs. The sensitivity of serology was similar in parasite-positive (84%), PCR-positive (86%) and post-infection (88%) samples. The sensitivity of serology varied during the course of infection, being lowest at the time of infection and high (93â100%) thereafter. Problems in determining the specificity of serology are discussed. The sensitivity and specificity of cellular responsiveness were low. These data suggest that PCR is most useful in detecting active or symptomatic infection, and that serology can be a more sensitive technique for the detection of all infected dogs
The importance of conscientiousness in the telemarketing industry
Researchers have long pondered the personality type-performance relationship. However, in the sales industry and especially in telemarketing, findings have been contradictory with regards to which personality trait best contributes to performance. The telemarketing industry is a rapidly growing industry, but is typically marred with high staff turnover and low levels of performance. An improved understanding of the factors that contribute towards performance in this industry is a key mandate of management. Personality-type has been shown to be a key driver of performance in the sales industry. However, it is unclear whether the personality types touted in typical sales positions (for example extroversion) are suited for the telemarketing environment. South Africa is one of the top emerging call centre locations in the world and as such the purpose of this study was to determine which personality type best contributes towards performance in the South African telemarketing industry. A census was conducted on telesales personnel in a large telemarketing organisation and the five-factor model was used to test employees' personality types. Contrary to current research, which suggests that extraversion is the five-factor trait best suited to salespeople, this study found that conscientiousness was the only personality trait that influenced sales performance in the call centre context
Sub-nanosecond signal propagation in anisotropy engineered nanomagnetic logic chains
Energy efficient nanomagnetic logic (NML) computing architectures propagate
and process binary information by relying on dipolar field coupling to reorient
closely-spaced nanoscale magnets. Signal propagation in nanomagnet chains of
various sizes, shapes, and magnetic orientations has been previously
characterized by static magnetic imaging experiments with low-speed adiabatic
operation; however the mechanisms which determine the final state and their
reproducibility over millions of cycles in high-speed operation (sub-ns time
scale) have yet to be experimentally investigated. Monitoring NML operation at
its ultimate intrinsic speed reveals features undetectable by conventional
static imaging including individual nanomagnetic switching events and
systematic error nucleation during signal propagation. Here, we present a new
study of NML operation in a high speed regime at fast repetition rates. We
perform direct imaging of digital signal propagation in permalloy nanomagnet
chains with varying degrees of shape-engineered biaxial anisotropy using
full-field magnetic soft x-ray transmission microscopy after applying single
nanosecond magnetic field pulses. Further, we use time-resolved magnetic
photo-emission electron microscopy to evaluate the sub-nanosecond dipolar
coupling signal propagation dynamics in optimized chains with 100 ps time
resolution as they are cycled with nanosecond field pulses at a rate of 3 MHz.
An intrinsic switching time of 100 ps per magnet is observed. These
experiments, and accompanying macro-spin and micromagnetic simulations, reveal
the underlying physics of NML architectures repetitively operated on nanosecond
timescales and identify relevant engineering parameters to optimize performance

Limited Neutralizing Antibody Specificities Drive Neutralization Escape in Early HIV-1 Subtype C Infection
We previously showed that HIV-1 subtype C viruses elicit potent but highly type-specific neutralizing antibodies (nAb) within the first year of infection. In order to determine the specificity and evolution of these autologous nAbs, we examined neutralization escape in four individuals whose responses against the earliest envelope differed in magnitude and potency. Neutralization escape occurred in all participants, with later viruses showing decreased sensitivity to contemporaneous sera, although they retained sensitivity to new nAb responses. Early nAb responses were very restricted, occurring sequentially and targeting only two regions of the envelope. In V1V2, limited amino acid changes often involving indels or glycans, mediated partial or complete escape, with nAbs targeting the V1V2 region directly in 2 cases. The alpha-2 helix of C3 was also a nAb target, with neutralization escape associated with changes to positively charged residues. In one individual, relatively high titers of anti-C3 nAbs were required to drive genetic escape, taking up to 7 weeks for the resistant variant to predominate. Thereafter titers waned but were still measurable. Development of this single anti-C3 nAb specificity was associated with a 7-fold drop in HIV-1 viral load and a 4-fold rebound as the escape mutation emerged. Overall, our data suggest the development of a very limited number of neutralizing antibody specificities during the early stages of HIV-1 subtype C infection, with temporal fluctuations in specificities as escape occurs. While the mechanism of neutralization escape appears to vary between individuals, the involvement of limited regions suggests there might be common vulnerabilities in the HIV-1 subtype C transmitted envelope
Barriers to Predicting the Mechanisms and Risk Factors of Non-Contact Anterior Cruciate Ligament Injury
High incidences of non-contact anterior cruciate ligament (ACL) injury, frequent requirements for ACL reconstruction, and limited understanding of ACL mechanics have engendered considerable interest in quantifying the ACL loading mechanisms. Although some progress has been made to better understand non-contact ACL injuries, information on how and why non-contact ACL injuries occur is still largely unavailable. In other words, research is yet to yield consensus on injury mechanisms and risk factors. Biomechanics, video analysis, and related study approaches have elucidated to some extent how ACL injuries occur. However, these approaches are limited because they provide estimates, rather than precise measurements of knee - and more specifically ACL - kinematics at the time of injury. These study approaches are also limited in their inability to simultaneously capture many of the contributing factors to injury
Mannose-rich glycosylation patterns on HIV-1 subtype C gp120 and sensitivity to the lectins, Griffithsin, Cyanovirin-N and Scytovirin.
Griffithsin (GRFT), Cyanovirin-N (CV-N) and Scytovirin (SVN) are lectins that inhibit HIV-1 infection by binding to multiple mannose-rich glycans on the HIV-1 envelope glycoproteins (Env). Here we show that these lectins neutralize subtype C primary virus isolates in addition to Env-pseudotyped viruses obtained from plasma and cervical vaginal lavages. Among 15 subtype C pseudoviruses, the median IC50 values were 0.4, 1.8 and 20.1 nM for GRFT, CV-N and SVN, respectively, similar to what was found for subtype B and A. Analysis of Env sequences suggested that concomitant lack of glycans at positions 234 and 295 resulted in natural resistance to these compounds, which was confirmed by site-directed mutagenesis. Furthermore, the binding sites for these lectins overlapped that of the 2G12 monoclonal antibody epitope, which is generally absent on subtype C Env. This data support further research on these lectins as potential microbicides in the context of HIV-1 subtype C infection
